Pretreatment with endoplasmic reticulum stress (ER stress) inhibitor tauroursodeoxycholic acid and knockdown of PERK expression attenuated the degradation of Mcl-1 caused by PEITC. In in vivo study, nude mice bearing NCI-H1299 xenograft were administrated with PEITC (50 mg/kg, ip) and Gefitinib (50 mg/kg, ig) for 15 days, the PEITC-Gefitinib combination treatment resulted in a significant synergistic reduction in tumor growth, and significantly induced both ER stress and Mcl-1 degradation in tumor tissues. In conclusion, we explored the prospect of PEITC in improving the efficacy of targeted drug therapy and demonstrated the synergistic effects and underlined mechanisms of PEITC combined with Gefitinib in NSCLC cells treatment. This study provided useful information for developing novel therapy strategies by combination treatment of PEITC with targeted drugs. © 2020 Wiley Periodicals, Inc.OBJECTIVES We assessed the incidence, risk factors and adverse birth outcomes associated with elevated liver enzymes and low platelets (HELLP) syndrome. DESIGN A retrospective population-based cohort study. SETTING Canada (excluding Quebec), 2012/13-2015/16. POPULATION Mothers with a singleton hospital live birth or stillbirth at ≥24 weeks gestation (N=1,078,323). METHODS HELLP syndrome was identified using ICD-10-CA diagnostic code from delivery hospitalization data. Congo Red We used logistic regression to identify independent risk factors for HELLP syndrome by obtaining adjusted odds ratios (AOR) and 95% confidence intervals (CI), and to assess the associations with adverse outcomes. MAIN OUTCOME MEASURES Adverse maternal (e.g., eclampsia) and fetal/neonatal outcomes (e.g., intraventricular hemorrhage, perinatal death). RESULTS The incidence of HELLP syndrome was 2.5 per 1,000 singleton deliveries (n=2,663). Risk factors included age ≥35 years, rural residence, nulliparity, parity ≥4, pre-pregnancy and gestational hypertension and diabetes, assisted reproduction, chronic cardiac conditions, systemic lupus erythematosus, obesity, chronic hepatic conditions, placental disorders (e.g., fetomaternal transfusion), and congenital anomalies; PROM and age less then 25 years were inversely associated with HELLP syndrome (p-values less then 0.05). Women with syndrome had a 10-fold higher maternal mortality (95% CI 1.6-84.3) and elevated severe maternal morbidity (9.6 vs. 121.7 per 1,000; AOR=12.5, 95% CI 11.1-14.1); and higher perinatal mortality (4.3 vs. 21.0 per 1,000; AOR=4.5, 95%CI 3.5-5.9) and perinatal mortality/severe neonatal morbidity (21.2 vs 202.4 per 1,000; AOR=10.7, 95% CI 9.7-11.8). CONCLUSION HELLP syndrome is associated with specific pre-pregnancy and pregnancy risk factors, higher rates of maternal death, and substantially higher severe maternal morbidity, perinatal mortality and severe neonatal morbidity. We demonstrate that laser irradiation of the molecular crystal can generate phase-dependent transient elastic lattice deformation . The resulting elastic lattice deformation that follows scanning irradiation of a laser can actuate the different phases of molecular crystal to move with different velocity and direction. Because the γ phase has a large Young's modulus (ca. 26 GPa), a force of 0.1 μN can be generated under one laser spot. The generated force is sufficient to actuate the γ-formed molecular crystals in a wide dimensional range to move longitudinally at a velocity of ca. 60 μm min -1 , which is two orders of magnitude faster than the α and β phases. © 2020 WILEY-VCH Verlag GmbH & Co. Approximately 5-20% of women at their reproductive age are diagnosed with endometriosis, which causes chronic pain and infertility. Here, we demonstrated that the bioactive flavonoid, 5,7-dimethoxyflavone (DMF), exhibited antiproliferative and apoptotic effects in VK2/E6E7 and End1/E6E7 cells which were established from vaginal and endocervical tissue taken from a premenopausal woman undergoing hysterectomy for endometriosis. DMF treatment significantly elevated DNA fragmentation resulting in apoptotic cell death in both cell lines. Furthermore, DMF induced loss of mitochondrial membrane potential, dysregulation of intracellular calcium level, and ROS production, which accelerate apoptosis. Additionally, DMF modulated the expression of the signaling molecules related to cell survival and endoplasmic reticulum stress in VK2/E6E7 and End1/E6E7 cells. Overall, DMF may ameliorate endometriosis and can be a potential alternative to hormonal and surgical therapy for endometriosis treatment. © 2020 John Wiley & Sons, Ltd.Cerebral small vessel disease (cSVD) is a leading cause of stroke and lies at the heart of vascular contributions to dementia. AIM A cross-sectional questionnaire survey was undertaken to describe the management strategies for DCL of vital primary teeth, focusing on the different caries removal techniques, among dentists practicing pediatric dentistry (DPPDs) in France. Their behavior was compared to members' one registered to European Academy of Pediatric Dentistry (EAPD). DESIGN A questionnaire was electronically administrated (2018-2019) to members of the Collège des Enseignants en Odontologie Pédiatrique (CEOP), the Société Française d'Odontologie Pédiatrique (SFOP) and the EADP. Descriptive and statistical analyses were performed. RESULTS Response rate were respectively for CEOP, SFOP and EAPD about 74%, 29% and 15%. About half of the respondents (53%) would perform a complete caries removal into one step when 12% would indicate a stepwise technique 68% of the DPPDs practicing in France would perform complete caries removal in one step when the preferred option in the other EAPD members was the selective excavation (44%) (p less then 0.
