We had previously reported an increase in trimethylamine N-oxide (TMAO) levels in patients with both compensated and decompensated heart failure (HF) and alteration in gut microbiota composition using 16S rRNA gene amplicon analysis. Although a metagenome-wide analysis showed that choline-TMA lyase levels increased in HF patients, which TMA generation pathway from choline, carnitine, or betaine contributes to the increase in TMAO levels in HF needs to be elucidated.

We conducted a metagenome-wide shotgun sequencing analysis of gut microbiota and measured the TMAO levels in plasma of 22 HF patients during the compensated phase and 11 age-, sex-, and comorbidity-matched control subjects, whose gut microbiota compositions were reported in a previous 16S rRNA-based analysis.

The abundance of cntA/B was positively correlated with TMAO, especially in HF patients, whereas that of cutC/D or betaine reductase was not correlated either in controls or HF patients. The abundance of cntA/B was mainly derived from the genera Escherichia and Klebsiella either in controls or HF patients.

Stress urinary incontinence (SUI) is prevalent among older women and can result from insufficient regeneration of the pudendal nerve (PN). Electrical stimulation (ES) of the PN upregulates brain derived neurotrophic factor (BDNF) and accelerates regeneration. Using tyrosine kinase B (TrkB) to reduce the availability of free BDNF, the aim of this study was to determine if BDNF is necessary for accelerated recovery via ES in a model of SUI.

Our SUI model consists of Female Sprague-Dawley rats, whose PNs were crushed bilaterally twice for 30s, followed by insertion of a modified Foley catheter into the vagina with balloon inflation for 4h. These rats were divided into 4 groups 1) Sham PN crush and sham vaginal distension without electrode implantation and with saline treatment (sham injury); 2) SUI with sham stimulation and saline treatment (SUI); 3) SUI and ES with saline treatment (SUI&ES); and 4) SUI and ES with TrkB treatment (SUI&ES&TrkB). The present analysis was conducted to validate the reliability of this interpretation with 129 pesticides more recently evaluated. Before applying, the decision tree was revised to be consistent in all steps. The pesticides showed similar characterization of increased ALP to the previous analysis in that the increase was more frequent than in rats and that liver hypertrophy and hepatotoxicity commonly coexisted with an increase in ALP in dogs. When short- and long-term studies of 58 pesticides inducing ALP activity in dogs were applied to the revised tree, the increased ALP in 8 pesticides was judged not adverse in either study. The revision of the tree did not affect the NOAEL judgment of these pesticides; however, the revised routes contributed to the judgment more robustly. In this study, chitosan-coated hollow zein particles using calcium phosphate as a sacrificing template (CS-HZ) were fabricated to co-encapsulate folic acid (FA) and caffeic acid (CA). Partition, photostability, and antioxidant capacity of bioactive compounds were also studied. The size, polydispersity index and ζ-potential of optimized CS-HZ were 176.3 nm, 0.14 and +39.3 mV, respectively, indicating their small and uniform dimension with excellent colloidal stability. FA interacted with chitosan to form complexes and then coated on the zein particles where CA was encapsulated. After co-encapsulation in CS-HZ, the photostability of both FA and CA was improved in comparison with encapsulation of single compound, with 85% of FA remaining after 240 min of UVA irradiation, and 90% of CA remaining after 80 min. Antioxidant activity of CA decreased upon encapsulation, but significantly increased after irradiation. It is used as emulsifier, thickener, stabilizer, and as binding agent in many industries. In the present project, it was planned to synthesize guar gum based polyurethanes by varying the amount of guar gum. Guar gum (GG) was used along with hydroxyl-terminated polybutadiene (HTPB) as soft segment, which was then reacted with isophorone diisocyanate (IPDI) to form PU pre-polymers. In last step, these -NCO terminated pre-polymers were extended with 1,4 butane diol as chain extender. The prepared polyurethane samples were then characterized by using FTIR, solid-state 1HNMR and X-ray diffraction (XRD). Thermal behavior of the samples was studied by thermogravimetric analysis (TGA) and differential scanning calorimetry (DSC).
