The expression of long non-coding RNA (lncRNA) prostate cancer-associated ncRNA transcripts 1 (PCAT1) is increased in non-small cell lung cancer (NSCLC). It stimulates tumour growth and metastasis, but its role in the radioimmune responses remain unknown. We aimed to explore the impacts of PCAT1 on tumorigenesis and radioimmune responses and the underlying molecular mechanisms in NSCLC.

Given the current popularity of magnetic resonance-guided FUS (MRgFUS), we have investigated the MR relaxation times T1 and T2 of different mixtures of agar-based phantoms. Nine TMPs were constructed containing agar as the gelling agent and various concentrations of silicon dioxide and evaporated milk. An agar-based phantom doped with wood powder was also evaluated. A series of MR images were acquired in a 1.5 T scanner for T1 and T2 mapping. T2 was predominantly affected by varying agar concentrations. A trend toward decreasing T1 with an increasing concentration of evaporated milk was observed. The addition of silicon dioxide decreased both relaxation times of pure agar gels. The proposed phantoms have great potential for use with the continuously emerging MRgFUS technology. The MR relaxation times of several body tissues can be mimicked by adjusting the concentration of ingredients, thus enabling more accurate and realistic MRgFUS studies.

To develop an automated image recognition software for the objective quantification of choroidal vascularity index (CVI) and choroidal thickness (CT) at different choroidal locations on images obtained from enhanced depth imaging optical coherence tomography (EDI-OCT), and to validate its reliability and investigate the difference and correlation between measurements made by manual and software.

Dacomitinib is the second-generation epidermal growth factor receptor (EGFR)-tyrosine kinase inhibitor (TKI) for mutant non-small cell lung cancer (NSCLC). This study evaluated clinical outcomes of dacomitinib in rechallenge setting.

Patients who received dacomitinib for advanced EGFR-mutant NSCLC who had progressed after EGFR-TKI in nine institutions in Japan were included in the analyses.

In total, 43 patients were analyzed. The median progression-free survival (PFS) was 4.3months (95% confidence interval [CI], 2.5-5.6). The overall survival (OS) was 10.5months (95% CI, 7.4-not reached). The overall response rate was 25.5% (95% CI, 13.1-33.7). Subset analysis indicated that patients with EGFR exon 21 L858R showed longer PFS than those with EGFR exon 19 deletion (5.8 vs. 4.1months) (p=0.018). Multiple factors may influence the risk of being homebound, including social isolation and race. This study examines the relationship between social isolation and homebound status by race over 9 years in a sample of adults.

Utilizing a representative sample of 7788 Medicare beneficiaries aged 65+ from 2011-2019, we assessed the odds of becoming homebound by social isolation. We defined social isolation as the objective lack of contact with others. We defined severe social isolation as scoring a 0 or 1 on a social connection scale from 0 to 4. Homebound status was defined as never leaving home or only leaving home with difficulty. Utilizing a multivariate Cox proportional hazards model adjusting for age, gender, marital status, income, and education, we examined the association between social isolation at baseline and becoming homebound during the study in those who were not initially homebound.

Older adults in this study were on average 78 years old. Overall, most were white (69%), female (56.3%), and msearch is needed to determine whether community-based strategies and policies that identify and address social isolation reduce homebound status among community-dwelling older adults.

Berberine is a plant-derived herbicidal alkaloid. The herbicidal mechanism of berberine is still not clear. In this study, our aim is to clarify the mechanism of berberine inhibiting the root growth of Arabidopsis thaliana, aiming at providing new insight into identifying the molecular targets of berberine.

The whole-genome RNA sequencing had revealed that 403 genes were down-regulated, and 422 genes were up-regulated in Arabidopsis roots with berberine treatment. According to KEGG and GO analysis, the expression of two genes AT5G48010 (Thas) and AT5G42600 (MRN1) which are in the sesquiterpenoid and triterpenoid biosynthesis pathway were affected most. These two genes belong to thalianol and marneral gene clusters. RT-PCR showed that Arabidopsis responds to berberine by inhibiting root growth through repressing the expression of thalianol and marneral gene clusters, which was independent of the upstream effectors ARP6 and HTA9-1. GC-MS analysis showed that berberine could inhibit THAH in the biosynthetic network of triterpenoid gene cluster in Arabidopsis and thus cause the accumulation of thalianol.

Our study indicated the repression of the thalianol and marneral gene clusters as the primary mechanism of action of berberine in Arabidopsis, which may result in plant growth defects by interrupting the thalianol metabolic pathway.
