Myocardial zinc dyshomeostasis is associated with caspase-3 activation, ErbB2 degradation and apoptosis during hypoxia/reoxygenation. Zinc pyrithione replenishes intracellular zinc, suppresses caspase-3, augments ErbB2 levels and improves cell survival. We hypothesize that zinc is capable of modulating redox and endoplasmic reticulum (ER) stress in the setting of cardiomyocyte hypoxia-reoxygenation. Hypoxia/reoxygenation lowered intracellular zinc, increased ER as well as oxidative stress in H9c2 cells, both of which were effectively attenuated by zinc supplementation. Silencing of gp91phox attenuated oxidative and ER stress, decreased caspase-3 activation and improved cell survival. Mimicking the oxidative insult using 50 μM H2O2 increased the caspase-3 activity that correlated with decreased ErbB2 levels, concomitant with augmented ER stress. N-Acetyl cysteine (NAC) administration completely suppressed ER stress as well as caspase-3 activity. Zinc depletion using TPEN also resulted in lowered ErbB2 and increased apoptosis, along with NOX2 mRNA upregulation, increased oxidative and ER stress. Repletion with zinc suppressed NOX2 mRNA, lowered oxidative as well as ER stress and decreased cell death. These results suggest that zinc dyshomeostasis, along with oxidative stress contribute to the unfolded protein response during myocardial H/R and that zinc replenishment corrects zinc homeostasis, alleviates associated stress and improves cardiomyocyte survival. Background The aim of the study was to (a) compare the accuracy of two different immobilization strategies for patients with head and neck tumors, and (b) compare the set-up errors on treatment units with different portal imaging systems. Patients and methods Variations in the position of the isocenter (IC) relative to the reference point determined on the computed tomography simulator were measured in a vertical (anterior-posterior), longitudinal (superior-inferior), and lateral (medial-lateral) direction in 120 head and neck cancer patients irradiated with curative intent. Depending on the treatment unit (unit A - 2D/2D image previews; unit B- 2D image previews) and the time of irradiation, patients were divided into 6 groups of 20 patients. In patients irradiated in 2014, standard head supports were used (groups 1 and 2), whereas in those treated in 2015 and 2017 (groups 3-6) individual head supports were employed. selleck chemical The clinical-to-planning target volume safety margin was calculated according to the formula proposed by Van Herk. Results In total, 2,454 portal images and 3,681 set-up errors were analysed. Implementation of individual head supports in 2015 resulted in a statistically significant reduction in the average inter-fraction displacement in the vertical direction and in decreased number of IC displacements in the vertical and longitudinal direction (applies to both treatment units). The largest reduction of the safety margin was calculated in the longitudinal direction and the safety margins were larger for unit B than for unit A. Conclusions The use of individual head supports and a more advanced imaging system were found to increase set-up precision. Background Ethanolic, aqueous and lipidic extracts of Spirulina platensis were evaluated for their bioactive substances (polyphenols and fatty acids) and antioxidant activities using two different assays diphenyl-1-picrylhydrazyl (DPPH) and azino-bis (ethylbenzthiazoline-6- sulfonicacid (ABTS)). Methods The phenolic compounds amounts were determined by colorimetric assays and were analyzed by high-performance and liquid chromatography (HPLC) method. The identification of phenolic compounds in the extract was accomplished by comparison of their retention times with those of pure standards. The chemical composition of fatty acids in lipidic extract was determined by GC/FIDs and GC/MS method. Results The obtained results revealed that using DPPH, the ethanolic extract recorded the highest activity with (IC50=449 μg/mL ± 83). It was characterized with a high levels of phenolic and flavonoid content 0.33 ± 0.01 mg GAE/g dw and 0.21 ± 0.01 mg quercetin/g dw, respectively. The ethanolic extract showed the presence oh antioxidant activity since they have two or more double bonds and chain lengths. The aqueous extract either with DPPH or ABTS methods, recorded low antioxidant activity. Its correlation was r=0.499, lower than found in the ethanolic extract. Conclusions These results showed that Spirulina platensis could be considered as a valuable source of fatty acids and phenolics with potent antioxidant activity. It may be employed in the manufacture of pharmaceutical drugs as an alternative source of natural antioxidants. Background Due to increasing prevalence of diabetes and associated endothelial dysfunction, this study was carried out to investigate the effects of co-administration of adrenoceptor blockers (prazosin and propranolol) and glibenclamide on plasma biomarkers of endothelial functions in diabetic rats. Methods Experiments were carried out on 35 male Wistar rats (170-200 g). They were divided into seven groups (n=5) as follows normal control, diabetic control, diabetic + glibenclamide (GLB-5mg/kg/day), diabetic+ prazosin (PRZ-0.5 mg/kg/day), diabetic + PRZ + GLB, diabetic + propranolol (PRP-10 mg/kg/day), diabetes + PRP + GLB. Experimental diabetes was induced with streptozotocin (60 mg/kg) and drugs were administered orally for 3 weeks. Blood pressure was measured and animals were sacrificed afterwards. Blood samples were collected by cardiac puncture, and major marker of endothelial functions, nitric oxide derivatives (NOx), as well as superoxide dismutase (SOD) and malondialdehyde (MDA) were measured on the plasma. The aorta was harvested for histological examination. Data were subjected to descriptive statistics and analysed using ANOVA at α 0.05. Results There was a significant increase in levels of NOx and SOD, and a decrease in MDA level in diabetic treated groups compared to diabetic control. Mean blood pressure increased in diabetic control and diabetic + GLB group when compared with normal control, while it was mildly reduced in diabetic group treated with PRZ and PRP, and co-administered GLB. More so, Aorta histology was altered in diabetic control groups when compared with normal control and all diabetic treated groups. Conclusions Results from this study suggest that PRZ, PRP, and GLB (singly and in combined therapy) could have a restorative effect on endothelial functions in diabetes. Objectives The involvement of adolescents in violent behaviors is becoming an important public health problem that is concerning physicians as it is associated with a decrease in adult life quality. In this context, bullying is a type of aggressive behavior that occurs repeatedly in interpersonal relationship where power imbalance exists increasing the risk of physical and psychosocial problems in all its intervenient. Prevalence of bullying in Portuguese adolescents is estimated to be ranging from 16.2 to 27.5%. Sleep disorders are a well-known problem related to bullying situations, either as a consequence or a risk factor; estimated prevalence in adolescence is 25% worldwide. In the present study we aimed to investigate the association between sleep quality and the involvement in school bullying in Portuguese adolescents in two high schools. Methods Data collected from a cross-sectional survey addressing questions on bullying behaviors and sleep quality was analyzed using SPSS 22.0. A two-tailed p-value ofblems in adolescence which may increase future risk for psychiatric symptoms, involvement in criminal activities, hyperactive, and inattentive behaviors. We verified a strong relation between insomnia and involvement in bullying situations with those who are involved reporting difficulties in sleep induction. The group of "bully-victims" was identified as potentially being the one with the most troubled adolescents since they combine characteristics of both bullies and victims. The authors conclude that there should be an investment in school-based interventions that help recognize and reduce school bullying and change lifestyle behaviors that may negatively influence adolescents' development.Introduction The health practices of pregnant women can affect maternal and fetal health, and pregnancy outcomes. Understanding the predictors of health practices in adolescent pregnant women can help improve these practices. The present study aimed to determine the predictors of health practices in adolescent pregnant women. link2 Materials and methods This cross-sectional study was conducted on 316 adolescent pregnant women in health centers of Tehran (capital of Iran). The participants were selected through the census method. The data were collected using the socio-demographic, obstetrics and health practices questionnaires and analyzed using Pearson correlation coefficient, independent t-test, one-way ANOVA, and general linear model. Results The mean score of health practice was 135.29 ± 9.08, ranging from 34 to 170. link3 The highest and the lowest mean score pertained to the avoidance of harmful drugs and opiates subscale (29.79) and the balance of rest and exercise subscale (13.15), respectively. The general linear model showed that a lower level of education, a history of abortion, lack of involvement in pre-pregnancy counseling, and delayed attendance for prenatal care, reduced the health practice score. However, high level of emotional support from the spouse increased the health practice score. Conclusions The findings highlighted the factors effective on health practices including involvement in pre-pregnancy counseling, early prenatal care and emotional support for the adolescent pregnant woman. Objectives It is widely accepted that sexual and reproductive health in adolescents are among the health priorities in the world that affects health aspects and the formation of beliefs and socioeconomic development in the community. However, achieving goals related to sexual and reproductive health is not possible without awareness and participation of the adolescents. On the other hand, proper knowledge about such issues can prevent many problems in this regard. Methods This qualitative study was carried out for one month in schools of Alborz University of Medical Sciences using content analysis. Data were collected from semi-structured, individual and in-depth interviews with female students. In addition, data analysis was performed using guided qualitative content analysis (theory-based). Results Analysis of the data led to the extraction of 11 primary codes classified in six categories of protection motivation constructs (perceived sensitivity, perceived severity, response efficiency, response cost, self-efficacy, and internal rewards) and two additional categories (communications and social contexts). Conclusion All interviewees found themselves exposed to sexually transmitted diseases and emphasized sexual self-care even though the majority of these individuals had inadequate self-efficacy.Objectives To determine the prevalence of muscle-enhancing behaviors in adolescents and young adults using a nationally representative sample in the USA and to examine differences by sex, race/ethnicity, age, socioeconomic status, body mass index, and participation in team sports. Methods Prospective cohort data from the National Longitudinal Study of Adolescent to Adult Health, Waves I through III (1994-2002) were analyzed. Engagement in muscle-enhancing behaviors including dietary changes, exercise and weightlifting, supplement use, performance-enhancing substances, and anabolic androgenic steroids were recorded. Multiple logistic regression models using generalized estimating equations, incorporating robust standard errors with clustering by school and within persons, and using national sample weighting, were used to determine associations with muscle-enhancing behaviors across three data collection waves. Results Of the 18,924 adolescents at baseline, 29.2% of males and 7.0% of females reported weight gain attempts, while 25.
