European Commission, Voyager Therapeutics, and EuroAtaxia.Given the interest in accurately predicting the Longitudinal Dispersion Coefficient (Dx) within the fields of hydraulic and water quality modeling, a wide range of methods have been used to estimate this parameter. In order to improve the accuracy of Dx predictions, this paper proposes the use of a Deep Convolutional Network (DCN), a sub-field of machine learning. The proposed deep neural network architecture consists of two parts; first, a one-dimensional convolutional neural network (CNN) to build informative feature maps, and second, a stack of deep, fully connected layers to estimate pollution dispersion (as Dx) in streams. To accurately predict Dx the developed model draws upon a large and diverse array of datasets in the form of three dimensionless parameters Width/Depth (W/H), Velocity/Shear Velocity (U/u*), and Longitudinal Dispersion Coefficient/(Depth * Shear Velocity) (Dx /Hu*). The model's accuracy is compared to that of several empirical models using a number of statistical measures. In addition,lues from river geometry and hydraulic datasets, with low errors across all ranges of Dx. The most significant advantage of DCN is that it tries to learn high-level features from data in an incremental manner.Early initiation of polysubstance use (PSU) is a strong predictor of subsequent addiction, however scarce individuals present resilience capacity. This neuroimaging study aimed to investigate structural correlates associated with cessation or reduction of PSU and determine the extent to which brain structural features accounted for this resilient outcome. Participants from a European community-based cohort self-reported their alcohol, tobacco and cannabis use frequency at ages 14, 16 and 19 and had neuroimaging sessions at ages 14 and 19. We included three groups in the study the resilient-to-PSU participants showed PSU at 16 and/or 14 but no more at 19 (n = 18), the enduring polysubstance users at 19 displayed PSU continuation from 14 or 16 (n = 193) and the controls were abstinent or low drinking participants (n = 460). We conducted between-group comparisons of grey matter volumes on whole brain using voxel-based morphometry and regional fractional anisotropy using tract-based spatial statistics. Random-forests machine-learning approach generated individual-level PSU-behavior predictions based on personality and neuroimaging features. Adolescents resilient to PSU showed significant larger grey matter volumes in the bilateral cingulate gyrus compared with enduring polysubstance users and controls at ages 19 and 14 (p less then 0.05 corrected) but no difference in fractional anisotropy. The larger cingulate volumes and personality trait "openness to experience" were the best precursors of resilience to PSU. Early in adolescence, a larger cingulate gyrus differentiated adolescents resilient to PSU, and this feature was critical in predicting this outcome. This study encourages further research into the neurobiological bases of resilience to addictive behaviors.Steroid concentrations in serum are fluctuating during pregnancy of many mammal species. The current knowledge about endocrinology of gestation is mainly based on immunoassays. However, the lack of specificity of these assays hampers the reliability of the results. In the present work, we developed and validated a methodology associating liquid chromatography (LC) and mass spectrometry (MS) to simultaneously quantify, with high specificity and accuracy, estrone-3-sulfate (E3S), progesterone (PRO), estrone (E1) and estradiol (E2) in serum of two different mammal species. The sample preparation procedure is based on a simple protein precipitation and a derivatization with dansyl chloride. After the chromatographical separation, compounds were analyzed with a triple-quadrupole mass spectrometer operating in multiple reaction monitoring. Mare and American bison serum samples were analyzed with the validated method and results were compared with concentrations measured with commercial radioimmunoassay (RIA), enzyme linked immunosorbent assay (ELISA) and chemiluminescent microparticle immunoassay (CMIA). Following these criterions relative standard deviation less then 15% and relative bias less then 15%, lower limits of quantification of 0.5 ng/mL (E3S), 0.1 ng/mL (PRO) and 2 pg/mL (E1 and E2) were achieved. Most of the comparison between immunoassays and LC-MS showed poor correlation and proportional differences. Our LC-MS method is able to simultaneously quantify several steroid hormones with high specificity, accuracy and sensitivity in serum of two different mammal species. Our method constitutes a useful and performant tool for veterinary clinicians and LC-MS should thus be used to update and refine the current knowledge about the endocrinology of pregnancy in mammals.Growing research evidence suggests that elevated TLR2 is closely related to the occurrence and development of nonalcoholic steatohepatitis (NASH). However, a little is known about its regulatory mechanism. Here, we found that IFN-γ and TLR2 expression is significantly upregulated in NASH associated rat liver specimens. Meanwhile, IFN-γ positively regulated the expression of TLR2 and its target genes in NR8383 rat macrophage cells in dose- & time-dependent manner. Importantly, IFN-γ also regulated the related transcriptional factors pSTAT1 and IRF1. Moreover, we identified that the DNA fragment from -1000 to -200 bp of the TLR2 promoter region is responsible for STAT1 binding, especially the STAT1-BS3 (-591∼-573 bp). Further investigation verified that STAT1β is essential in this process, rather than STAT1α. Overall, our findings suggest that IFN-γ promotes TLR2 transcription and its target genes expression by STAT1β. In this study, we tested the chemical compositions of masticated and ingested different plastic types by G. mellonella. High throughput sequencing of 16S rRNA gene was used to characterize the alteration of the microbial communities derived from salivary glands, gut contents and whole G. mellonella larvae. In this study we identify Quiescin sulfhydryl oxidase 1 (QSOX1) acting as a cellular pro-oxidant, specifically in the context of sorafenib treatment of HCC. QSOX1 disrupts redox homoeostasis and sensitizes HCC cells to oxidative stress by inhibiting activation of the master antioxidant transcription factor NRF2. A negative correlation between QSOX1 and NRF2 expression was validated in tumor tissues from 151 HCC patients. Mechanistically, QSOX1 restrains EGF-induced EGFR activation by promoting ubiquitination-mediated degradation of EGFR and accelerating its intracellular endosomal trafficking, leading to suppression of NRF2 activity. Additionally, QSOX1 potentiates sorafenib-induced ferroptosis by suppressing NRF2 in vitro and in vivo. In conclusion, the data presented identify QSOX1 as a novel candidate target for sorafenib-based combination therapeutic strategies in HCC or other EGFR-dependent tumor types.The fine structure of the female reproductive system of a cheyletid mite Bakericheyla chanayi (Trombidiformes Cheyletidae) is investigated for the first time. This system consists of an unpaired ovary, glandular oviduct, receptaculum seminis, long cuticle-lined vagina, and genital atrium terminating in the genital opening. A separate sperm access system has not been found. The receptaculum seminis opens into the distal oviduct region, where fertilization apparently takes place. The ovary contains clusters of oogonia (cystocytes), clustered early meiotic cells, a few growing previtellogenic oocytes, and 3 large nurse cells. The dorsal ovarian region is occupied by the clusters of bacteriocytes which harbor symbiotic bacteria. Oocytes undergo vitellogenesis in individual ovarian pouches, each connected to the corresponding nurse cell by an intercellular bridge. The fine structure of the bridge suggests transport between the interconnected cells in the course of vitellogenesis. The population of cystocytes was shown to be heterogenic. The electron-light cells enter meiosis and develop into the oocytes or nurse cells. The electron-dense cystocytes do not show meiotic transformation and probably give rise to the bacteriocytes.
