Under adverse circumstances, bacteria enter the viable but non-culturable (VBNC) state, a dormancy-like state for survival. The altered gene regulation underlying the entry of the VBNC state has not yet been well elucidated. Here, we reported that a subpopulation of cells (23.8 %) in Vibrio alginolyticus cultures enters the VBNC state in response to nutrient limitation at alkaline pH. The proteolysis of pivotal virulence regulator ToxR at these conditions is associated with VBNC formation. Meantime, ToxR abrogation impaired the mobility and the expression of virulence-associated genes, resulting in attenuated virulence in V. alginolyticus. RNA-seq and ChIP-seq analyses of the cells grown in VBNC-inducing conditions revealed that ToxR directly controls the expression of ∼8 genes including ahpC and dps involved in reactive oxygen species (ROS) resistance. ToxR binds to the promoter regions of kdgR, ppiC, ahpC, and dps and further controls their respective expression under oxidative stress conditions. The cells with impaired ToxR accumulated detrimental intracellular ROS. Moreover, these genes contribute to bacterial culturability as their in-frame deletion strains exhibiting severely decreased plate counts and the complementary strain showed rescued viability. Collectively, this study revealed the role of ToxR in switching on the VBNC state by sensing unfavorable environmental signals such as endogenous ROS (hydrogen peroxide, H2O2) in V. alginolyticus and provided mechanistic insights into Vibrio lifestyle adaptation in the marine environment.A strategy is proposed to develop a stationary phase for hydrophilic interaction liquid chromatography (HILIC) using the synergistic effect of polyhedral oligomeric silsesquioxane (POSS) and copolymer brushes. Octahedral octa-aminopropylsisesquioxane (8NH2-POSS) was first bound to silica gel, followed by bromination to form a cubic initiator. Then, using acrylamide (AM) and dihydroxypropyl methacrylate (DPMA) as mixed monomers, surface initiated-atom transfer radical polymerization was conducted to prepare a stationary phase comprising cubic copolymer brushes with amide and diol groups. The characterization of the stationary phase confirmed the successful synthesis of Sil-NH2-POSS/Poly(AM-co-DPMA). The chromatographic properties were investigated using nucleosides, organic acids and β-agonists to find that our designed column has superior hydrophilic property, better separation performance compared with classical HILIC columns consisting of diol- or amino-modified silica. The systematic investigation of the retention mechanism and separation selectivity using various types of polar compounds revealed that Sil-NH2-POSS/Poly(AM-co-DPMA) follows a mixed-mode retention composed of HILIC and electrostatic interactions. Besides, it exhibits good column efficiency and stability. The role of 8NH2-POSS in the separation was evaluated by comparing the performance of Sil-NH2-POSS/Poly(AM-co-DPMA) and poly(AM-co-DPMA)-modified silica without 8NH2-POSS. In conclusion, our designed based on POSS and hydrophilic copolymer brushes can contribute to the development of HILIC separation materials with enhanced performance.Biogenic amines are quality control criteria for foods that are potentially toxic to humans. In this study, amidation derivatization for biogenic amines and liquid-solid phase transition microextraction were carried out simultaneously for food sample pretreatment. The derivatization reaction was executed in one pot with coumarin-3-carboxylic acid as the derivatizing reagent and (1-cyano-2-ethoxy-2-oxoethylidenaminooxy)dimethylamino-morpholino-carbenium hexafluorophosphate as the coupling agent. Liquid-solid phase transition microextraction was achieved by the salting-out effect, using a phase change salt (1 M disodium hydrogen phosphate) solution. The combined derivatization and microextraction process was completed within 3 min at 30 °C, and the liquid top phase was easily obtained by placing the tube in an ice bath. Finally, a narrowbore liquid chromatograph coupled with a UV detector was used to determine the levels of six biogenic amines. The coupling agent-assisted derivatization and liquid-solid phase transition microextraction parameters were also investigated. The quantitative linear ranges were 3-400 μM for histamine, putrescine, spermidine, cadaverine, and tyramine and 5-400 μM for spermine, and the detection limit was 1 μM. The relative standard deviations of the intra- and inter-batches were less then 5.3% and 8.4%, respectively, while the relative error was less then 4.5% for both. We successfully applied this simultaneous derivatization-microextraction method to determine the biogenic amines in fermented foods.Currently, the shape and variance of the analyte band entering the second dimension column when injected from an open loop interface in two-dimensional liquid chromatography is not fully understood. This is however important as it is connected to several other variables encountered when developing 2D-LC methods, including the first dimension flow rate, the sampling (modulation) time and the loop volume. Both numerical simulation methods and experimental measurements were used to understand and quantify the dispersion occurring in open tubular interface loops. Variables included are the analyte diffusion coefficient (Dmol), loop filling and emptying rates (Ffill & Fempty), loop inner diameter or radius (Rloop) and loop volume (Vloop). For a straight loop capillary, we find that the concentration profile (as measured at the loop outlet) depends only on a single dimensionless parameter tempty*=VloopFempty·DmolRloop2 and the ratio of the filling and emptying flow rates Fempty/Ffill. A model depending only on these two parameters was developed to predict of the peak variance resulting from the filling and emptying of a straight capillary operated in the first-in-last-out (FILO) modulation mode. Comparison of the concentration profiles and the corresponding variances obtained by either numerical simulation or experiments with straight capillaries shows the results generally agree very well. When the straight capillary is replaced by a tightly coiled loop, significantly smaller (20-40%) peak variances are observed compared to straight capillaries. The magnitude of these decreases is not predicted as well by simulations, however the simulation results are still useful in this case, because they represent an upper boundary (i.e., worst-case scenario) on the predicted variance.

To investigate the hidden curriculum of professionalism in the emergency department.

This is a qualitative study that consisted of six in-depth semi-structured interviews and a focus group with psychiatry residents of a teaching hospital program about their perception of professionalism in emergency department. A simple content analysis method has been conducted for the extraction of findings.

Common themes that were detected in the hidden curriculum were "Disregarding the priority of the patients"; "Overriding patient autonomy"; "Non-empathetic care"; "Breaching Confidentiality"; "Injustice in providing care"; and "Dishonesty" PRACTICE IMPLICATIONS Professional behaviors are mostly learned through the hidden curriculum. The hidden and formal curricula of professionalism are different in several domains. Paying attention to organizational culture and social environment is critical for promoting professionalism.

Major depressive disorder is the leading cause of non-fatal burden, and disability in adulthood. Even though depression is well-treated in the acute term,psychosocial functioning does not get back to the premorbid level most of the time. In this present study, it is aimed to evaluate the outcome of the acute term treatment of major depressive disorder in terms of psychosocial functioning.

The study is an open-label, observational, multi-center follow-up study for four months of patients with major depressive disorder according to DSM-5. Patients were evaluated with Montgomery Asberg Depression Rating Scale (MADRS), Sheehan Disability Scale (SDS) and Short Form-36 (SF-36) at the beginning, and at the 2., 4., 8., 12. and 16.weeks.

100 patients were invited to the study and 56 patients completed the study.As a result of the treatment, the mean MADRS and SDS scores decreased significantly. All domains of SF-36 were improved significantly with the treatment. Unfortunately patients suffering from MDD could not reach the normative data,especially on the domains of social functioning, role emotional, pain, and general health perception. Treatment outcomes show that SNRI users presented higher scores on the domains of pain and physical functioning. However SSRI users showed better outcomes on the domains of mental health and vitality.
