Prey-selective venoms and toxins have been documented across only a few species of snakes. The lack of research in this area has been due to the absence of suitably flexible testing platforms. In order to test more species for prey specificity of their venom, we used an innovative taxonomically flexible, high-throughput biolayer interferometry approach to ascertain the relative binding of 29 α-neurotoxic venoms from African and Asian elapid representatives (26 Naja spp., Aspidelaps scutatus, Elapsoidea boulengeri, and four locales of Ophiophagus hannah) to the alpha-1 nicotinic acetylcholine receptor orthosteric (active) site for amphibian, lizard, snake, bird, and rodent targets. Our results detected prey-selective, intraspecific, and geographical differences of α-neurotoxic binding. The results also suggest that crude venom that shows prey selectivity is likely driven by the proportions of prey-specific α-neurotoxins with differential selectivity within the crude venom. Our results also suggest that since the α-neurotoxic prey targeting does not always account for the full dietary breadth of a species, other toxin classes with a different pathophysiological function likely play an equally important role in prey immobilisation of the crude venom depending on the prey type envenomated. The use of this innovative and taxonomically flexible diverse assay in functional venom testing can be key in attempting to understanding the evolution and ecology of α-neurotoxic snake venoms, as well as opening up biochemical and pharmacological avenues to explore other venom effects.Few topics are able to channel the interest of researchers, the public, and industries, like nutraceuticals. The ever-increasing demand of new compounds or new sources of known active compounds, along with the need of a better knowledge about their effectiveness, mode of action, safety, etc., led to a significant effort towards the development of analytical approaches able to answer the many questions related to this topic. Therefore, the application of cutting edges approaches to this area has been observed. Among these approaches, metabolomics is a key player. Herewith, the applications of NMR-based metabolomics to nutraceutical research are discussed after a brief overview of the analytical workflow, the use of NMR-based metabolomics to the search for new compounds or new sources of known nutraceuticals are reviewed. Then, possible applications for quality control and nutraceutical optimization are suggested. Finally, the use of NMR-based metabolomics to study the impact of nutraceuticals on human metabolism is discussed.Poor diet quality and obesity, especially abdominal obesity, have been associated with systemic inflammation. The neutrophil-to-lymphocyte Ratio (NLR) is an available and inexpensive inflammation biomarker. The aim of the present study was to determine the association of dietary patterns and obesity with an inflammatory state. A group of 1747 Spanish noninstitutionalized older adults individuals were included, and a food-frequency questionnaire was applied. The Global Food Score (GFS) and Healthy Eating Index for Spanish population (SHEI) were calculated. Weight, height and waist (WC) and hip circumferences were measured, and BMI, waist-to-height ratio (WHtR), and waist-to-hip ratio (WHR) determined. In addition, body-fat percentage was measured by bioimpedance. NLR was calculated (NLR ≥ p80 2.6; 2.8 and 2.4 as inflammatory status in the entire population, men and women, respectively). The men with inflammatory status presented significative higher values of WC, WHtR, WHR, and body-fat percentage (101.82 ± 10.818 (0.688-0.974); p = 0.024 and OR = 0.829 (0.730-0.942); p = 0.004, respectively). WHtR and quality of diet is related to the inflammation status in older adults regardless to the sex.The near infrared (NIR) laser drilling of a carbon fiber reinforced polymer (CFRP) composite in the continuous wave (CW) mode and the ms pulse mode was investigated by an experiment and a numerical simulation. The relationships between the laser penetrating time, entrance hole diameter, surface heat affected zone (HAZ) width, and material ablation rate and the laser irradiation time and laser peak power densities were obtained from the experiment. For the same average power density of the laser output, 3.5 kW/cm2, it was found that the ms pulse laser mode, which had a higher peak power density, had a higher drilling efficiency. When drilling the same holes, the pulse laser mode, which had the highest peak power density of 49.8 kW/cm2, had the lowest drilling time of 0.23 s and had the smallest surface HAZ width of 0.54 mm. In addition, it was found that the laser penetrating time decreased sharply when the peak power density was higher than 23.4 kW/cm2. After analyzing the internal gas pressure by the numerical simulation, it was considered that a large internal gas pressure appeared, which resulted from polymer pyrolysis, causing a large amount of the mechanical erosion of the composite material to improve the drilling efficiency. It is important to elucidate whether genetic and epigenetic stability is altered in stored seeds and in seedlings produced from them. Non-stored and stored rye seeds at different stages of ageing were compared, as well as the seedlings obtained from them. Seeds were stored at 35 °C and 15% water content, under vacuum or air atmosphere. DNA of seeds and seedlings was isolated at three stages of the deterioration curve P75 (13 days), P20 (29 days), and P0 (36 days). Genetic stability was assessed by RAPD technique, and epigenetic changes by MSAP markers. While seeds showed genetic stability after storage, the similarity of seedlings obtained from seeds stored for 29 days was lower (95%) when compared to seedlings from control seeds. Epigenetic changes were between 15% and 30% (both de novo methylation and demethylation) in the stored seeds compared to control seeds, with no differences between 13 and 29 days of storage with either air or vacuum atmospheres. In seedlings, epigenetic changes significantly increased with storage time. In conclusion, ageing increased epigenetic instability in both seeds and seedlings, when compared to controls.The garden pea (Pisum sativum L.) is a legume crop of immense economic value. Extensive breeding has led to the emergence of numerous pea varieties, of which some are distinguished by accelerated development in various stages of ontogenesis. One such trait is rapid seed maturation, which, despite novel insights into the genetic control of seed development in legumes, remains poorly studied. This article presents an attempt to dissect mechanisms of early maturation in the pea line Sprint-2 by means of whole transcriptome RNA sequencing in two developmental stages. By using a de novo assembly approach, we have obtained a reference transcriptome of 25,756 non-redundant entries expressed in pea seeds at either 10 or 20 days after pollination. Differential expression in Sprint-2 seeds has affected 13,056 transcripts. A comparison of the two pea lines with a common maturation rate demonstrates that while at 10 days after pollination, Sprint-2 seeds show development retardation linked to intensive photosynthesis, morphogenesis, and cell division, and those at 20 days show a rapid onset of desiccation marked by the cessation of translation and cell anabolism and accumulation of dehydration-protective and -storage moieties. Further inspection of certain transcript functional categories, including the chromatin constituent, transcription regulation, protein turnover, and hormonal regulation, has revealed transcriptomic trends unique to specific stages and cultivars. Among other remarkable features, Sprint-2 demonstrated an enhanced expression of transposable element-associated open reading frames and an altered expression of major maturation regulators and DNA methyltransferase genes. To the best of our knowledge, this is the first comparative transcriptomic study in which the issue of the seed maturation rate is addressed.Owing to its pharmacological potential, betulin has attracted substantial attention in the past two decades. The present work attempts to extract betulin from Betula platyphylla Suk. bark by the ultrasonic-assisted ethanol method and to evaluate its potential bioactivities. The critical process variables affecting the yield were optimized by a four-factor, three-level, central composite response surface methodology (RSM). A betulin yield of 92.67% was achieved under the optimum conditions 65% ethanol concentration, 125 ratio of white birch bark to solvent, an extraction temperature of 30 °C, and an extraction time of 30 min. The ratio of solid to solvent is the most significant parameter in terms of yield. The optimal conditions were validated through experiments, and the observed value (92.67 ± 2.3%) was interrelated with the predicted value (92.86 ± 1.5%). The betulin extract was analyzed quantitatively by HPLC and quantitatively by LC/MS, before its potential biological activities were evaluated. Bioactivity surveys confirmed that the betulin extract showed not only no embryo deformity through zebrafish administration experiments, but also no cytotoxicity through MTT assays. Furthermore, the betulin extract had strong antioxidant activities in vitro by scavenging ferric reducing power (FRAP), 1, 1-diphenyl-2-picryl hydrazyl(DPPH), 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), and chelating metal ions. This study demonstrates that ultrasonic-assisted ethanol extraction may be a green, efficient method for the extraction of betulin from white birch bark, and that betulin extracts are potentially useful in cosmetics, food supplements, or pharmaceutical applications.
