We further examine the efficacy of treatments targeting oxidative stress and redox regulation in animal and human epilepsies in the literature that warrant further investigation. Treatment approaches aimed at rectifying oxidative stress and aberrant redox signaling may enable control of neuroinflammation and improve patient outcomes.Human serum albumin (HSA) nanoparticles are promising biocompatible, nontoxic, and non-immunogenic platforms for biomedical applications such as bioimaging and drug and gene delivery. The development of nonviral gene delivery vectors is a great challenge for efficient and safe gene therapy. Sulforaphane (SF) can stimulate the expression of antioxidant genes via activation of a nuclear transcription factor, the erythroid-2 related factor 2 (Nrf-2). Here, we use polyethyleneimine (PEI)-stabilized HSA nanoparticles to stimulate endogenous antioxidant defense mechanisms in lung epithelial cells L-132 through the combinatorial effect of SF drug and antioxidant superoxide dismutase 1 gene (pSOD1 plasmid) delivered by HSA-PEI-SF-pSOD1 nanocomposites (NCs). The developed NCs demonstrated high biocompatibility (L-132 viability, >95%, MTT assay) and high antioxidant activity because of efficient entry of the SOD1 gene and SF-loaded NCs at a very low (3 μg) dose in L-132 cells. A high transfection efficiency of L-132 cells (∼66%, fluorescent microscopy) was obtained with the GFP-tagged transgene SOD1-GFP. We speculate that the antioxidant activity of HSA-PEI-SF-pSOD1 NCs in L-132 cells is due to the initial release of SF followed by subsequent SOD1 gene expression after three to four days of incubation. Hence, the developed HSA-based NCs can be efficient biocompatible nanocarriers for safe and effective drug and gene delivery applications to treat diseases with high oxidative stress due to combinatorial SF and SOD1 gene mechanisms.Actin is a cytoskeletal filament involved in numerous biological tasks, such as providing cells a shape or generating and transmitting forces. Particularly important for these tasks is the ability of actin to grow and shrink. To study the role of actin in living cells this dynamic needs to be targeted. In the past, such alterations were performed by destabilizing actin. In contrast, we used the natural compound miuraenamide A in living retinal pigmented epithelial (RPE-1) cells to stabilize actin filaments and show that it decreases actin filament dynamics and elongates filament length. Cells treated with miuraenamide A increased their adhesive area and express more focal adhesion sites. These alterations result in a lower migration speed as well as a shift of nuclear position. We therefore postulate that miuraenamide A is a promising new tool to stabilize actin polymerization and study cellular behavior such as migration.Duchenne Muscular Dystrophy (DMD) is an X-linked neuromuscular disease which to date is incurable. The major cause of death is dilated cardiomyopathy however, its pathogenesis is unclear as existing cellular and animal models do not fully recapitulate the human disease phenotypes. In this study, we generated cardiac organoids from patient-derived induced pluripotent stem cells (DMD-COs) and isogenic-corrected controls (DMD-Iso-COs) and studied if DMD-related cardiomyopathy and disease progression occur in the organoids upon long-term culture (up to 93 days). Histological analysis showed that DMD-COs lack initial proliferative capacity, displayed a progressive loss of sarcoglycan localization and high stress in endoplasmic reticulum. Additionally, cardiomyocyte deterioration, fibrosis and aberrant adipogenesis were observed in DMD-COs over time. RNA sequencing analysis confirmed a distinct transcriptomic profile in DMD-COs which was associated with functional enrichment in hypertrophy/dilated cardiomyopathy, arrhythmia, adipogenesis and fibrosis pathways. Moreover, five miRNAs were identified to be crucial in this dysregulated gene network. In conclusion, we generated patient-derived cardiac organoid model that displayed DMD-related cardiomyopathy and disease progression phenotypes in long-term culture. We envision the feasibility to develop a more complex, realistic and reliable in vitro 3D human cardiac-mimics to study DMD-related cardiomyopathies.In teleosts, cardiac plasticity plays a central role in mediating thermal acclimation. Previously, we demonstrated that exposure to elevated temperatures throughout development (+4°C) improved acute thermal tolerance of the heart in juvenile Atlantic salmon. Fish raised in a warmer thermal regime also displayed higher proportions of compact myocardium within their ventricles. In the present study, we investigated the molecular mechanisms supporting this temperature-specific phenotype by comparing relative protein abundance in ventricular tissue from the same experimental fish using mass spectrometry. We provide the first description of the ventricular proteome in juvenile Atlantic salmon and identify 79 proteins displaying differential abundance between developmental treatments. The subset of proteins showing higher abundance in fish raised under elevated temperatures was significantly enriched for processes related to ventricular tissue morphogenesis, and changes in protein abundance support a hypertrophic model of compact myocardium growth. Proteins associated with the vasculature and angiogenesis also showed higher abundance in the warm-developmental group, suggesting capillarization of the compact myocardium in the hearts of these fish. Proteins related to oxidative metabolism and protein homeostasis also displayed substantive shifts in abundance between developmental treatments, underscoring the importance of these processes in mediating thermal plasticity of cardiac function. While rapid growth under warm developmental temperatures has been linked to cardiomyopathies in farmed salmon, markers of cardiac pathology were not implicated in the present study. Thus, our findings offer a molecular footprint for adaptive temperature-dependent plasticity within the ventricle of a juvenile salmonid.Objectives To review the evidence of associations between adverse birth outcomes (ABO) and industrial air pollution. Methods Searches were conducted in PubMed, and Scopus databases, and additional articles were found from snowball search techniques. The included studies feature a study population of mothers with live-born babies exposed to industrial air pollutants, and they examine the effects of industrial pollutants on adverse birth outcomes-namely, low birth weight, term low birth weight, preterm birth, and small for gestational age. Results Altogether, 45 studies were included in this review. Exposure to PM2.5, PAHs, benzene, cadmium, and mixtures of industrial air pollutants and living near an industrial area affect birth outcomes. Conclusion This study concludes that industrial air pollution is an important risk factor for ABO, especially low birth weight and preterm birth. The strongest evidence is associations between ABO and air pollution from power plants and petrochemical industries. Understanding of specific chemicals that are critical to birth outcomes is still vague. Accumulate evidence indicated that moxibustion has a positive effect on pain treatment. However, the bibliometric analysis of moxibustion on pain remains uncertain. Herein, this study aimed to explore the current hotspots and the research trends over the past decade to bring a great panorama in this field.

Publications were retrieved from the database Web of Science Core Collection (WoSCC) related to moxibustion therapy for pain treatment from January 1, 2012, to December 31, 2021. CiteSpace and Excel were applied to analyze the volume of publications, journals, cited journals, dual-map analysis, countries, institutes, authors, references, and keywords with citation bursts.

A total of 360 publications were retrieved from 2012 to 2021. The annual number of publications increased steadily with some fluctuations over the past 10 years. "The Evid Based Complement Alternat Med" and "the Gut" were the top-cited journals in frequency and centrality.

In the study, a multi-purpose reverse logistics network has been designed to create effectual management of medical waste (MW) generated in 39 districts of Istanbul, a heavily populated city, during the COVID-19 pandemic as well as that to be generated in the next decade. learn more With the model, the medical waste management system in Istanbul is analyzed during the pandemic and for the next 10 years. The model attempts to integrate economic, environmental, and social objectives within the sustainable development goals. It aims to maximize the number of personnel and government earnings for the estimated MW of a megacity while minimizing the total fixed cost and the cost of carbon emissions and transportation. The results indicated that the existing facilities are sufficient for the treatment and disposal of MW generated even under pandemic conditions. However, the capacity of the sterilization facility could be insufficient to treat the estimated amount of MW in the next decade. Opening a sterilization facility near the sanitary landfill in Komurcuoda with a total management cost of 62,450,332 €/year would be an optimum solution for Istanbul MW. In comparison to the single-purpose model results, the multi-purpose model resulted in approximately 42,000 € more in total cost. Sensitivity analyses show that the amount of MW has the most significant effect on the total cost. This simple model created an effective MW management proposal for Istanbul, which can be a model for megacities.

This study applies the conservation of resource (COR) theory to ground the statistical results investigating the moderation and moderated mediation effects of COVID-19 anxiety in relation to the UTAUT model. According to the COR theory, anxiety linked with the COVID-19 pandemic endangers resources such as human connections (death of friends and relatives), job security and finances, as well as physical health and psychological well-being. Since resource loss is more salient according to COR theory, academicians are more willing to employ technologies like Zoom, MS Teams, and Google Classroom since the COVID-19 threat is existential and the resource loss is eminent. To test the theory, data was collected through an anonymous survey during the months of December 2020 and January 2021.
