A series of methoxy naphthyl substituted cyclopenta[d]pyrimidine compounds, 4-10, were designed and synthesized to study the influence of the 3-D conformation on microtubule depolymerizing and antiproliferative activities. NOESY studies with the N,2-dimethyl-N-(6'-methoxynaphthyl-1'-amino)-cyclopenta[d]pyrimidin-4-amine (4) showed hindered rotation of the naphthyl ring around the cyclopenta[d]pyrimidine scaffold. In contrast, NOESY studies with N,2-dimethyl-N-(5'-methoxynaphthyl-2'-amino)-cyclopenta[d]pyrimidin-4-amine (5) showed free rotation of the naphthyl ring around the cyclopenta[d]pyrimidine scaffold. The rotational flexibility and conformational dissimilarity between 4 and 5 led to a significant difference in biological activities. Compound 4 is inactive while 5 is the most potent in this series with potent microtubule depolymerizing effects and low nanomolar IC50 values in vitro against a variety of cancer cell lines. The ability of 5 to inhibit tumor growth in vivo was investigated in a U251 glioma xenograft model. The results show that 5 had better antitumor effects than the positive control temozolomide and have identified 5 as a potential preclinical candidate for further studies. The influence of conformation on the microtubule depolymerizing and antitumor activity forms the basis for the development of conformation-activity relationships for the cyclopenta[d]pyrimidine class of microtubule targeting agents.Due to the increasing number of ecosystem invasions with the introduction of exogenous species via ballast water, the International Maritime Organization adopted the Ballast Water Convention (BWMC). The BWMC establishes standards for the concentration of viable organisms in a ballast water discharge. Ultraviolet (UV) irradiation is commonly used for treating ballast water; however, regrowth after UV irradiation and other drawbacks have been reported. In this study, improvement in UV treatment with the addition of hydrogen peroxide or peroxymonosulfate salt was investigated using the microalgae Tetraselmis suecica as the target organism. Results reported that each of these reagents added in a concentration of 10 ppm reduced the concentration of initial cells by more than 96%, increased the UV inactivation rate, and enabled reaching greater level of inactivation with the treatment. These improvements imply a reduction of the UV doses required for a consistent compliance with the BWMC standards.Microbeads are solid primary microplastics less then 5 mm in diameter that are added to cosmetic products for cleansing and/or exfoliation of the skin. After use, they are discharged into the drain and end up in Wastewater Treatment Plants (WWTPs), from which they can escape into waters. Once disposed, there is no efficient method of recovery and the environmental conditions do not allow full biodegradation. Focusing mainly on the possible effect of microbeads on the environment and the human health, and taking into account that there have always been alternatives with similar cost, scientists suggested banning microbeads from cosmetic products. Despite the increasing knowledge on the microplastics' effects, the pressure from non-governmental organizations (NGO's) and the increasing public concern, few European and other countries worldwide have taken legislative steps against microbeads, and even fewer against other microplastics used in cosmetic products, mainly because it is a common belief that cosmetic industries responded massively to the call for a phase-out. Although EU acted soon after scientists focused on microbeads' effects by restricting cosmetics with microbeads to bare the EU Ecolabel in 2014, there is still no European-wide ban, which will probably take place as of 2022. Present study offers a thorough literature review on the presence of microbeads in cosmetics up to date, focusing primarily on the actions against their use, and questioning whether future pollution from microbeads or/and microplastics in cosmetics has been successfully handled.Humans rely fundamentally on the marine environment, which is at the same time subject to an increasingly broad range of anthropogenic pressures, leading to growing concerns and the need for effective management for marine protection. One activity is the dredging of ports and harbours which is necessary to maintain safe channels for commercial and recreational navigation. Regulatory authorities in developing countries have few resources to conduct full EIAs for determining dredged material disposal sites but are required to do so under international obligations. The Tool in this paper provides an effective, pragmatic, transparent, consistent, and robust approach to protect the environment whilst using limited technical and scientific resources through a risk based approach to defining need, characterising and designating disposal sites at sea for dredged material. Whilst this approach for dredged material disposal sites was developed for use in UK, this process is equally applicable to other waste types, worldwide.Right Brain-Damaged patients (RBD) with left spatial neglect (N+), are characterised by deficits in orienting and re-orienting attention to stimuli in the contralesional left side of space. In a recent ERPs study with visual stimuli (Lasaponara et al., 2018) we have pointed out that the pathological attentional bias of N+ is matched with exaggerated novelty reaction and contextual updating of targets in the right ipsilesional space and reduced novelty reaction and contextual updating of targets in the left contralesional space. To characterise further the attentional performance of N+, here we measured Pupil Dilation (PDil), which is a reliable marker of noradrenergic-locus coeruleus activity and response to unexpected events/rewards. Compared to Neutral and Valid targets, N+ patients displayed a pathological reduction of PDil in response to infrequent Invalid targets in the left side of space, while in Healthy Controls (HC) and RBD without neglect (N-) the same targets enhanced PDil with respect to Neutral and frequent Valid targets. Invalid targets in the right side of space enhanced PDil in all experimental groups. Interestingly, both N- and N+ showed a consistent number of target omissions both in the left and right side of space. With respect to seen targets, N- showed reduced PDil in response to unseen targets both in the left and right side of space. In contrast, N+ had reduced PDil in response to unseen targets in the left side of space though not in the right side, where seen and unseen targets evoked comparable levels of PDil. These results disclose, for the first time, the PDil correlates of spatial attention in left spatial neglect and suggest that the pathological attentional bias suffered by N+ might enhance the autonomic responses reflected in PDil to unseen ipsilesional stimuli. click here This enhancement can contribute to biasing contextual updating and predictive coding of stimuli in the ipsilesional space, thus worsening the pathological attentional bias of N+.
