To date, drugs to attenuate cytokine storm in severe cases of Corona Virus Disease 2019 (COVID-19) are not available. In this study, we investigated the effects of intragastric and atomized administration of canagliflozin (CAN) on cytokine storm in lung tissues of lipopolysaccharides (LPS)-induced mice. Results showed that intragastric administration of CAN significantly and widely inhibited the production of inflammatory cytokines in lung tissues of LPS-induced sepsis mice. Simultaneously, intragastric administration of CAN significantly improved inflammatory pathological changes of lung tissues. Atomized administration of CAN also exhibited similar effects in LPS-induced sepsis mice. Furthermore, CAN significantly inhibited hypoxia inducible factor 1α (HIF-1α) and phosphofructokinase-2/fructose-2,6-bisphosphatase 3 (PFKFB3) protein levels in LPS-treated lung tissues. Its action mechanism might involve the regulation of HIF-1α and glycolysis in vivo. Autoimmune thyroid disease (AITD) is a common autoimmune disease. This review introduces the history and origins of Bregs. It summarizes the different phenotypes and functionalities of Breg cells related to AITD and analyzes the reasons for the differences in Breg expression frequencies in Graves disease (GD) and Hashimoto's Thyroiditis (HT). A number of functional defects of regulatory B cells may be the newly discovered cause of AITD. This paper sheds new light on the role and prospects of Bregs in the progression and treatment of AITD.The interplay between the immune system, sleep dysfunction and cognitive impairment participates in the progression of disability in multiple sclerosis (MS). Our aim was to identify molecular pathways and B cell associated with separate components of MS disability. Benign MS, non-benign MS patients and healthy controls were recruited. Patients underwent polysomnography and cognitive studies. Microarray and bioinformatics analysis performed using peripheral blood mononuclear cell samples identified B cell-associated genes with the most significantly altered expression. Expression levels of these genes were validated by real-time PCR and peripheral blood cell subsets were examined by flow cytometry. Putative correlations among clinical and laboratory parameters were investigated by correlation network analysis. Sleep and cognitive functions were equally impaired in BMS and NBMS. BMS patients showed significantly reduced memory B cell and increased regulatory B cell percentages than NBMS patients. Among genes that were selected by bioinformatics, levels of BLK, BLNK, BANK1, FCRL2, TGFB1 and KCNS3 genes were significantly different among study subgroups. Correlation network analysis showed associations among physical-cognitive disability and sleep dysfunction measures of MS versus expression levels of selected genes. BMS and NBMS differ by physical disability but not cognitive and sleep dysfunction. Different components of disability in MS are associated with peripheral blood B cell ratios and B cell related gene expression levels. Thus, it is likely that altered B cell functions participate in the progression of disability in MS.
Relapsing MS (RMS) is a lifelong disease without a cure, usually diagnosed between 20-40 years of age. Being newly diagnosed with RMS is a highly stressful event due to the unpredictable disease course after diagnosis. Thus, it is imperative that persons with MS have the skills and support to cope with the negative physical and emotional effects of the disease. The objective of this study was to assess whether a mindfulness-based intervention (MBI) would improve coping skills and thus lessen the negative consequences of stress due to being newly diagnosed with RMS.
This was a single-blind (assessor), randomized, prospective study of a 10-week MBI vs. usual standard of care in persons newly diagnosed (within 1 year) with RMS, recruited from one tertiary care MS clinic in London (ON), Canada. The MBI was administered in group format with a trained MBI facilitator. Primary outcomesincluded the Brief COPE measure and the Hospital Anxiety and Depression Scale (HADS) subscales. Secondary outcomes included measurceived stress in newly diagnosed (within one year) persons with RMS in the short term. Future research to confirm these results, as well as further investigate measures to extend the benefit beyond the immediate intervention.
The purpose of the current study was improving the measurement precision of the Transition Readiness Assessment Questionnaire (TRAQ 5.0-20 item) in order to gain better decimation of transition readiness skills across the 5 Stages of Change-from Precontemplation to Mastery.
In stage 1, starting with the TRAQ 5.0 20-item, 5 domain subscale questionnaire, we eliminated the five lowest discriminating items using Item response theory (IRT) in MPlus v7.4,which eliminated the domain subscale Managing Daily Activities, and we e added 15 more difficult and better discriminating items. We added items to both to the remaining 4 domain subscales and created a new domain subscale entitled Future Planning. The revised 30-item TRAQ was piloted among 386 youth between 16 and 24 years old (mean = 20 years; 54% female; 87% White).
After examining the model fit, discrimination and difficulty coefficients, and modification indices, we eliminated 10 items and the new Future Planning domain subscale we eliminated. By adding more difficult items to the 4 resulting domain subscales, we have demonstrated improved item discrimination and difficulty, and therefore can better measure acquisition of transition readiness skills across the five stages of change from pre-contemplation to contemplation to initiation to action and finally to mastery.EG (ethylene glycol) is a good model system for the study of the fundamental hydrogen bonds in aqueous solutions. Using Raman spectroscopy, we have investigated the EG volume fraction induced variation in the hydrogen bonding interactions and conformations of EG-H2O (water) binary solutions. New hydrogen bonding networks is evidenced by the appearance of remarkable changes in Raman spectra and the full width at half maximum (FWHM) when the mixing volume ratio is 0.5. The H-bond in water molecules firstly strengthened and then weakened with the increasing concentration of EG. Meanwhile, the dominant association structure also changed from H2O-H2O to EG-H2O in binary solutions in this process. We provide a simple but effective method for studying EG-H2O binary solutions. It also has exciting potential prospects and can be easily extended to other mixing situations.In this paper, a reagent-free simultaneous and direct detection method of three analytes in human blood based on Fourier-transform Raman (FT-Raman) spectroscopy with 1064 nm laser radiation was proposed for the first time. A total of 161 human blood samples were characterized by FT-Raman spectroscopy under the excitation laser source of 1064 nm. In order to achieve a robust regression model, the Nonlinear Iterative Partial Least Squares (NIPALS) with orthogonal signal correction (OSC) algorithm and sample set partition based on a joint x-y distance (SPXY) is used to establish multivariate calibration models. The root means square error of cross-validation (RMSECV), root mean square error of prediction (RMSEP), correlation coefficients (R2) and ratio of performance to deviation (RPD) were 0.34255 mg/dL, 0.3662 mg/dL, 0.99982 and 56.3524 for glucose, 0.33656 mg/dL, 0.75736 mg/dL, 0.99967 and 34.9169 for total cholesterol (TC), and 0.29956 mg/dL, 0.27469 mg/dL, 0.99998 and 173.5098 for triglycerides (TG), respectively. The analysis results showed that the proposed method could be able to accurately predict the concentration of glucose, TC and TG in blood. This method can instantaneous multi-component detection on whole blood.Based on the specific interaction of Ag+ and cytosine-cytosine (C-C) base mismatch and using berberine (Ber) as the fluorescent probe and Exonuclease I (Exo I) as the background fluorescence reducing tool, a label-free Exo I-assisted fluorescence aptamer sensing platform was established for the detection of silver ions with high sensitivity and selectivity. Exo I reduced the fluorescence background of the Ber/Ag+-aptamer complex to a level similar to that of Ber itself in the absence of Ag+. After introducing Ag+ into the sensing system, it induces the aptamer rich in base C to form C-Ag+-C i-motif structure which are resistant to degradation mediated by Exo I. The concentration of Ber, Ag+-aptamer, Exo I and the temperature and reaction time for Exo I were all optimized. Under the optimal experimental conditions, the detection limit of Ag+ was 4.4 nM and the linear range was from 0.0059 μM to 235.48 μM with a coefficient of determination (R2) > 0.99. Moreover, the proposed strategy had been successfully applied to the detection of Ag+ in tap water and human serum with a good recovery ranging from 88.4% to 106.9%.Rare earth-carbon dots (RE-CDs) hybrid nanomaterials with the merits of both RE and CDs have rapidly emerging as highly promising functional materials in biochemical analysis. In this work, a new kind of water-soluble RE-CDs nanocomposite (CDs@CaF2Eu3+) was developed for the ratiometric determination of doxycycline (DOX). The CDs@CaF2Eu3+ under the excitation at 365 nm displayed blue emission of CDs at 440 nm and no obvious emission of Eu3+. With the addition of DOX, substantial fluorescence quenching of the CDs at 440 nm and enhancement of Eu3+ at 613 nm were observed, resulting in a ratiometric fluorescent response toward DOX. A wide linear range from 0.1 µM to 30 µM was achieved in the detection of DOX with a lowest detection limit of 43 nM. In particular, the probe could discriminate DOX from other tetracycline antibiotics through unique fluorescence response. Moreover, we have successfully applied the method for the determination of DOX in milk and honey samples.The reaction system between the simplest Criegee intermediate, CH2OO, and the greenhouse gas CO2 has been investigated by Fourier transform microwave spectroscopy. The CH2OO-CO2 weakly bound complex was identified in the rotational spectrum, where inversion doublets due to the tunnelling motion between two equivalent configurations of the complex, with CO2 located at one side or the other side of the CH2OO plane, were observed. Using a two-state torsion-rotation Hamiltonian, a complete set of rotational and centrifugal distortion constants for both tunneling states were derived. In addition, the torsional energy difference between both states could be accurately determined, being 23.9687 MHz. The non-observation of the cycloaddition reaction product is in agreement with our ab initio calculations and with previous results that concluded that the reactivity of CIs toward CO2 is measured to be quite limited.
