Aedes aegypti is a principal vector for several viruses including dengue virus, chikungunya virus and zika virus. Economic burden of mosquito-borne diseases, relative failure of traditional control strategies and the resistance development against insecticides enforces towards genetic manipulation of Ae. aegypti. Hence, a key gene doublesex (Aedsx) which regulate sex differentiation and alternatively splices to form male and female specific transcripts (AedsxM and AedsxF ). CRISPR/Cas9 technique was employed to sex specifically disrupt the female-specific isoforms, AedsxF1 and AedsxF2 , both of which were shown to be expressed only in female mosquitoes. Targeting of dsxF at the developmental stage has resulted in various phenotypic anomalies of adult females. The rate of adult mutation phenotype was recorded between 29 and 37% along with anomalies of wing size, proboscis length and reduction in the sizes of pre-blood-meal and after blood-meal ovaries in dsxF1 and dsxF2 microinjected groups, respectively. These findings can be correlated with reduced fecundity rate of Go female, where AedsxF1 and AedsxF2 groups showed reduction rate in range of 23-31%. Furthermore, hatching inhibition rate of 28 to 36% was also observed in G1 generation when compared to the wildtype. Overall, these results demonstrated that AedsxF disruption has resulted in multiple female traits disruption including decreased fertility of the female that could directly or indirectly associated with reproduction and its disease transmitting abilities. All these findings suggesting that CRISPR works to alter the developmental pathways as predicted, and therefore this method potentially gives us the basis for the sex-ratio distortion system as genetic control approach for the management of this vector.Red pepper (Capsicum annuum L.) is one of the most commonly cultivated vegetable in the Mediterranean region. This study evaluated the effects of biochar derived from corncob and poultry litter on growth of red pepper (Capsicum annuum L.) and some chemical properties of a silty clay soil. The experiment consisted of two factors, i.e., biochar doses (0, 0.5, 1.0 and 2%) and poultry litter doses (0, 0.5, 1.0 and 2%). The number of days to 50% flowering, plant height, stem diameter, total number of leaves per plant, the number of main branches per plant, fresh root weight, root length, dry shoot weight, macro (P and K) and micro (Fe, Zn, Cu and Mn) nutrient concentrations of leaves were determined to compare the efficiency biochar and poultry litter. Moreover, post-harvest soil analysis was conducted to measure pH, organic matter, and macro and micronutrient contents. Biochar had varying impact on plant growth parameters, whereas poultry litter alone or in combination with biochar increased macro and micronutrient concentrations of soil and improved most of the growth parameters of red pepper. In contrast, sole biochar application had no significant impact on most of the growth parameters. Wider C/N ratio (107.7) of corncob derived biochar restricted the nitrogen supply for plant growth. The combination of 0.5% biochar and 2% poultry litter resulted in the highest plant height (36.7 cm) and stem diameter (0.69 cm). The results revealed that application of single biochar derived from corncob is insufficient to supply adequate nutrients for optimal plant growth. The application of biochar alone enhances carbon sequestration in soils, however most biochars like cornconb biochar do not contain sufficient available plant nutrients. Therefore, biochars should be applied along with mineral fertilizers or organic materials such as poultry manure which is rich in available plant nutrients.Fifteen alfalfa populations were tested for resistance to the seedling damping-off disease sourced by Rhizoctonia solani, Fusarium solani, and Macrophomina phaseolina. In a laboratory experiment, saponin treatment significantly diminished the mycelial growth of the causal fungi of alfalfa damping-off disease. Roots of the fifteen alfalfa populations varied in saponin and lignin content. Selection for the considerably resistant plants leads to the best growth performance, desirable yield, and high nutritive values such as crude protein (CP), crude fier (CF), nitrogen free extract (NFE), ash, and ether extract (EE) contents. The experiments took place in 2019 on a research station with an area of 10 ha (Moscow region, Russian Federation), divided into two even sites of 5 ha (control, experimental). Eleven germination experiments were carried out to determine the influence of livestock wastewater irrigation on radish seeds (1 - control; 10 - irrigation with liquid and solid phases of wastewater samples mixed with pure water). The experimental and control plots appeared to differ in terms of the bulk density of soil. Changes occurred in all horizons (p ≤ 0.05) but a soil layer with a depth of 0.2-0.4 m. Soil horizons in the experiment plots all exhibited lower porosity (p ≤ 0.05) except for the topsoil, and the water capacity was higher in the topsoil (p ≤ 0.05) and near-surface layer (p ≤ 0.05). The experiment showed higher concentrations of hummus (p ≤ 0.01) and phosphorus (p ≤ 0.01). As for nitrogen, significant changes only occurred in the topsoil (p ≤ 0.01). In the germination experiments, more than 90% of radish seeds germinated. Besides, their root length was higher compared to the control (p ≤ 0.05). The results of the study suggest that livestock wastewater can benefit crop cultivation after preliminary treatment. Finally, the experiments revealed a reduced soil salt accumulation.Marine natural products have displayed numerous advantageous effects on biological activities, including antioxidants and cytotoxicity. The total lipids, carotenoids, chlorophyll a and b content, total phenolic content (TPC), total flavonoid content (TFC), and antioxidant activity of methanolic crude extract of the green seaweed Halimeda opuntia were all measured in this study. The TPC of the extracts was determined according to the Folin-Ciocalteu method, yielding a result of 55.04 ± 0.98 mg GAE/g of extract. As determined by the aluminium chloride colorimetric method, the TFC of the extract was 40.02 ± 0.02 mg QE/g of extract. Antioxidant activity was determined by using a 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay with different concentrations that ranged between 200 and 1000 µg/mL, noted H. opuntia as the highest in DPPH reduction (63.61 %) at 1000 µg/mL concentration. Total antioxidant capacity (TAC) of the extract was 57.36 ± 0.004 mg AAE/g extract at concentration of 1.0 mg/mL. The cytotoxic activity of this seaweed was pre-screened against a panel of cell lines including estrogen receptor-positive human breast adenocarcinoma (MCF-7), estrogen negative human breast adenocarcinoma (MDA-MB-231), human colorectal adenocarcinoma (HT-29), human hepatocellular carcinoma (HepG2), and mouse embryonic fibroblast (3T3) using the MTT assay. The content of total lipids in H. opuntia was 1.60 ± 0.002 %. Total carotenoids were 115.57 ± 0.98 µg/g, while chlorophyll a and b were 148.73 ± 2.60 µg/g and 290.83 ± 9.46 µg/g, respectively. In terms of cytotoxicity activity, methanolic extract of H. opuntia was found to be highly cytotoxic to MCF-7 cells, with an IC50 of 25.14 ± 1.02 g/mL, and slightly less so to 3T3 cells (IC50 65.23 ± 0.25 µg/mL). This research was aimed to assess the risk factors for intestinal parasites in school children of Malakand, Pakistan. Two hundred and eighty eight students were enrolled between February and June 2016. Out of the total enrolled 184 were agreed to collect stool specimens. A questionnaire was also used to collect the data on socio-demographic and socio-economic characteristics of the participants. All the students were guided to collect at least 10gof their own stool specimens. Each of the stool specimens was diagnosed for the presence of any stage of helminth or protozoal parasites. Formal ether concentration method and wet mount techniques were applied. One way ANOVA was used for calculation of P value when it was less than 0.05 which was considered significant. Eighty two percent of the participants were found infected with one species of parasite while 69.9% of the participants were infected with more than one species of intestinal parasites. The most prevalent parasite was hook worm 33.4% (n = 99/296) followed by Taenia saginata 28.7% (n = 85/296), Ascaris lumbricoides 27.7% (n = 82/296), Hymenolepis nana 6.08% (n = 18/296), Entamoeba histolytica 3.37% (n = 10/296) and least for each Enterobius vermicularis and Fasciola hepatica 0.37% (n = 1/296). However, few studies have been done on the effect of culinary treatments. Therefore, the aim of the present study is to evaluate the effect of the most popular culinary treatments (boiled, steamed and sous vide) on the physicochemical properties of Ulva lactuca. The treatments were applied at temperatures of 100 °C in the case of both boiling and steaming whereas 50 and 75 °C on sous vide culinary. learn more Results illustrated that both the chemical composition and physical properties of Ulva lactuca greatly affected depending on the culinary method and time. The culinary processes produce an increase in water activity (0.962-0.989) with respect to the raw algae (0.952). All the applied culinary treatments showed an increase in the content of pH, chlorophylls and carotenoids compared to the raw algae, and the highest rise was after boiling for 5 min, although this effect decreases slightly at longer times of boiling.
