The optimal workflow was successfully applied to multidrug-resistant Salmonella Kentucky isolates, where the transfer of an ESBL-gene-containing fragment from a plasmid to the chromosome was detected. This study highlights a strategy including wet and dry lab parameters that allows accurate plasmid reconstruction, which will contribute to an improved monitoring of circulating plasmids and the assessment of their risk of transfer.

The aim of the study was to investigate perceptions of staff about the promotion of physical activity (PA) in selected group residences of Hong Kong (HK), some of which had experienced a multi-component PA program.

Focus group interviews with nineteen staff members from four group homes (two of which received the program) were conducted.

A SWOT analysis provided important insights into residential staff views about key influences on the quality of PA programs for residents with intellectual disabilities (ID). Positive (strengths and opportunities) and negative (weaknesses and threats) influences were identified. They were associated with characteristics of residents, staff, and group residence. Increasing age and low motivation are impediments to PA engagement of adults with ID. Staff competence and prior unsuccessful experience in promoting PA are also implicated.

The PA program quality is mediated by the quality of staff interpersonal interactions with their clients and their commitment in encouraging such adults with ID to join and persistent in PA as well as staff seeking external resources and support as well as using initiative to adapt PA promotion activities in their specific group residential context.

Herein, we report a novel class of RNA ligase deoxyribozymes that utilize 5'-adenylated RNA (5'-AppRNA) as the donor substrate, mimicking the activated intermediates of protein-catalyzed RNA ligation. Four new DNAzymes were identified by in vitro selection from an N40 random DNA library and were shown to catalyze the intermolecular linear RNA-RNA ligation via the formation of a native 3'-5'-phosphodiester linkage. The catalytic activity is distinct from previously described RNA-ligating deoxyribozymes. Kinetic analyses revealed the optimal incubation conditions for high ligation yields and demonstrated a broad RNA substrate scope. Together with the smooth synthetic accessibility of 5'-adenylated RNAs, the new DNA enzymes are promising tools for the protein-free synthesis of long RNAs, for example containing precious modified nucleotides or fluorescent labels for biochemical and biophysical investigations. To explore the association between the personal social capital and loneliness among the widowed older adults in China. Data from 1497 widowed older adults were extracted from China's Health-Related Quality of Life Survey for Older Adults 2018. The Chinese version of the Personal Social Capital Scale (PSCS-16) was used to evaluate the participants' status of bonding and bridging social capital (BOC and BRC). Loneliness was assessed by the short-form UCLA Loneliness Scale (ULS-8). Multiple linear regression models were established to examine the relationship between social capital and loneliness. The BOC and BRC of rural widowed older people were significantly lower than those of widowed older people in urban areas, while loneliness of rural widowed older people was higher than that of widowed older people in urban areas. The result of the final model showed that loneliness of rural participants was significantly associated with both BOC (B = 0.141, p = 0.001) and BRC (B = -0.116, p = 0.003). The loneliness of the urban widowed sample had no association with both BOC and BRC (p > 0.05). These findings suggested that more social support and compassionate care should be provided to enrich the personal social capital and thus to reduce loneliness of widowed older adults, especially those in rural areas. The orofacial pain pathway projects to the parabrachial and amygdala, and sex steroids have been shown to affect neuronal activity in these regions. GABA positive cells in the amygdala are influenced by sex steroid metabolites to affect pain, and sex steroids have been shown to alter the expression of genes in the parabrachial, changing neuronal excitability. Mechanisms by which sex steroids affect amygdala and parabrachial signaling are unclear. The expression of genes in the parabrachial and amygdala in diestrus (low estradiol) and proestrus (high estradiol) female rats were evaluated in this study. First, varicella zoster virus was injected into the whisker pad of female rats to induce a pain response. Second, gene expression was quantitated using RNA-seq one week after injection. Genes that had the greatest change in expression and known to function in pain signaling were selected for the quantitation of protein content. Protein expression of four genes in the parabrachial and seven genes in the amygdala were quantitated by ELISA. In the parabrachial, neurexin 3 (Nrnx3) was elevated at proestrus. Nrnx3 has a role in AMPA receptor and GABA signaling. Neuronatin (Nnat) and protein phosphatase, Mg2+/Mn2+ dependent 1E (Ppm1e) were elevated in the parabrachial of diestrus animals both genes having a role in pain signaling. Epoxide hydroxylase (Ephx2) was elevated in the parabrachial at proestrus and the vitamin D receptor (Vdr) was elevated in the amygdala. Ephx2 antagonists and vitamin D have been used to treat neuropathic pain. In conclusion, sex steroids regulate genes in the parabrachial and amygdala that might result in the greater pain response observed during diestrus.Since 2005, The Michael J. Fox Foundation for Parkinson's Research (MJFF) has invested significant funding and non-funding effort to accelerate research and drug development activity around the Parkinson disease (PD)-associated protein LRRK2. MJFF has spearheaded multiple public/private pre-competitive collaborations that have contributed to our understanding of LRRK2 function; de-risked potential safety questions around the therapeutic use of LRRK2 kinase inhibitors; and generated critical research tools, biosamples, and data for the field. Several LRRK2-targeted therapies are now in human testing due to the hard work of so many in the PD community. In this perspective, we present a holistic description and model of how our Foundation's support targeted important barriers to LRRK2 research and helped move the field into clinical trials. The use of drones in combination with remote sensors have displayed increasing interest over the last years due to its potential to automate monitoring processes. In this study, a novel approach of a small flying e-nose is proposed by assembling a set of AlphaSense electrochemical-sensors to a DJI Matrix 100 unmanned aerial vehicle (UAV). The system was tested on an outdoor field with a source of NO2. read more Field tests were conducted in a 100 m2 area on two dates with different wind speed levels varying from low (0.0-2.9m/s) to high (2.1-5.3m/s), two flight patterns zigzag and spiral and at three altitudes (3, 6 and 9 m). The objective of this study is to evaluate the sensors responsiveness and performance when subject to distinct flying conditions. A Wilcoxon rank-sum test showed significant difference between flight patterns only under High Wind conditions, with Spiral flights being slightly superior than Zigzag. With the aim of contributing to other studies in the same field, the data used in this analysis will be shared with the scientific community.Responses of broiler chickens to dietary glycine equivalent (Glyequi) are affected by dietary cysteine and choline. Hence, this study investigated interactive effects among dietary Glyequi, cysteine, and choline on the growth of broiler chickens. Male Ross 308 broiler chickens were maintained in 105 metabolism units (10 birds/unit) from days 7 to 22. Excreta were collected in 12-h intervals from days 18 to 21. Blood was sampled on day 22 (1 bird/unit). Five levels each of Glyequi (9-21 g/kg), cysteine (2-5 g/kg), and choline (0.5-1.7 g/kg) were tested under 15 diets in 7 replicates each following a fractional central composite design. Another diet was provided to five metabolism units (15 birds/unit) to measure prececal amino acid digestibility. Data were evaluated using neural networks. The gainfeed ratio (GF) increased with digestible Glyequi intake. Differences between low and high digestible cysteine intake were low. Effects of choline intake on GF were low. Nitrogen-utilization efficiency (NUE) was high (≥77%), with low variation among treatments. Plasma metabolites varied among treatments and indicated that metabolism of Glyequi, cysteine, and choline was influenced. These findings showed that interactive effects of dietary Glyequi, cysteine, and choline on growth were small, possibly because NUE was barely influenced.Nucleic acid aptamers show clear promise as diagnostic reagents, as highly specific strands were reported against a large variety of biomarkers. They have appealing benefits in terms of reproducible generation by chemical synthesis, controlled modification with labels and functionalities providing versatile means for detection and oriented immobilization, as along with high biochemical and temperature resistance. Aptamers against immunoglobulin targets-IgA, IgM, IgG and IgE-have a clear niche for diagnostic applications, therefore numerous aptamers have been selected and used in combination with a variety of detection techniques. The aim of this review is to overview and evaluate aptamers selected for the recognition of antibodies, in terms of their design, analytical properties and diagnostic applications. Aptamer candidates showed convincing performance among others to identify stress and upper respiratory tract infection through SIgA detection, for cancer cell recognition using membrane bound IgM, to detect and treat hemolytic transfusion reactions, autoimmune diseases with IgG and detection of IgE for allergy diseases. However, in general, their use still lags significantly behind what their claimed benefits and the plethora of application opportunities would forecast.Downy mildew of grapevine is one of the most destructive grapevine diseases worldwide. Nowadays, downy mildew control relies almost exclusively on the use of chemical pesticides, including copper products, which are efficient but controversial due to their environmental toxicity. Natural plant protection products have become important solutions in the quest for the sustainable production of food and pest management. However, most biocontrol agents currently on the market, such as biofungicides or elicitors, have a limited efficacy; thus, they cannot replace chemical compounds in full. Our innovation is a natural active substance, which is a lysate of the amoeba Willaertia magna C2c Maky. This active substance is not only able to elicit grapevine defenses, but it also demonstrates direct fungicidal activity against Plasmopara viticola. The efficacy of this new natural substance was demonstrated both in a greenhouse and in a field. The amoeba lysate provided up to 77% protection to grapevine bunches in the field in a natural and safe way.
