The expansion of pharmacy technicians' roles in community pharmacies allows pharmacists the opportunity to focus on providing clinical services to patients. This study explores the tasks pharmacy technicians' perform to support Med Sync programs in community pharmacies. Pharmacy staff members at North Carolina pharmacies with more than fifty percent of their prescription volume being dispensed as part of a Med Sync program were recruited to participate in semi-structured interviews. Inductive coding and summary analysis were used to analyze the interview data. Study participants described pharmacy technicians' roles in identifying patients for marketing and enrollment, reviewing patients' medications list, choosing alignment dates based on patient preference, contacting patients in preparation for dispensing and, lastly, engaging in pickup or delivery of medications. This study highlights technicians' vital role in completing tasks that support Med Sync programs in community pharmacies.Insulin resistance, the hallmark of type 2 diabetes mellitus (T2DM), is linked to hyperinsulinemia, which develops to counterbalance initial peripheral hormone resistance. Studies indicate that chronically elevated levels of insulin lead to skeletal muscle insulin resistance by deregulating steps within the insulin signaling cascade. The polyphenol resveratrol (RSV) has been shown to have antidiabetic properties in vitro and in vivo. In the present study, we examined the effect of RSV on high insulin (HI)-induced insulin resistance in skeletal muscle cells in vitro and investigated the mechanisms involved. Parental and GLUT4myc-overexpressing L6 rat skeletal muscle cells were used. [3H]2-deoxyglucose (2DG) uptake was measured, and total and phosphorylated levels of specific proteins were examined by immunoblotting. Exposure of L6 cells to HI levels (100 nM) for 24 h decreased the acute-insulin-stimulated 2DG uptake, indicating insulin resistance. HI increased ser307 and ser636/639 phosphorylation of IRS-1 (to 184% ± 12% and 225% ± 28.9% of control, with p less then 0.001 and p less then 0.01, respectively) and increased the phosphorylation levels of mTOR (174% ± 6.7% of control, p less then 0.01) and p70 S6K (228% ± 33.5% of control, p less then 0.01). Treatment with RSV abolished these HI-induced responses. Furthermore, RSV increased the activation of AMPK and restored the insulin-mediated increase in plasma membrane GLUT4 glucose transporter levels. These data suggest that RSV has a potential to counteract the HI-induced muscle insulin resistance.The main objective of this work is to study the effect of hydroxylamine sulfate or stabilizer states (solid vs liquid) on the storage hardening of natural rubber (NR). Several types of natural rubber samples were prepared unstabilized NR samples and stabilized NR samples (i) dry NR with 0.2 and 2.0 parts per hundred rubber (phr) of dry hydroxylamine sulfate, and (ii) natural latex with 0.2 and 2.0 phr of liquid hydroxylamine sulfate. The samples were characterized immediately (time 0) and after 12 weeks of storage at room temperature, respectively. We found that the Mooney viscosity, gel content, and Wallace plasticity of NR without a stabilizer increases with storage hardening for 12 weeks. However, two types of stabilized NR samples represent constant values of those three parameters, because hydroxylamine sulfate inhibits network and gel formation in NR. Interestingly, the mixing states (solid vs liquid) between natural rubber and the stabilizer affect the properties of stabilized NR. This could be explained by the better dispersion and homogeneous nature of liquid stabilizers in natural latex (liquid state), and thus the higher loading of the stabilizer in the liquid state. This is important, as the stabilization of NR properties as a function of time is required by rubber industry. Analysis of historical data revealed a strong association between abundance of Bifidobacterium in the gut microbiome of breasted infants and stool pH, suggesting that this taxon plays a key role in determining the pH in the gut. Bifidobacterium longum subsp. infantis is uniquely equipped to metabolize human milk oligosaccharides (HMO) from breastmilk into acidic end products, mainly lactate and acetate. The presence of these acidic compounds in the infant gut is linked to a lower stool pH. Conversely, infants lacking B. infantis have a significantly higher stool pH, carry a higher abundance of potential pathogens and mucus-eroding bacteria in their gut microbiomes, and have signs of chronic enteric inflammation. This suggests the presence of B. infantis and low intestinal pH may be critical to maintaining a protective environment in the infant gut. In this work, we first confirmed a positive correlation of ADAMTS1 with endothelial-like phenotype of human melanoma cells together with the finding of associated signatures, including key genes such as endothelial CDH5. Using a CRISPR-Cas9 approach, we observed that the inhibition of ADAMTS1 in an aggressive uveal melanoma model compromised its endothelial-like properties, and more importantly, caused a robust blockade on the progression of tumor xenografts. Although vasculature emerged affected in ADAMTS1-deficient tumors, the most relevant action implied the downregulation of endothelial CDH5 in tumor cells, in association with stemness markers. Indeed, melanoma sphere assays also revealed a deficient commitment to form spheres in the absence of ADAMTS1, directly correlating with stemness markers and, remarkably, also with CDH5.
