Superoxide dismutase (SOD) and peroxidase (POX) activities in hairy roots of both species were higher as compared to other organs. The hairy roots of both species showed an ability to synthesize steroidal sapogenins. These results presented that hairy roots could be a suitable procedure for producing sapogenins compounds that have medicinal value in Trigonella. © Prof. H.S. Srivastava Foundation for Science and Society 2020.Citrus limon (L.) Osbeck cultivated all over the world is a valuable source of aromatic essential oil. To develop tetraploids of C. limon, four different concentration of colchicine (0.025, 0.05, 0.1 and 0.2%; w/v) and three varied exposure time (12, 24 and 48 h) were employed. The ploidy level of diploids (2n = 2x = 18) and tetraploids (2n = 4x = 36) were determined by direct chromosome count and confirmed by flow cytometric analyses. Successful result with maximum tetraploidy frequency was observed in plantlets developed from seeds treated with 0.025% colchicine for 24 h. Morphological and stomatal characteristics indicated that tetraploids were taller with increased leaf and root length. On the other side, the leaves of tetraploids had fewer and larger stomata with a greater number of chloroplasts in guard cells in contrast with diploids. GC-GC/MS analyses showed cyclic monoterpene, limonene had increased significantly in tetraploids and was further confirmed by HPLC quantification. RT-PCR analyses revealed unaltered expression of monoterpene synthase, sesquiterpene synthase and flavone synthase and remarkable upregulation of genes such as limonene synthase, chalcone synthase and phenylalanine ammonia lyase in tetraploids. Antioxidant activity of essential oil of tetraploids was higher than diploids in all the five test systems studied. Overall, the findings of the present study prove that colchicine induced tetraploidy in C. limon could be a greater source of essential oil with improved composition and of economic significance. © Prof. H.S. Srivastava Foundation for Science and Society 2019.Mesophyll conductance (g m) has received over time much less attention than stomatal conductance (g s), although it affects leaf photosynthesis to about the same extent as stomatal conductance does. The objective of this study was to analyze the g m trend in five understory herbaceous species growing in a close-canopy forest in the north-west of Italy. In particular, three of analyzed species were monocots Carex brizoides Lam., Carex pilosa Scop., and Oplismenus undulatifolius P. Beauv and the others dicots species Circaea lutetiana L., and Pulmonaria officinalis Ced. The results showed, on one hand, the absence of correlation between g m and the considered environmental variables in the forest understory (i.e. air temperature, photosynthetic photon flux density and carbon dioxide concentration). Moreover, we carried out a principal component analysis considering all the analyzed morphological and physiological variables for the five species. The following correlation between the first component, related to the leaf mass per unit of leaf area and the leaf tissue density, and g m seem to suggest a key role of the leaf structural features in determining g m variations across the five species. © Prof. H.S. Srivastava Foundation for Science and Society 2020.This study investigated the effects of different temperatures, photoperiods and spermidine concentrations on the flowering time regulation of Anoectochilus roxburghii by measuring changes in the soluble sugar, soluble protein, malondialdehyde and proline contents, and the peroxidase, superoxide dismutase and catalase activities in A. ARRY-382 solubility dmso roxburghii flower buds. The flowering time could be advanced under 25/20 °C (day/night), 16/8-h (day/night) long day conditions or low spermidine concentrations. The plants grew more rapidly and flowering rates were greater. The flowering time could be delayed under a low temperature of 20/15 °C or 8/16-h short day conditions, resulting in a low flowering rate. Under a high temperature of 30/25 °C or high spermidine concentrations, the plants could not flower normally and even died. There were significant differences in the seven measured indices among the various treatments. Thus, different treatments had significant effects on the flowering time regulation and flowering quality of A. roxburghii, providing a reliable theoretical basis for further studies on the flowering-related regulatory mechanisms of A. roxburghii. © Prof. H.S. Srivastava Foundation for Science and Society 2020.Polyamines (PAs) are positively charged molecules known to mitigate drought stress; however, little is known about their mechanism of alleviating drought stress. We investigated the effects of PAs exogenously applied as a seed primer and as a foliar spray on the growth, membrane stability (MS), electrolyte leakage (EL), Na+ and K+ cations, reactive oxygen species (ROS), catalase (CAT; EC 1.11.1.6) and guaiacol peroxidase (GPX; EC 1.11.1.7) activity and chloroplast ultra-structure in wheat (Triticum aestivum L.; cv. Sakha-94) under drought stress. Three PA solutions, namely, putrescine, spermine and a mixture of the two (Mix), were each applied at a concentration of 100 µM. Our study demonstrated that the retardation of chlorophyll loss and elevation of Rubisco levels were involved in PA-enhanced growth under drought stress. These relationships were mainly reflected in elevated fresh weight and dry weight in response to foliar spraying with all PA solutions and seed priming with the Mix solution. The elevated growth seemed to be due to increased photosynthetic pigments, protein and Rubisco. In contrast, drought decreased growth, photosynthetic pigments, protein and Rubisco. MS was enhanced by PAs applied as a seed primer or foliar spray, as shown by clear reductions in EL %, malondialdehyde (MDA) content and the Na+/K+ ratio as well as reduced ROS markers and elevated CAT (but not GPX) activity. Further study showed that the Mix solution of PAs, applied either during seed priming or as a foliar spray, improved chloroplast ultra-structure, suggesting that improvements in Rubisco and photosynthetic pigments were involved in PA maintenance of chloroplast stability. Therefore, the present study showed that elevated CAT activity is the main mechanism through which PAs reduce ROS and MDA, thereby improving MS and protecting mesophyll cells structurally and functionally under drought stress in wheat. © Prof. H.S.
