Atopic dermatitis (AD) can affect multiple body regions and is especially burdensome when involving exposed skin areas. Rapid, effective treatment of AD across body regions remains an unmet need, particularly for difficult-to-treat areas such as the head and neck area. We investigated the temporal and regional patterns of clinical improvement in AD with the use of abrocitinib, an orally available Janus kinase 1 selective inhibitor under development for the treatment of moderate-to-severe AD.

We performed a post hoc analysis of data from JADE COMPARE, a phase 3, multicenter, randomized, double-blind, double-dummy trial that evaluated the efficacy and safety of abrocitinib 200mg once daily, abrocitinib 100mg once daily, dupilumab 300mg subcutaneous injection every 2weeks, and placebo in adult patients with moderate-to-severe AD who were concomitantly receiving medicated topical therapy. Assessments included the Eczema Area and Severity Index (EASI) and SCORing Atopic Dermatitis (SCORAD) index.

With abrocitinib 200mg, time to ≥ 75% improvement in EASI (EASI-75) occurred at a median of 29days across body regions, including the head and neck region. With abrocitinib 100mg, EASI-75 response was achieved at a median of 30-32days for the trunk and lower limbs, and at 56-57days for the head and neck region and upper limbs. With dupilumab, EASI-75 response was achieved at a median of 43days for the trunk and 57days for other regions. EASI body region scores significantly improved with abrocitinib 200mg and 100mg versus placebo at week 2 (p < 0.0001 for all comparisons). Improvements with abrocitinib were maintained up to week 16.

Rapid and persistent improvement in AD across all body regions was observed with abrocitinib treatment. Abrocitinib may be useful in patients with AD that affects difficult-to-treat anatomical areas or who require a rapid response.

He had no history of human immunodeficiency virus (HIV) infection or treatment with immunosuppressive agents. We performed renal biopsy to investigate possible acute kidney injury. Pathological findings showed inflammatory cell infiltration, including granulomatous lesions in the interstitium. We diagnosed the patient with acute granulomatous tubulointerstitial nephritis. We initiated prednisolone (PSL) 40 mg/day (0.6 mg/kg), in combination with isoniazid for a latent tuberculosis infection, because of positive results in interferon-γ release assays. The patient's fever and malaise promptly disappeared, and his renal function improved. After the patient had been discharged, Mycobacterium intracellulare grew in cultures of his renal tissue and urine. We gradually reduced the dose of PSL; we initiated combination therapy with ethambutol, clarithromycin, and rifampin. After 2 years of follow-up, the patient continued treatment for chronic kidney disease; it has since enabled him to avoid renal replacement therapy. This report describes a rare instance of nontuberculous mycobacteria-associated tubulointerstitial nephritis in a patient without a history of HIV infection or organ transplantation. In differential diagnosis of granulomatous tubulointerstitial nephritis, clinicians should consider drugs, sarcoidosis, tubulointerstitial nephritis and uveitis syndrome, vasculitis, and infections (e.g., involving mycobacteria). Prompt microbiological examinations, especially of urine or biopsy cultures, are vital for diagnosis.The multi-attribute method has been recognized as an elegant quantification tool for post-translational modifications (PTMs) of therapeutic proteins, since it can evaluate several attributes spontaneously and site-specifically. Here, the abundance of PTMs calculated by three different types of formula were compared and there was little difference among the results. For the method evaluation, two different kinds of peptides were used as internal standards (ISs) and one of the IS was used as the "standard peak" to define the signal strength of MS. A standard account is that people develop a sense of the size of symbolic numbers by learning to map symbolic numbers (e.g., 6) to their corresponding numerosities (e.g. ) and concomitant approximate magnitude system (ANS) representations. However, we here demonstrate that adults are capable of extracting fractional numerical quantities from non-symbolic visual ratios (i.e., labeling a ratio of two circle areas with the appropriate symbolic fraction). Not only were adult participants able to perform this task, but they were remarkably accurate linear regressions on median estimates yielded slopes near 1, and accounted for 97% of the variability. Participants also performed at least as well on line-estimation and ratio-estimation tasks using non-numeric circular stimuli as they did in earlier experiments using non-symbolic numerosities, which are frequently considered to be numeric stimuli. We discuss results as consistent with accounts suggesting that non-symbolic ratios have the potential to act as a reliable and stable ground for symbolic number, even when composed of non-numeric stimuli.Attention has multiple influences on visual working memory (VWM). Fluctuations in sustained attention predict VWM performance. Furthermore, focusing selective attention in VWM by retro-cuing the to-be-tested item during maintenance boosts retrieval. So far, we lack knowledge how the ability to focus selective attention relates to the state of sustained attention during the VWM trial. Here, we combined a retro-cue task and a self-rated attention protocol to test whether focusing selective attention via retro-cues (1) mitigates spontaneous attention fluctuations, in which case retro-cues should be more helpful under low levels of self-rated attention; (2) depends on an optimal state of sustained attention, in which case retro-cue benefits should be largest under high levels of self-rated attention; or (3) is independent of sustained attention, in which case retro-cue benefits and self-rated attention effects should be additive. Our data supported the additive hypothesis. Across four experiments, self-rated attention levels predicted continuous reproduction of colors. Retro-cue trials produced better recall and higher rated attention. Critically, retro-cues improved recall to a similar extent across all levels of self-rated attention. This indicates that attention has multi-faceted and independent contributions to VWM.Real-time quality assessment (rtQA) of functional magnetic resonance imaging (fMRI) based on blood oxygen level-dependent (BOLD) signal changes is critical for neuroimaging research and clinical applications. The losses of BOLD sensitivity because of different types of technical and physiological noise remain major sources of fMRI artifacts. Due to difficulty of subjective visual perception of image distortions during data acquisitions, a comprehensive automatic rtQA is needed. To facilitate rapid rtQA of fMRI data, we applied real-time and recursive quality assessment methods to whole-brain fMRI volumes, as well as time-series of target brain areas and resting-state networks. We estimated recursive temporal signal-to-noise ratio (rtSNR) and contrast-to-noise ratio (rtCNR), and real-time head motion parameters by a framewise rigid-body transformation (translations and rotations) using the conventional current to template volume registration. In addition, we derived real-time framewise (FD) and micro (MD) dispof fMRI acquisitions by means of substantiating decisions about the necessity of the interruption and re-start of the experiment and increasing the confidence in neural estimates.Masked priming is one of the most important paradigms in the study of visual word recognition, but it is usually thought to require a laboratory setup with a known monitor and keyboard. To test if this technique can be safely used in an online setting, we conducted two online masked priming lexical decision task experiments using PsychoPy/PsychoJS (Peirce et al., 2019). Importantly, we also tested the role of prime exposure duration (33.3 vs. 50 ms in Experiment 1 and 16.7 vs. 33.3 ms in Experiment 2), thus allowing us to examine both across conditions and within-conditions effects. We found that our online data are indeed very similar to the masked priming data previously reported in the masked priming literature. Additionally, we found a clear effect of prime duration, with the priming effect (measured in terms of response time and accuracy) being stronger at 50 ms than 33.3 ms and no priming effect at 16.7 ms prime duration. From these results, we can conclude that modern online browser-based experimental psychophysics packages (e.g., PsychoPy) can present stimuli and collect responses on standard end user devices with enough precision. These findings provide us with confidence that masked priming can be used online, thus allowing us not only to run less time-consuming experiments, but also to reach populations that are difficult to test in a laboratory.This paper introduces the CommonLit Ease of Readability (CLEAR) corpus, which provides unique readability scores for ~ 5000 text excerpts along with information about the excerpt's year of publishing, genre, and other metadata. The CLEAR corpus will provide researchers interested in discourse processing and reading with a resource from which to develop and test readability metrics and to model text readability. The CLEAR corpus includes a number of improvements in comparison to previous readability corpora including size, breadth of the excerpts available, which cover over 250 years of writing in two different genres, and unique readability criterion provided for each text based on teachers' ratings of text difficulty for student readers. This paper discusses the development of the corpus and presents reliability metrics for the human ratings of readability.Facial expressions play an essential role in social interactions. Databases of face images have furnished theories of emotion perception, as well as having applications in other disciplines such as facial recognition technology. However, the faces of many ethnicities remain largely underrepresented in the existing face databases, which can impact the generalizability of the theories and technologies developed based on them. ATN161 Here, we present the first survey-validated database of Iranian faces. It consists of 248 images from 40 Iranian individuals portraying six emotional expressions-anger, sadness, fear, disgust, happiness, and surprise-as well as the neutral state. The photos were taken in a studio setting, following the common scenarios of emotion induction, and controlling for conditions of lighting, camera setup, and the model's head posture. An evaluation survey confirmed high agreement between the models' intended expressions and the raters' perception of them. The database is freely available online for academic research purposes.
