Phosphonic acid-based fungicides, also referred to as phosphonates, have been used extensively as crop protectants in horticulture since the late 1970s, and more recently in native ecosystems and forestry. Discovering that phosphonates are effective against foliar and soilborne oomycete diseases, such as those caused by species of Phytophthora, Pythium and Plasmopara, was a significant breakthrough, especially for soilborne pathogens that are notoriously difficult to manage. Phosphonates have played an important role in protection of forests and sensitive natural ecosystems, under threat from these pathogens. Since introduction, their increased application in management of non-oomycete diseases, along with other functionalities, demonstrates their versatility in agriculture and more broadly. Continued use of phosphonic acid crop protectants will be underpinned by demonstrated efficacy and safety, and a better understanding of specific interactions within the plant, pathogen and environment. © 2020 Society of Chemical Industry.Prior knowledge and long-term memory can guide our attention to facilitate search for and detection of subtle targets embedded in a complex scene. A number of neuropsychological and experimental studies have investigated this effect, yet results in the field remain mixed, as there is a lack of consensus regarding the neural correlates thought to support memory-guided attention. The purpose of this systematic review and meta-analysis was to identify a common set of brain structures involved in memory-guided attention. Statistical analyses were computed on functional magnetic resonance imaging (fMRI) studies that presented participants with a task that required them to detect a target or a change embedded in repeated and novel complex visual displays. After a systematic search, 10 fMRI studies met the selection criteria and were included in the analysis. The results yielded four significant clusters. Activity in right inferior parietal (Brodmann area [BA] 9) and right superior parietal (BA 7) lobes suggests involvement of a fronto-parietal attention network, while activity in left mid-cingulate cortex (BA 23) and right middle frontal gyrus (BA 10) suggests involvement of a fronto-parietal control network. These findings are consistent with the notion that fronto-parietal circuits are important for interfacing retrieved memories with attentional systems to guide search. Dengvaxia, the only licensed vaccine, may not be adequately safe in young and seronegative patients; hence, development of a safer, more effective vaccine is of great public health interest. Virus-like particles (VLPs) are a safe and very efficient vaccine strategy, and DENV VLPs have been produced in various expression systems. Here, we describe the production of DENV VLPs in Nicotiana benthamiana using transient expression. The co-expression of DENV structural proteins (SP) and a truncated version of the non-structural proteins (NSPs), lacking NS5 that contains the RNA-dependent RNA polymerase, led to the assembly of DENV VLPs in plants. These VLPs were comparable in appearance and size to VLPs produced in mammalian cells. Contrary to data from other expression systems, expression of the protein complex prM-E was not successful, and strategies used in other expression systems to improve the VLP yield did not result in increased yields in plants but, rather, increased purification difficulties. Immunogenicity assays in BALB/c mice revealed that plant-made DENV1-SP + NSP VLPs led to a higher antibody response in mice compared with DENV-E domain III displayed inside bluetongue virus core-like particles and a DENV-E domain III subunit. These results are consistent with the idea that VLPs could be the optimal approach to creating candidate vaccines against enveloped viruses.End stage renal disease (ESRD) is associated with a high mortality rate among patients hospitalized with COVID-19. To the best of our knowledge, there is limited data on the clinical features, ethnicity, inpatient glycaemic control and outcomes in patients with diabetes related ESRD in the literature. We report the clinical features and outcomes of 39 consecutive ESRD patients (28 on haemodialysis [HD] and 11 with renal transplant) secondary to diabetic kidney disease admitted to a university hospital with COVID-19. We observed a high prevalence of patients of Afro-Caribbean ethnicity hospitalized with COVID-19 with a 73% and 54% prevalence in renal transplant and HD groups respectively. The mortality rate of our cohort was 36%. Nearly a one-third of HD patients and one-fifth of transplant patients had hypoglycaemic events during COVID-19 hospitalization. Adjustment of diabetes treatment was frequently required. Our data highlight the importance of integrated multidisciplinary care of patients with diabetes related ESRD hospitalized with COVID-19.Platelet activation and thrombus formation have been implicated to be detrimental for intraportal pancreatic islet transplants. The platelet-specific collagen receptor glycoprotein VI (GPVI) plays a key role in thrombosis through cellular activation and the subsequent release of secondary mediators. In aggregometry and in a microfluidic dynamic assay system modeling flow in the portal vein, pancreatic islets promoted platelet aggregation and triggered thrombus formation, respectively. While platelet GPVI deficiency did not affect the initiation of these events, it was found to destabilize platelet aggregates and thrombi in this process. Interestingly, while no major difference was detected in early thrombus formation after intraportal islet transplantation, genetic GPVI deficiency or acute anti-GPVI treatment led to an inferior graft survival and function in both syngeneic mouse islet transplantation and xenogeneic human islet transplantation models. selleck chemical These results demonstrate that platelet GPVI signaling is indispensable in stable thrombus formation induced by pancreatic islets. GPVI deficiency resulted in thrombus destabilization and inferior islet engraftment indicating that thrombus formation is necessary for a successful intraportal islet transplantation in which platelets are active modulators.
