Melatonin is an important bioactive molecule in plants. Two synthetases, N-acetylserotonin methyltransferase (ASMT) and serotonin N-acetyltransferase (SNAT) are involved in the final two steps of melatonin synthesis. Selleckchem Ciforadenant Melatonin participates in responses to a variety of biotic and abiotic stresses in plants, but few studies have addressed the roles of endogenous melatonin in pathogen resistance. We investigated the role of endogenous melatonin in resistance to Botrytis cinerea infection in an Arabidopsis thaliana model system. Plant lines that overexpressed ASMT or SNAT through genetic manipulation showed upregulated expression of resistance genes PR1 and PR5, transcription factor gene WRKY33, and jasmonic acid (JA) defense pathway marker gene PDF1.2, and downregulated transcription factor gene MYC2 in JA signaling pathway. Higher melatonin content also enhanced the activity of antioxidant enzymes superoxide dismutase (SOD) and peroxidase (POD), increased JA content, reduced plant disease symptoms, and reduced lesion size in leaves. These findings indicate that endogenous melatonin enhances plant resistance to B. cinerea infection. In contrast, ASMT and SNAT gene silencing lines showed opposite results and were more susceptible to B. cinerea. Thus, it can be demonstrated that melatonin functions as an effective regulator of plant stress resistance at the genetic level. A schematic model is presented for its role in resistance to B. cinerea infection. Our findings also helped to elucidate the associated signal transduction pathways and interactions between melatonin and other plant hormones.Halophytes are capable of resisting salinity, and their root system is the part in direct contact with the saline soil environment. The aim of this study was to compare the responses of root morphology and rhizosphere characteristics to salinity between a halophyte, Suaeda salsa (suaeda), and a glycophyte, Beta vulgaris L. (sugar beet). The soil salt content was set to four levels (0.7, 1.2, 1.7, and 2.7%) by NaCl-treated plants. We investigated the soil pH, EC, nutrients and soil, plant ion (Na+, Cl-, K+, and Mg2+) concentration to evaluate the rhizospheric processes, and salt tolerance of suaeda by the root mat method. The highest biomass was in the 1.2% salt level for suaeda and in the 0.7% salt level for sugar beet. The root length and root surface area of suaeda showed similar trends to biomass, but the root diameter decreased by 11.5-17.9% with higher salinity. The Na+, Cl-, and K+ accumulations in the shoot of suaeda displayed higher than that in sugar beet, while the Mg2+ accumulation was lower in suaeda than that in sugar beet. High salinity resulted in increased pH and EC values in the rhizosphere for suaeda, but lower values of these parameters for sugar beet. Under high salinity, the Olsen phosphorus content was 0.50 g·kg-1 and 0.99 g·kg-1 higher in the rhizosphere than in the non-rhizosphere for suaeda and sugar beet. We concluded that the two species [halophyte, Suaeda salsa (suaeda), and a glycophyte, B. vulgaris L. (sugar beet)] showed diverse approaches for nutrient absorption under salinity stress. Suaeda altered its root morphology (smaller root diameter and longer roots) under salt stress to increase the root surface area, while sugar beet activated rhizospheric processes to take up more nutrients.In order to analyze the effect of cadmium in Ulva compressa (Chlorophyta), the alga was cultivated with 10, 25, and 50 μM of cadmium for 7 days, and the level of intracellular cadmium was determined. Intracellular cadmium showed an increase on day 1, no change until day 5, and an increase on day 7. Then, the alga was cultivated with 10 μM for 7 days, and the level of hydrogen peroxide, superoxide anions, and lipoperoxides; activities of antioxidant enzymes ascorbate peroxidase (AP), dehydroascorbate reductase (DHAR), and glutathione reductase (GR); the level of glutathione (GSH) and ascorbate (ASC); and the level of phytochelatins (PCs) and transcripts encoding metallothioneins (UcMTs) levels were determined. The level of hydrogen peroxide increased at 2 and 12 h, superoxide anions on day 1, and lipoperoxides on days 3 to 5. The activities of AP and GR were increased, but not the DHAR activity. The level of GSH increased on day 1, decreased on day 3, and increased again on day 5, whereas ASC slightly increased on days 3 and 7, and activities of enzymes involved in GSH and ASC synthesis were increased on days 3 to 7. The level of PC2 and PC4 decreased on day 3 but increased again on day 5. The level of transcripts encoding UcMT1 and UcMT2 increased on days 3 to 5, mainly that of UcMT2. Thus, cadmium accumulation induced an oxidative stress condition that was mitigated by the activation of antioxidant enzymes and synthesis of GSH and ASC. Then, the alga cultivated with inhibitors of calcium-dependent protein kinases (CDPKs), calmodulin-dependent protein kinases (CaMKs), calcineurin B-like protein kinases (CBLPKs), and MAPKs and 10 μM of cadmium for 5 days showed a decrease in intracellular cadmium and in the level of GSH and PCs, with the four inhibitors, and in the level of transcripts encoding UcMTs, with two inhibitors. Thus, CDPKs, CaMK, CBLPKS, and MAPKs are involved in cadmium accumulation and GSH and PC synthesis, and GSH and PCs and/or UcMTs may participate in cadmium accumulation.Salt stress is an important environmental limiting factor. Water dropwort (Oenanthe javanica) is an important vegetable in East Asia; however, its phenotypic and physiological response is poorly explored. For this purpose, 48 cultivars of water dropwort were grown hydroponically and treated with 0, 50, 100, and 200 mm NaCl for 14 days. Than their phenotypic responses were evaluated, afterward, physiological studies were carried out in selected sensitive and tolerant cultivars. In the present study, the potential tolerant (V11E0022) and sensitive (V11E0135) cultivars were selected by screening 48 cultivars based on their phenotype under four different levels of salt concentrations (0, 50, 100, and 200 mm). The results depicted that plant height, number of branches and leaves were less effected in V11E0022, and most severe reduction was observed in V11E0135 in comparison with others. To this end, assessment of genetic gain for key traits is important to inform breeding processes. Despite the improvements made to increase production and productivity of cassava in Uganda at research level, there has been limited effort to quantify associated genetic gains. Accordingly, a study was conducted in Uganda to assess whether or not genetic improvement was evident in selected cassava traits using cassava varieties that were released from 1940 to 2019. Thirty-two varieties developed during this period, were evaluated simultaneously in three major cassava production zones; central (Namulonge), eastern (Serere), and northern (Loro). Best linear unbiased predictors (BLUPs) of the genotypic value for each clone were obtained across environments and regressed on order of release year to estimate annual genetic gains. We observed that genetic trends were mostly quadratic. On average, cassava mosaic disease (CMD) resistance increased by 1.9% per year, while annual genetic improvements in harvest index (0.0%) and fresh root yield (-5 kg per ha or -0.03% per ha) were non-substantial. For cassava brown streak disease (CBSD) resistance breeding which was only initiated in 2003, average annual genetic gains for CBSD foliar and CBSD root necrosis resistances were 2.3% and 1.5%, respectively. It's evident that cassava breeding has largely focused on protecting yield against diseases. This underpins the need for simultaneous improvement of cassava for disease resistance and high yield for the crop to meet its current and futuristic demands for food and industry.Salinity affects plant growth and development as shown with the glycophyte model plant, Arabidopsis thaliana (Arabidopsis). Two Arabidopsis accessions, Wassilewskija (Ws) and Columbia (Col-0), are widely used to generate mutants available from various Arabidopsis seed resources. However, these two ecotypes are known to be salt-sensitive with different degrees of tolerance. In our study, 3-week-old Col-0 and Ws plants were treated with and without 150 mM NaCl for 48, 72, or 96 h, and several physiological and biochemical traits were characterized on shoots to identify any specific traits in their tolerance to salinity. Before salt treatment was carried out, a different phenotype was observed between Col-0 and Ws, whose main inflorescence stem became elongated in contrast to Col-0, which only displayed rosette leaves. Our results showed that Col-0 and Ws were both affected by salt stress with limited growth associated with a reduction in nutrient uptake, a degradation of photosynthetic pigments, an increase in icipate in CWI. Altogether, these data indicate that Col-0 and Ws trigger similar mechanisms to cope with salt stress, and specific changes are more likely related to the developmental stage than to their respective genetic background.Knowing the nutrient removal by soybean grain harvest in different varieties, locations, and over time is essential to correctly adjust agronomic recommendations, update farmers' practices, and increase nutrient use efficiency. A field-based research trial was carried out to assess macronutrients [nitrogen (N), phosphorus (P), potassium (K), magnesium (Mg), calcium (Ca), and sulfur (S)] removed in grain by modern soybean varieties from southern Brazil introduced between 2007 and 2016. We examined changes between our set of modern varieties and a dataset of historical values encompassing a wide range of varieties introduced before 2007. Moreover, we undertook a synthesis analysis using scientific literature published after 2007 to investigate nutrient removal by grain among modern Brazilian soybeans and a dataset that included field trials from Argentina, United States, and India. There were no yield gains across the years for modern soybean varieties introduced among 2007 and 2016 in Brazil, although the graiin nutrient removal patterns between Brazil and other countries indicates an integrated effect of management, genotype, and environment on nutrient removal. These findings provide guidance for optimal nutrient management and specific information for plant breeding programs to understand nutrient variability.While biochar use in agriculture is widely advocated, how the effect of biochar on plant growth varies with biochar forms and crop genotypes is poorly addressed. The role of dissolvable organic matter (DOM) in plant growth has been increasingly addressed for crop production with biochar. In this study, a hydroponic culture of rice seedling growth of two cultivars was treated with bulk mass (DOM-containing), water extract (DOM only), and extracted residue (DOM-free) of maize residue biochar, at a volumetric dosage of 0.01, 0.05, and 0.1%, respectively. On seedling root growth of the two cultivars, bulk biochar exerted a generally negative effect, while the biochar extract had a consistently positive effect across the application dosages. Differently, the extracted biochar showed a contrasting effect between genotypes. In another hydroponic culture with Wuyunjing 7 treated with biochar extract at sequential dosages, seedling growth was promoted by 95% at 0.01% dosage but by 26% at 0.1% dosage, explained with the great promotion of secondary roots rather than of primary roots.
