A new procedure for classifying brain structures described by SPHARM is presented. We combine a dimension reduction technique (functional principal component analysis or functional independent component analysis) with stepwise variable selection for linear discriminant classification. This procedure is compared with many well-known methods in a novel classification problem in neuroeducation, where the reversal error (a common error in mathematical problem solving) is analyzed by using the left and right putamens of 33 participants. The comparison shows that our proposal not only provides outstanding performance in terms of predictive power, but it is also valuable in terms of interpretation, since it yields a linear discriminant function for 3D structures.Gastrointestinal function is known to be regulated by steroid molecules produced by the gonads, the adrenal glands and the gut microbiota. However, we have a limited knowledge on the functional significance of local steroid production by gastrointestinal tract tissue. In this study, we demonstrate the application of AcmB in the synthesis of 1-dehydro-3-ketosteroids and investigate the influence of reaction conditions on the catalytic performance of the enzyme. The recombinant AcmB expressed in E. coli BL21(DE3)Magic exhibits a broad pH optimum and pH stability in the range of 6.5 to 9.0. The activity-based pH optimum of AcmB reaction depends on the type of electron acceptor (2,6-dichloroindophenol - DCPIP, phenazine methosulfate - PMS or potassium hexacyanoferrate - K3[Fe(CN)6]) used in the biocatalytic process yielding the best kinetic properties for the reaction with a DCPIP/PMS mixture (kcat/Km = 1.4·105 s-1·M-1 at pH 9.0) followed by DCPIP (kcat/Km = 1.0·105 s-1·M-1 at pH = 6.5) and K3[Fe(CN)6] (kcat/Km = 0.5·102 s-1·M-1 at pH = 8.0). The unique feature of AcmB is its capability to convert both testosterone derivatives (C20-C22) as well as steroids substituted at C17 (C27-C30) such as cholest-4-en-3-one or (25R)-spirost-4-en-3-one (diosgenone). Apparent steady-state kinetic parameters were determined for both groups of AcmB substrates. In a batch reactor synthesis, the solubility of water-insoluble steroids was facilitated by the addition of a solubilizer, 2-hydroxypropyl-β-cyclodextrin, and organic co-solvent, 2-methoxyethanol. The aim of this study was to evaluate the in vitro anticancer properties of Hitchner B1 (HB1) strain of NDV on TC-1 cell line and underlying molecular mechanisms. The cytolytic effects of oncolytic HB1 strain of NDV was determined by lactate dehydrogenase (LDH) release assay. Apoptosis, intracellular reactive oxygen species (ROS) levels, cleaved caspase-3 and autophagy were evaluated by flow cytometry. Cytochrome-C and survivin protein levels were distinguished by Enzyme-Linked Immunosorbent Assay (ELISA). Our results from LDH method showed that the viability of the TC-1 cell line following HB1 NDV infection was dose-dependent and decreased significantly with increasing the dose of HB1 NDV infection (MOIs 5, 10, and 15). Other evaluations also revealed that HB1 strain of NDV potentially led to the ROS production, and apoptosis and autophagy induction in TC-1 cell line in a dose-dependent manner. The in vitro experiments also presented that NDV treatment significantly up-regulated the expression of cytochrome-C and down-regulated the expression of survivin, as detected by ELISA assay. Our results confirmed that the HB1 NDV could be introduced as a powerful candidate for the therapy of cervical cancer. However, further examinations are needed to explain the underlying mechanisms of the HB1 NDV against TC-1 cell line and cervical cancer.Vibrio cholerae is a natural inhabitant of aquatic environments and causes the epidemic diarrheal disease known as cholera. Fatty acid metabolism is closely related to the pathogenicity of V. cholerae. The TetR family transcriptional repressor PsrA regulates the β-oxidation pathway in Pseudomonas aeruginosa; however, little is known about its regulation in V. cholerae. In this study, qRT-PCR revealed that the expression of vc1741 (psrA) increased 40-fold in the small intestines of infant mice compared with that grown in LB medium. The Δvc1741 mutant showed a significant defected in the ability to colonize the small intestines of infant mice with a competitive index (CI) of 0.53. EMSAs indicated that VC1741 could directly bind to the promoter regions of vc1741-fadE1, fadBA, and fadIJ operons, and these bindings were reversed upon addition of the long-chain fatty acid (LCFA), oleic acid. The expression levels of the fadB, fadA, fadI, and fadJ genes were all elevated by approximately 2-fold in the Δvc1741 mutant strain compared with that in the wild-type strain in LB medium, indicating that VC1741 is a repressor for these genes involved in fatty acid degradation.
