Adenine nucleotide translocator 1 (ANT1), the only 4q35 gene involved in mitochondrial function, is strongly expressed in FSHD skeletal muscle biopsies. However, its role in FSHD is unclear. In this study, we evaluated ANT1 overexpression effects in primary myoblasts from healthy controls and during Xenopus laevis organogenesis. We also compared ANT1 overexpression effects with the phenotype of FSHD muscle cells and biopsies. Here, we report that the ANT1 overexpression-induced phenotype presents some similarities with FSHD muscle cells and biopsies. ANT1-overexpressing muscle cells showed disorganized morphology, altered cytoskeletal arrangement, enhanced mitochondrial respiration/glycolysis, ROS production, oxidative stress, mitochondrial fragmentation and ultrastructure alteration, as observed in FSHD muscle cells. ANT1 overexpression in Xenopus laevis embryos affected skeletal muscle development, impaired skeletal muscle, altered mitochondrial ultrastructure and led to oxidative stress as observed in FSHD muscle biopsies. Opportunistic salpingectomy (OS) is an attractive method for primary prevention of ovarian cancer. Although OS has not been associated with a higher complication rate, it may be associated with earlier onset of menopause.
To provide a systematic review and meta-analysis of the effect of OS on both age at menopause and ovarian reserve.
A search was conducted in the Cochrane Library, Embase and MEDLINE databases from inception until March 2022. We included randomized clinical trials and cohort studies investigating the effect of OS on onset of menopause and/or ovarian reserve through change in anti-Müllerian hormone (AMH), antral follicle count (AFC), estradiol (E2), follicle stimulating hormone (FSH) and luteinizing hormone (LH). Data was extracted independently by two researchers. Random-effects meta-analyses were conducted to estimate the pooled effect of OS on ovarian reserve.
The initial search yielded 1047 studies. No studies were found investigating the effect of OS on age of menopause. Fifteen studies were included in the meta-analysis on ovarian reserve. Meta-analyses did not result in statistically significant differences in mean change in AMH (MD -0.07ng/ml, 95%CI -0.18;0.05), AFC (MD 0.20 n, 95% CI -4.91;5.30), E2 (MD 3.97pg/ml, 95%CI -0.92;8.86), FSH (MD 0.33mIU/ml, 95%CI -0.15;0.81) and LH (MD 0.03mIU/ml; 95%CI -0.47;0.53).

Registration number PROSPERO CRD42021260966.Classic studies of medical education have examined how professional socialization reproduces the prevailing professional culture, as well as how students actively negotiate their place in educational processes. However, sociological research has not re-examined student culture in light of structural transformations in medical education, such as the introduction of new assessment types and their use as modes of commensuration. In this paper, we examine data from two studies of online forums where medical trainees and applicants to medical school discuss their experiences preparing for tests of professional skills, including judgment, empathy, and communication. Examining how medical students talk about these tests on such forums allows us to understand the meaning-making processes at work as students negotiate the commensuration processes such tests enable. We examine how these negotiations take place in online forums, where participants confront common challenges, form common perspectives, and share common solutions, all hallmarks of student culture. Through qualitative analysis, we find that online communities are spaces where students grapple with these new forms of commensuration, interrogate the standards and quantifications that underlie them, and collectively negotiate how to approach these assessments. Using the case of online forum communities, our findings advance past work on student culture in medical sociology by theorizing student culture as an extra-organizational phenomenon that spans multiple career stages. In so doing, we highlight the importance of online forum data for studying social processes.Pongo fossils with precise absolute age brackets are rare, limiting our understanding of their taxonomy and spatiotemporal distribution in southern China during the Late Pleistocene. Twenty-four isolated teeth of fossil orangutans were recently discovered during excavations at Yicun Cave in Guangxi Zhuang Autonomous Region, southern China. Here, we dated the fossil-bearing layer using Uranium-series dating of the associated flowstone and soda straw stalactites. Our results date the Yicun orangutan fossils to between 66 ± 0.32 ka and 57 ± 0.26 ka; thus, these fossils currently represent the last appearance datum of Pongo in southern China. We further conducted a detailed morphological comparison of the Yicun fossil teeth with large samples of fossil (n = 2454) and extant (n = 441) orangutans from mainland and island Southeast Asia to determine their taxonomic position. Compared to other fossil and extant orangutan samples, the Yicun Pongo assemblage has larger teeth and displays greater variation in occlusal structure. Based on the high frequency of cingular remnants and light to moderate enamel wrinkling of the molars, we assigned the Yicun fossils to Pongo weidenreichi, a species that was widespread in southern China throughout the Pleistocene. Lastly, we used published stable carbon isotope data of Early to Late Pleistocene mammalian fossil teeth from mainland Southeast Asia to reconstruct changes in the paleoenvironment and to interpret dental size variation of Pongo assemblages in a broader temporal and environmental context. The carbon isotope data show that dental size reduction in Pongo is associated with environmental changes. These morphological changes in Pongo appear to coincide with the expansion of savannah biomes and the contraction of forest habitats from the Middle Pleistocene onward. The variation in dental size of forest-dwelling Pongo in mainland Southeast Asia may have resulted from habitat differentiation during the Pleistocene.
Absent septum pellucidum (ASP) is a brain abnormality often associated with neuroanatomic abnormalities including septo-optic dysplasia (SOD). We aimed to determine how frequently prenatally diagnosed isolated ASP is confirmed by postnatal imaging and to examine clinical outcomes for ASP.
This was a retrospective study of maternal-fetal dyads referred to Children's National Hospital from January 1, 2012, to June 30, 2019. We included cases with fetal diagnosis of isolated or complex ASP. Diagnosis was based on ASP and the presence or absence of additional neuroanatomic findings. Data included obstetric and birth history, genetic testing, imaging, and neurodevelopmental outcomes.
ASP was diagnosed in 35 fetuses. Of 17 fetuses with isolated ASP, 10 had postnatal evaluation. In five (50%) isolated ASP cases, postnatal imaging revealed additional brain abnormalities. The five children with postnatally confirmed isolated ASP had lower rates of hydrocephalus (0% vs 54%) and abnormal feeding (0% vs 20%), hearing (0% vs 14%), and vision (0% vs 14%) than those with complex ASP (n= 17). Children with isolated ASP had lower rates of developmental delay (33% vs 50%) and seizures (11% vs 30%) than children with complex ASP. One child with prenatal isolated ASP was diagnosed with SOD (10%).

This prospective, observational study assessed TIA/ischaemic stroke patients before (baseline; N=60), at 14 ±7days (14d, N=39) and≥90days (90d, N=31) after adding dipyridamole to aspirin. Platelet function/reactivity at high shear stress (PFA-100® C-ADP) and low shear stress (VerifyNow® P2Y12 and Multiplate® ADP assays), and platelet activation status (% expression of CD62P, CD63 and leucocyte-platelet complexes on whole blood flow cytometry) were quantified. 'Dipyridamole-high on-treatment platelet reactivity (HTPR)' was defined as failure to inhibit ADP-induced platelet aggregation +/- adhesion compared with the patient's baseline on aspirin monotherapy by more than twice the coefficient-of-variation of the assay after adding dipyridamole to aspirin.
