Intrahepatic cholangiocarcinoma is a malignant tumor originating from intrahepatic bile ducts. Surgical therapy, radiotherapy, and chemotherapy are taken to treat this disease, but it is prone to recurrence and metastasis, with poor prognosis. Therefore, it is of great significance to explore new targets and molecular mechanisms for the development of cholangiocarcinoma cells. Clinical cholangiocarcinoma tissues from patients and four human cholangiocarcinoma cell lines were analyzed for microRNA-373 (miR-373) expression. For investigating whether miR-373 directly modulated unc-51 like autophagy activating kinase 1 (ULK1), dual-luciferase reporter assay was performed. In addition, CCK-8 assay, flow cytometry, western blot, and immunofluorescence were applied to evaluate the proliferation, apoptosis, and autophagy of cholangiocytic hepatocellular carcinoma cells. miR-373 downregulation was observed in clinical tissues and cell lines of cholangiocarcinoma. The dataset will be continuously improved and advanced based on the experience and enhanced understanding of data collection in the forthcoming years. We describe an infant with a clinical diagnosis of Cornelia de Lange syndrome (CdLS) in whom chromosome analysis revealed a de novo complex balanced translocation, 46,XY,t(5;7;6)(q11.2;q32;q13)dn. Subsequent molecular characterization by whole-genome sequencing (WGS) identified 23 breakpoints, delineating segments derived from four chromosomes (5;6;7;21) in ancestral or inverted orientation. One of the breakpoints disrupted a known CdLS gene, NIPBL. Further investigation revealed paternal origin of the CCR allele, clustering of the breakpoint junctions, and molecular repair signatures suggestive of a single catastrophic event. Notably, very short DNA segments (25 and 41 bp) were included in the reassembled chromosomes, lending additional support that the DNA repair machinery can detect and repair such segments. Interestingly, there was an independent paternally derived miniscule complex rearrangement, possibly predisposing to subsequent genomic instability. In conclusion, we report a CCR causing a monogenic Mendelian disorder, urging WGS analysis of similar unsolved cases with suspected Mendelian disorders. DESIGN This study was a qualitative study in which data were collected and analyzed in accordance with the methodological principles of Reflective Lifeworld Research with a phenomenological approach. METHODS Face-to-face interviews were performed with 17 adolescents after undergoing the first colonoscopy. RESULTS The phenomenon of undergoing colonoscopy as experienced by adolescents can be described as a collision between emotions and a desire to obtain answers to questions about the examination, as well as concerns about its result and the meaning of undergoing colonoscopy. The essential meaning is additionally described through its constituents a sense of vulnerability, an opportunity for symptom explanation, and sensibility regarding information. CONCLUSIONS The results can be concluded in terms of the knowledge that for adolescents a colonoscopy means more than an examination. Although colonoscopy is not experienced as painful, it evokes different emotions that affect adolescents. Therefore, a psychological preparation, on an individual level, is required before the colonoscopy. Our results showed that adolescents need to understand the connection between their symptoms, their body, and the colonoscopy. © 2020 The Authors. Journal for Specialists in Pediatric Nursing Published by Wiley Periodicals, Inc.The use of fish embryo toxicity (FET) data for hazard assessments of chemicals, in place of acute fish toxicity (AFT) data, has long been the goal for many environmental scientists. The FET test was first proposed as a replacement to the standardized AFT test nearly 15 years ago, but as of now, it has still not been accepted as a standalone replacement by regulatory authorities such as the European Chemicals Agency (ECHA). However, the ECHA have indicated that FET data can be used in a weight of evidence (WoE) approach, if enough information is available to support the conclusions related to the hazard assessment. To determine how such a WoE approach could be applied in practice has been challenging. To provide a conclusive WoE for FET data, we have developed a Bayesian Network (BN) to incorporate multiple lines of evidence to predict AFT. There are four different lines of evidence in this BN model including 1) physico-chemical properties; 2) AFT data from chemicals in a similar class or category; 3) ecotoxicity data from other trophic levels of organisms (e.g. daphnids and algae) and 4) measured FET data. The BN model was constructed from data obtained from a curated database [the company] and conditional probabilities assigned for the outcomes of each line of evidence. To evaluate the model, 20 data rich chemicals, containing a minimum of three AFT and FET test data points, were selected to ensure a suitable comparison could be performed. The results of the AFT predictions indicated that the BN model could accurately predict the toxicity interval for 80% of the chemicals evaluated. For the remaining chemicals (20%), either daphnids or algae were the most sensitive test species, and for those chemicals, the daphnid or algal hazard data would have driven the environmental classification. All rights reserved.BACKGROUND The National Institute for Health and Care Excellence (NICE) Evidence search Student Champion Scheme aims to enable undergraduate health and social care students to teach their peers skills for information literacy (IL), thereby encouraging future evidence-based practice. OBJECTIVES To analyse the Student Champions' teaching methods; discover what effects the Scheme had on their IL; and uncover any differences between disciplines. METHODS Fifty-one reflective reports, written by Student Champions and submitted to NICE, were thematically analysed using a non-linear six-stage model. Four health disciplines from academic year 2017/2018 were featured. RESULTS (a) Students preferred active teaching methods; (b) reported benefits of participation included gaining/developing new skills and increased confidence; (c) students believed that participating improved their skills for IL; (d) multiple recommendations for improving the Scheme were given; and (e) students wanted the Scheme to be offered earlier in their degrees. DISCUSSION Champions from all disciplines positively benefit from participating in the Scheme. However, they also have concerns which are not well-documented in the literature. CONCLUSION Student Champions have overall positive experiences. There is demonstrated improvement in their IL, and they become familiar with a useful evidence-based practice resource. They also offer recommendations for future improvements to the Scheme. © 2020 Health Libraries Group.Self-management helps patients understand their illnesses and learn about disease-related skills so they can better manage their situation. However, published studies on the effectiveness of self-management for chronic obstructive pulmonary disease (COPD) are heterogeneous. Quasi-experimental design was used to investigate the effectiveness of a self- management program in self-care efficacy and quality of life in patients with COPD. Sixty participants who had experienced COPD were assigned to control group (usual care) or experimental group (self-care program) (11 ratio) according to a purposeful sampling at a Medical Center in Taiwan. The Medical Research Council dyspnea scale, the COPD Self-Efficacy Scale, the Clinical COPD Questionnaire were used to analyze data. The Medical Research Council dyspnea scale was more significant improvement in the two months after discharge in the experimental group than in the control group. The COPD Self-Efficacy Scale and Clinical COPD Questionnaire scores in the experimental group within 3 months after discharge were significantly improved compared with the control group. All rights reserved.Herein, we report a degradable film that can be coated on various substrates by the codeposition of dopamine and cystamine. The thickness of the resulting film (pDC) varies depending on the initial ratio of dopamine/cystamine dissolved in a solution; the thickest film (ca. 60 nm) is obtained under optimized codeposition conditions. Selective degradation of pDC occurs in the presence of tris(2-carboxyethyl)phosphine (TCEP), the reaction kinetics of which are highly dependent on the TCEP concentration. For further application as a drug-delivery platform, doxorubicin can be loaded within the pDC film, which is released actively under film degradation in response to TCEP. We expect that the developed pDC film will be a useful tool for developing drug delivery cargo, antibacterial surface, and cell surface coating for various biomedical applications. © 2020 Wiley-VCH Verlag GmbH & Co.
