Nonreplicating rotavirus vaccine (NRRV) candidates are being developed with the aim of serving the needs of developing countries. A significant proportion of the cost of manufacturing such vaccines is the purification in multiple chromatography steps. Crystallization has the potential to reduce purification costs and provide new product storage modality, improved operational flexibility, and reduced facility footprints. This communication describes a systematic approach for the design of the crystallization of an NRRV candidate, VP8 subunit proteins fused to the P2 epitope of tetanus toxin, using first-principles models and preliminary experimental data. The first-principles models are applied to literature data to obtain feasible crystallization conditions and lower bounds for nucleation and growth rates. Crystallization is then performed in a hanging-drop vapor diffusion system, resulting in the nucleation and growth of NRRV crystals. The crystals obtained in a scaled-up evaporative crystallization contain proteins truncated in the P2 region, but have no significant differences with the original samples in terms of antibody binding and overall conformational stability. These results demonstrate the promise of evaporative crystallization of the NRRV.Polymerized whey protein and low-methoxyl pectin were used as thickening agents in fermented dairy food formulations. Effects of polymerized whey protein and pectin (PP) system on particle size distributions, rheological properties, molecular weight distributions of polysaccharides, microstructures, and volatile compounds of goat milk kefir mild and kefir were investigated. Clustering analysis indicated that the PP system addition could increase the particle size distributions and rheological properties of both kefir products. Molecular weight distributions of polysaccharides shown that an intermolecular aggregation occurred between pectin and exopolysaccharides (produced by microorganisms from starter cultures). Micrographs suggested that polymerized whey protein interacted with casein network to form a dense structure. Meanwhile, PP system did not change the characteristics flavor of goat milk kefir mild and kefir. Results indicated that the PP system may be a good combination to improve the physical properties of kefir products. PRACTICAL APPLICATION Goat milk kefir has gained more and more attention due to its specific flavor and functional properties. However, poor texture of milk curd limits the consumption of fermented goat milk. As a new combination of thickening agents, PP system could specifically improve the particle size distributions and rheological properties of fermented goat milk. And the characteristic flavor of goat milk kefir was not changed. Results indicated that PP system can be used as thickening agents for formulations of different fermented goat milk products.Cysteinyl leukotrienes (CysLTs) are inflammatory lipid mediators that play a central role in the pathophysiology of several inflammatory diseases. Understanding the molecular mechanisms underlying the role of CysLTs and their receptors in cancer progression will help investigate the potential of targeting CysLTs signalling for anti-cancer therapy. Despite the wide variety of scaffold materials currently available, such as collagen, critical problems in achieving bone regeneration remain, including a rapid absorption period and low tensile strength as well as high costs. Inspired by extracellular matrix protein and topographical cues, we developed a polycaprolactone-based scaffold for bone regeneration using a soluble eggshell membrane protein (SEP) coating and a nanotopography structure for enhancing the physical properties and bioactivity. The scaffold exhibited adequate flexibility and mechanical strength as a biomedical platform for bone regeneration. The highly aligned nanostructures and SEP coating were found to regulate and enhance cell morphology, adhesion, proliferation, and differentiation in vitro. In a calvaria bone defect mouse model, the scaffolds coated with SEP applied to the defect site promoted bone regeneration along the direction of the nanotopography in vivo. These findings demonstrate that bone-inspired nanostructures and SEP coatings have high potential to be applicable in the design and manipulation of scaffolds for bone regeneration.

The CIT algorithms are extensions of the classification and regression tree algorithm that use splitting criteria based on subgroup-specific treatment effect estimators appropriate for observational data. We describe inverse probability weighting, g-formula, and doubly robust estimators of subgroup-specific treatment effects, derive their asymptotic properties, and use them to construct splitting criteria for the CIT algorithms. We study the performance of the algorithms in simulations and implement them to analyze data from an observational study that evaluated the effectiveness of right heart catheterization for critically ill patients.Some studies have reported the association between maternal serum zinc (Zn) levels and pre-eclampsia. Hence, this systematic review and meta-analysis was planned to generate summarized evidence on the association between maternal serum Zn levels and pre-eclampsia in African women. Four electronic databases such as PubMed, Hinari, Google Scholar, and African Journals Online were searched for studies published in English. Joanna Briggs Institute Meta-Analysis of Statistics Assessment and Review Instrument, and Newcastle-Ottawa Scale were used for data extraction and quality assessment of the included studies. The meta-regression analysis was performed by the Stata 14 software. The standardized mean difference (SMD) values of lipid profiles were computed to assess their association with pre-eclampsia at 95% CI. A total of 12 observational studies were included. The mean values of serum Zn level were significantly lower in pre-eclamptic women as compared with normotensive pregnant women (Zn = 59.40 ± 22.80 μg/dL and 80.24 ± 16.04 μg/dL), respectively. The pooled SMD of Zn was significantly reduced in pre-eclamptic women as compared with normotensive pregnant women with the SMD of -1.45 (95% CI -2.26, -0.65) at 95% CI. In this review, we found that the maternal Zn serum level was significantly reduced in pre-eclamptic women than normotensive pregnant women. This suggests that Zn could be involved in the etio-pathogenesis of pre-eclampsia. However, the specific functions of Zn in pre-eclampsia pathogenesis should be proved in large-scale clinical trial studies.

To assess the safety of centrifugal therapeutic plasma exchange (TPE) in pediatric patients.

The authors did a retrospective analysis of all TPE procedures performed in pediatric patients over a period of 19 y (2001-2019). Procedures were done on different apheretic devices, daily or on alternate days depending on the clinical condition of the patient. Adverse events during the procedure were noted and analyzed.

A total of 672 TPE (with mean of 6.77 ± 4.85) procedures were performed for 99 pediatric patients with different indications like hematological (n = 68), renal (n = 12), neurology (n = 18) and hepatology (n = 1). The mean age was 7.00 ± 3.11 y and weight was 20.72 ± 9.17kg. Adverse events (AEs) were observed during 34 (5%) procedures, most common were allergic reactions to replacement fluid (2.24%) followed by hypotension (1.04%), symptomatic hypocalcemia (1.04%), line occlusion (0.59%), and febrile non hemolytic transfusion reaction (0.41%). A significant correlation of AEs was observed with weight (p = 0.045), total blood volume of the patient (p = 0.04), increasing number of procedures (p = 0.000) and replacement fluid [Fresh frozen plasma (FFP)] (p = 0.04). All AEs were managed as per departmental standard operating procedures (SOPs) completing procedures successfully except one which was abandoned. No mortality was observed during the procedures.

To examine the relationship between peer victimization and psychosomatic symptoms among school-going children.

A total of 213 school-going children (Mean = 11.49 y, SD = 1.44) studying in grades 5 to 8 were recruited from 2 schools from a north Indian city. The children were administered the California Bullying Victimization Scale (CBVS), Pre-Adolescent Adjustment Scale (PAAS), and the Children's Somatization Inventory (CSI). The total score on the somatization inventory and the total number of psychosomatic symptoms endorsed were used as the main outcome measures.

Out of the 213 students, 42.7% were involved in bullying either as victims or as perpetrators. A little more than one-fourth (26.8%) were victims. A consistent positive association between victimization and somatic complaints was found. The most common physical symptoms reported by victims included headache (60.7%), chest pain (35.7%), stomach pain (33.9%), weakness (30.4%), and pain in arms and legs (19.6%).
