Several fungal endophytes were isolated from some medicinal plants and screened for their ability to produce the anticancer drug vinblastine.
An isolate was found to produce vinblastine (205·38μgl
), and the identity of the fungal vinblastine was confirmed by UV spectroscopic, high-performance liquid chromatography and electrospray ionization mass spectrometry analyses. Based on both morphological and molecular studies, the vinblastine-producing strain was identified as Alternaria alternata. Cytotoxic activities of the fungal vinblastine were evaluated against CHO-K1, MCF-7 and HepG-2 cell lines by the MTT assay. The proliferation of these cell lines was inhibited after treatment with fungal vinblastine and the recorded IC
values of the respective cell lines were 12·15, 8·55 and 7·48μgml
. A strain improvement programme for improving vinblastine productivity by the fungal strain was also used. In addition, 10 broth media were evaluated for further increasing the production of vinblastine. The yield of vinblastine was intensified by 3·98-fold following gamma irradiation at 1000Gy, and a stable mutant strain was isolated. Among the screened media, M1D broth (pH 6·0) stimulated the highest vinblastine production of 1553·62μgl
by the isolated mutant strain.

Hypomineralised second primary molar (HSPM) is a developmental enamel defect associated with dental caries.
To evaluate the impact of HSPM on oral health-related quality of life (OHRQoL) in preschoolers through hierarchical analysis.
This population-based cross-sectional study assessed 834 5-year-old preschoolers, in Teresina, Piauí, Brazil, using sociodemographic questionnaires and Early Childhood Oral Health Impact Scale (ECOHIS). Dental examination was performed by two calibrated examiners (κ≥0.80) for the diagnosis of HSPM (EAPD), dental caries (dmft and pufa indices), and malocclusion (Foster and Hamilton criteria). Analysis of the determinants of OHRQoL was stratified at three levels (P<.05).
Severe HSPM was associated with OHRQoL in bivariate analysis (P<.05). In adjusted final multivariate analysis (model 1), the presence of dental caries with or without clinical consequences was associated with worse OHRQoL (P<.05). In model 2, after the removal of dental caries, preschoolers with severe HSPM had greater probability of having a negative impact on OHRQoL in the child impact section (RR=2.14; 95% CI=1.26-3.65), family impact section (RR=1.99; 95% CI=1.16-3.42), and total score (RR=2.09; 95% CI=1.25-3.49) of the ECOHIS.

ZO-1 is a key regulatory tight junction protein that plays an important role in maintaining gastrointestinal health. In this study, we investigated the protective effect and regulation mechanism of the probiotic Enterococcus faecium HDRsEf1 on tight junction protein ZO-1 at the cellular and molecular levels.
We established LPS-induced intestinal epithelial cell injury model, and detected the protective effect of HDRsEf1 on ZO-1 in IPEC-J2 cells by Real-time PCR and Western blot. The results showed that HDRsEf1 inhibited the downregulation of ZO-1 expression induced by LPS. HDRsEf1 stabilized the destruction of the ZO-1 structure caused by LPS in an immunofluorescence assay. Through gene overexpression and siRNA interference tests, we found that transcription factor AP-1 inhibited the level of ZO-1 expression. Silencing experiment further supported that the protective effect of HDRSEF1 might mediated by suppression of LPS-provoked activation of ASK1/MKK7/JNK signalling pathways. In addition, HDRsEf1 could stabilize ZO-1 expression by increasing TLR2 expression and competing with LPS for the TLR4 binding site.
