HSP70 triggered the production of inflammatory mediators in a TLR4-dependent manner. Moreover, HSP70 inhibition in vivo ameliorated diabetes-induced albuminuria, inflammatory response and tubular injury. Finally, we found that individuals with DN had higher levels of TLR4 and HSP70 in the dilated tubules than non-diabetic controls. Thus, activation of the HSP70-TLR4 axis, stimulated at least in part by albumin, in the tubular cell is a newly identified mechanism associated with induction of tubulointerstitial inflammation and aggravation of pre-existing microalbuminuria in the progression of DN.We have developed a method to label and image myeloid cells infiltrating the mouse retina and choroid in vivo, using a single depot injection of indocyanine green dye (ICG). This was demonstrated using the following ocular models of inflammation and angiogenesis endotoxin-induced uveitis, experimental autoimmune uveoretinitis and laser-induced choroidal neovascularization model. BTK inhibitor A near-infrared scanning ophthalmoscope was used for in vivo imaging of the eye, and flow cytometry was used on blood and spleen to assess the number and phenotype of labelled cells. ICG was administered 72 h before the induction of inflammation to ensure clearance from the systemic circulation. We found that in vivo intravenous administration failed to label any leukocytes, whereas depot injection, either intraperitoneal or subcutaneous, was successful in labelling leukocytes infiltrating into the retina. Progression of inflammation in the retina could be traced over a period of 14 days following a single depot injection of ICG. Additionally, bright-field microscopy, spectrophotometry and flow cytometric analysis suggest that the predominant population of cells stained by ICG are circulating myeloid cells. The translation of this approach into clinical practice would enable visualization of immune cells in situ. This will not only provide a greater understanding of pathogenesis, monitoring and assessment of therapy in many human ocular diseases but might also open the ability to image immunity live for neurodegenerative disorders, cardiovascular disease and systemic immune-mediated disorders.To investigate the initial symptoms, treatment, prognosis, and 1-, 3-, and 5-year survival of patients with bronchopulmonary carcinoid, clinical and pathological data were collected retrospectively from 74 patients diagnosed with bronchopulmonary neuroendocrine tumors at the Cancer Hospital, Chinese Academy of Medical Science, from January 2004 through December 2009. The data collected included age, initial symptoms, primary tumor sites, pathological types, lymphatic metastasis, and distant metastasis. The Kaplan-Meier method was used for survival analysis and the log-rank test was used for univariate analysis of prognostic factors. A Cox proportional hazard regression model was used for multivariate analysis. The 74 patients included 56 men and 19 women, and their average age was 56.07 years. The most common initial symptom was cough (51.35%), and the major lesion site was the left upper lobe of the lung (38.84%). Of the 59 patients (79.73%) who underwent surgery, most (76.27%) received a pulmonary lobectomy. The patients' 1-, 3-, and 5-year survival rates were 92.7, 80.3, and 71.9%, respectively. Univariate analysis showed that both local lymphatic and distant metastases were prognostic factors (P less then 0.05), whereas multivariate analysis showed that the pathological type (typical carcinoid and atypical carcinoid) was an independent prognostic factor (P=0.006). These data indicate that cough is the major presenting symptom of patients with bronchopulmonary carcinoid and the left upper lobe of the lung is the most commonly involved site. Following treatment, mostly by pulmonary lobectomy, the 5-year survival rate is 71.9%. The pathological tumor type is an independent prognostic factor.Bufalin, a cardiotonic steroid isolated from toad venom, has been shown to kill various types of tumor cells. Our previous study showed that triple-negative breast cancer (TNBC) cells were less sensitive to bufalin than other types of breast cancer cells, but the reason for this lower sensitivity remains unclear. In this study, we showed that bufalin induced apoptosis in MDA-MB-231 and MCF-7/ADR TNBC cell lines, accompanied by increased miR-155-5p expression. Overexpression of miR-155-5p promoted proliferation and reduced bufalin-induced apoptosis of TNBC cells. In contrast, downregulation of miR-155-5p increased sensitivity to bufalin and upregulated the expression of FOXO3A. Bufalin also downregulated DNA methyltransferases 1 and 3a (DNMT1 and DNMT3a), and concurrent inhibition of DNMT1 and DNMT3a significantly increased miR-155-5p expression. These results indicate that miR-155-5p antagonizes bufalin sensitivity in TNBC cells, and that downregulation of DNMT1 and DNMT3a may be responsible for the bufalin-induced upregulation of miR-155-5p.Bisphenol A (BPA) is widespread in the aquatic environment, and early life-stage exposure to this chemical affects growth and development in fish. We tested the hypothesis that BPA accumulation in eggs, mimicking maternal transfer, impacts the development and function of the hypothalamus-pituitary-interrenal (HPI) axis in rainbow trout (Oncorhynchus mykiss). To mimic maternal transfer, oocytes were loaded with 0 (control), 1, 4 and 40ng BPA embryo(-1). We measured the temporal changes in whole body cortisol content, transcript levels of corticosteroidogenesis-related genes and glucocorticoid receptor (GR) and mineralocorticoid receptor (MR) protein expressions during early development. In addition, trout larvae were subjected to an acute stressor at key developmental stages (at hatch, just prior to first feed and post hatch) and whole body cortisol levels measured to assess the functional integrity of the HPI axis. The developmental profile of whole body cortisol content was altered in the 1 and 40ng BPA groups compared to the control group. Also, the two key rate-limiting steps in steroidogenesis, the steroidogenic acute regulatory protein (StAR) and cytochrome P450 side chain cleavage (P450scc) enzyme transcript levels were disrupted in the larvae originating from BPA accumulated eggs prior to first feed. Whole body GR and MR protein expressions were higher in the 40ng BPA group compared to other groups prior to first feed. Stressor-induced elevation in whole body cortisol levels were evident at 65 days post-fertilization (dpf) and later, but not at 42 dpf in trout larvae. This cortisol response to a stressor exposure was attenuated in the 4 and 40ng BPA groups at 65 dpf, but not at 140 dpf. Together, accumulation of BPA in eggs, mimicking maternal transfer of this chemical, disrupts the ontogeny of cortisol stress response in trout larvae and may compromise the target tissue responsiveness to cortisol stimulation during early development.Despite the focus of most ecotoxicological studies on cyanobacteria on a select group of cyanotoxins, especially microcystins, a growing body of evidence points to the involvement of other cyanobacterial metabolites in deleterious health effects. In the present study, original, self-developed reporter gene-based cellular biosensors, detecting activation of the main human xenobiotic stress response pathways, PXR and NFkappaB, were applied to detect novel potentially toxic bioactivities in extracts from freshwater microcystin-producing cyanobacterial blooms. Crude and purified extracts from cyanobacteria containing varying levels of microcystins, and standard microcystin-LR were tested. Two cellular biosensor types applied in this study, called NHRTOX (detecting PXR activation) and OXIBIOS (detecting NFkappaB activation), successfully detected potentially toxic or immunomodulating bioactivities in cyanobacterial extracts. The level of biosensor activation was comparable to control cognate environmental toxins. Despite the fact that extracts were derived from microcystin-producing cyanobacterial blooms and contained active microcystins, biosensor-detected bioactivities were shown to be unrelated to microcystin levels. Experimental results suggest the involvement of environmental toxins (causing a response in NHRTOX) and lipopolysaccharides (LPS) or other cell wall components (causing a response in OXIBIOS) in the potentially harmful bioactivity of investigated extracts. These results demonstrate the need for further identification of cyanobacterial metabolites other than commonly studied cyanotoxins as sources of health risk, show the usefulness of cellular biosensors for this purpose and suggest a novel, more holistic approach to environmental monitoring.The π Echo Planar Imaging (PEPI) method was modified to compensate for concomitant magnetic fields by waveform symmetrization. Samples with very short T2(∗) (a few hundred microseconds) and short T2 (tens of milliseconds to hundreds of milliseconds) were investigated. Echo spacings as short as 1.2 ms were achieved with the gradient pre-equalization method, enabling rapid 3D imaging of short relaxation time species with sub-millimeter resolution. The PEPI method yields superior quality images, compared to the Fast Spin Echo (FSE) method, with significantly reduced gradient duty cycle. Accelerated PEPI measurements with a variable number of centric interleaves are presented. Restricted k-space sampling was demonstrated for specific sample geometries, notably a Locharbriggs sandstone core plug, with the acquisition time further reduced. These methods generate proton density weighted images considering the echo time to sample T2 ratio. These methods are principally designed for 3D studies of fluid saturation in rock core plugs, evolving in time due to some manner of external perturbation, such as water flooding.In this study we have focused on 3,5,6-trichloro-2-pyridinol (TCP), degradation product of chlorpyrifos. Photolysis experiments were conducted in order to elucidate its degradation mechanism. Identification of products was performed using the LC-MS technique. To evaluate the mineralization efficiency, TiO2 photocatalytic study was performed. Under photolytic experimental conditions, the concentration of TCP after 120 min of irradiation reached 5.9 ± 1.5% of the initial concentration, while chloride concentration reached approximately 73% of total chloride concentration. The TOC measurements after 120 min of photocatalytic degradation experiment revealed high mineralization rate, i.e. 53.6 ± 1.9%, while chloride concentration reached 26.6 mg L(-1) what means almost quantitative transformation of organic chlorine into chloride. TIC chromatogram (ESI, negative ion mode) of the reaction mixture after 30 min of irradiation revealed the presence of several peaks. One of them has already been reported previously. Two other products have been identified in this study for the first time. They have been formed by radical attack of the reactive OH(•) species on the carbonyl group followed by the corresponding N-C or C-C bond cleavages and recyclization with formation of the pyrrol structures substituted with carboxylic groups. Both deprotonated molecules easily lose CO2 in ESI conditions.
