This paper describes a selenium-promoted electrophilic cyclization of arylpropiolamides allowing the synthesis of 3-organoselenyl spiro[4,5]trienones via a 5-endo-dig ipso-mode. The 3-organoselenyl-quinolinone derivative formation via 6-endo-dig was avoided using an electrophilic organoselenium species in a metal-free protocol. The use of phenylselenyl bromide (1.3 equiv.), as the electrophilic source, in nitromethane (3 mL) at 90 °C enabled the cyclization of N-(2-methoxyphenyl)-N-methyl-3-phenylpropiolamides, giving 3-organoselenyl[4,5]triene-2,6-dione derivatives. The extension of the standard conditions to the N-(4-methoxyphenyl)-phenylpropiolamides led to the corresponding 3-organoselenyl spiro[4,5]trienones having the carbonyl group at the 8-position. Besides, we demonstrated a general application of our approach by using 3-organoselenyl spiro[4,5]trienones as substrates in Suzuki cross-coupling reactions, which gave the cross-coupled products in good yields.Coadsorption experiments reveal an unexpected increase of the D2/H2 selectivity with loading in pure silica chabazite at 47 K. This effect is correlated with the appearance of a step in the adsorption isotherms of H2 and D2. Grand canonical Monte Carlo simulations show that this phenomenon is related to a molecular rearrangement of the adsorbed phase induced by its strong confinement. In the case of a H2 and D2 mixture, this rearrangement favors the adsorption of D2 having a smaller size due to quantum effects.CRISPR-Cas9 is a versatile genome-editing technology that is a promising gene therapy tactic. However, the delivery of CRISPR-Cas9 is still a major obstacle to its broader clinical application. Here, we confirm that the components of CRISPR-Cas9-sgRNA and Cas9 protein-can be packaged into exosomes, where sgRNA and Cas9 protein exist as a sgRNACas9 ribonucleoprotein complex. Although exosomal CRISPR-Cas9 components can be delivered into recipient cells, they are not adequate to abrogate the target gene in recipient cells. To solve this, we engineered a functionalized exosome (M-CRISPR-Cas9 exosome) that could encapsulate CRISPR-Cas9 components more efficiently. To improve the loading efficiency of Cas9 proteins into exosomes, we artificially engineered exosomes by fusing GFP and GFP nanobody with exosomal membrane protein CD63 and Cas9 protein, respectively. Therefore, Cas9 proteins could be captured selectively and efficiently loaded into exosomes due to the affinity of GFP-GFP nanobody rather than random loading. sgRNA and Cas9 protein exist as a complex in functionalized exosomes and can be delivered into recipient cells. To show the function of modified exosomes-delivered CRISPR-Cas9 components in recipient cells visually, we generated a reporter cell line (A549stop-DsRed) that produced a red fluorescent signal when the stop element was deleted by the sgRNA-guided endonuclease. Using A549stop-DsRed reporter cells, we showed that modified exosomes loaded with CRISPR-Cas9 components abrogated the target gene more efficiently in recipient cells. Our study reports an alternative tactic for CRISPR-Cas9 delivery.The potential role of borophene as a radiosensitizer in PT and BNCT was investigated. Our study focused on two aspects (1) the synthesis and characterization of borophene nanomaterials; and (2) biocompatibility and dose enhancement. To overcome the limitation of vapor-based technology, we successfully deployed the liquid-phase exfoliation (LPE) method to produce borophene targeting for biomedical applications. Bringing together spatial distribution and dose deposition, the in vitro microdosimetry study was carried out in the presence of borophene. A quantitative study of the dose enhancement ratio (DER) was performed with Monte-Carlo simulation.
