Nuclear factor 90 (NF90) is a novel virus sensor that serves to initiate antiviral innate immunity by triggering stress granule (SG) formation. However, the regulation of the NF90-SG pathway remains largely unclear. We found that Tim-3, an immune checkpoint inhibitor, promotes the ubiquitination and degradation of NF90 and inhibits NF90-SG-mediated antiviral immunity. Vesicular stomatitis virus (VSV) infection induces the up-regulation and activation of Tim-3 in macrophages, which in turn recruit the E3 ubiquitin ligase TRIM47 to the zinc finger domain of NF90 and initiate a proteasome-dependent degradation via K48-linked ubiquitination at Lys297. Targeted inactivation of Tim-3 enhances the NF90 downstream SG formation by selectively increasing the phosphorylation of protein kinase R and eukaryotic translation initiation factor 2α, the expression of SG markers G3BP1 and TIA-1, and protecting mice from VSV challenge. These findings provide insights into the crosstalk between Tim-3 and other receptors in antiviral innate immunity and its related clinical significance.Enterotoxigenic Escherichia coli (ETEC) expressing the colonization pili CFA/I are common causes of diarrhoeal infections in humans. Here, we use a combination of transposon mutagenesis and transcriptomic analysis to identify genes and pathways that contribute to ETEC persistence in water environments and colonization of a mammalian host. ETEC persisting in water exhibit a distinct RNA expression profile from those growing in richer media. Multiple pathways were identified that contribute to water survival, including lipopolysaccharide biosynthesis and stress response regulons. The evolution of the genomics era has led to the generation of huge volumes of sequencing data at an unprecedented rate due to the ever-reducing costs of whole-genome sequencing (WGS). We have developed the Rapid Microbial Analysis Pipeline (rMAP), a user-friendly pipeline capable of profiling the resistomes of ESKAPE pathogens (Enterococcus faecium, Staphylococcus aureus, Klebsiella pneumoniae, Acinetobacter baumannii, Pseudomonas aeruginosa and Enterobacter species) using WGS data generated from Illumina's sequencing platforms. rMAP is designed for individuals with little bioinformatics expertise, and automates the steps required for WGS analysis directly from the raw genomic sequence data, including adapter and low-quality sequence read trimming, de novo genome assembly, genome annotation, single-nucleotide polymorphism (SNP) variant calling, phylogenetic inference by maximum likelihood, antimicrobial resistance (AMR) profiling, plasmid profiling, virulence factor determination, multi-locus sequence typing (MLST), pangenome analysis and insertion sequence characterization (IS). Once the analysis is finished, rMAP generates an interactive web-like html report. rMAP installation is very simple, it can be run using very simple commands. It represents a rapid and easy way to perform comprehensive bacterial WGS analysis using a personal laptop in low-income settings where high-performance computing infrastructure is limited.Co-endemicity of SARS-CoV-2 and dengue virus (DV) infection is becoming a matter of serious concern as it has been already reported that antibodies (Ab) elicited by SARS-CoV-2 infection can produce false-positive results in dengue IgG and IgM rapid tests and vice versa. The electronic version of the article has already been corrected.This study investigated social support exchanges on social media among male homosexuals who are HIV-positive (MHHP) in China and explored how online self-disclosure relates to such activity. Analyses were based on 9,459 Weibo messages posted by 188 targeted users.Results showed that the informational and emotional support messages were prevalent in the Chinese context, and esteem social support was first found to be the most salient by the current investigation. Results of independent samples t-tests suggested that MHHP who engaged in significant self-disclosure on Weibo tended to have more frequent social support exchanges than those who did not. In particular, MHHP who expressed sex-seeking intentions and companionship seeking intentions were likely to post more supportive messages than those who did not express such intentions. Moreover, MHHP who disclosed their concerns over discrimination tended to post more emotional, informational, and esteem support messages than those who did not express discrimination concerns.The phenomenon of the secondary reactions of volatiles prevails during the pyrolysis process of scrap tires, but less is known about the influence of volatiles' residence time and temperature on the pyrolytic oil compositions. This study sought to investigate the effect of a lower limb IPC protocol, with either a 5-min or 45-min post-application delay, on vastus lateralis tissue saturation index (TSI) and systemic cardiac hemodynamics at rest and during short-duration intense cycling. Twelve young adults randomly completed four interventions IPC (at 220 mmHg) with 5-min delay (IPC5), IPC with 45-min delay (IPC45), SHAM (at 20 mmHg) with 5-min delay (SHAM5), and SHAM with 45-min delay (SHAM45). Following IPC intervention and recovery delay, participants completed 5, 60-s high-intensity (100% Wpeak) cycle sprints separated by 120-sec of active recovery (30% Wpeak). Compared to baseline, TSI immediately following IPC5, but pre-exercise, remained lower than the equivalent for IPC45 (-5.9 ± 1.5%, p = .002). IPC, imposed at least 45-min before the completion of five 60-s sprint cycling efforts, significantly enhanced TSI during active recovery between sprint intervals compared to a 5-min delay (6.6 ± 2.4%, p = .021), and identical SHAM conditions (SHAM5 5.8 ± 2.2%, p = .024; SHAM45 6.2 ± 2.5%, p = .029). A 45-min delay following IPC appears to provide heightened skeletal muscle metabolic rebound prior to intense sprint cycling as compared to a 5-min delay. Furthermore, IPC followed by a 45-min delay enhanced recovery of skeletal muscle oxygenation during low intensity active sprint recovery, despite an unchanged decline in skeletal muscle oxygenation during near-maximal sprinting efforts.In this study, we explored the factors relating to face and body visibility in young men who have sex with men (YMSM) from online dating applications. Research on visual self-presentation in online dating applications, especially in YMSM, remains scarce. The sample consisted of 339 young men (ages 18-24 years) who had casual sex with one or more male partners from online dating applications. Participants completed an online survey asking how visible their face and body were on their profile pictures. For body visibility, participants rated the visibility of their upper and lower body separately. We also computed difference scores between face and body visibility to measure face visibility in relation to body visibility (e.g., showing more-visible face compared to body). Predictors included self-rated attractiveness, fear of being identified on the applications, and motives for application use. Participant age and duration of application use were statistically controlled in all analyses. Results showed that higher perceived attractiveness related to higher upper- and lower-body visibility on profile pictures. Greater fear of being identified related to lower face visibility and less-visible face relative to upper- and lower-body visibility. Having a relationship motive was associated with higher face visibility. Older age and longer duration of application use were associated with more-visible face relative to lower body. The findings contribute to the literature on visual self-presentation in online dating applications in young and vulnerable sexual minorities.The scoping review examines and summarises the available knowledge base on intervention techniques focused on positive memories. An iterative series of PsycInfo and Medline searches was conducted up to April 2021 following PRISMA-ScR guidelines. Thirty-nine studies, spanning 12 intervention techniques, were selected and described including (1) theoretical basis; (2) type of study, sample, and measures; (3) intervention protocol; and (4) results of empirical studies if applicable. Results indicated that most techniques have only been tested in one-two studies with modest sample sizes and, when follow-ups are conducted, they are typically short. Results indicate that working with positive memories has the greatest impact on improving positive affect and reducing depressive symptoms, and that these effects are often temporary. This review serves as a quick reference guide to help professionals' access to descriptions and information on empirical evidence of positive memory techniques, improving their therapeutic arsenal to enhance well-being and therapeutic outcomes in their patients.Evidence from theories of Developmental Origins of Health and Disease (DOHaD) suggests that experiencing adverse early life conditions subsequently leads to detrimental adult health outcomes. The bulk of empirical DOHaD literature does not consider the nature and magnitude of the impact of adverse early life conditions at the population level. In particular, it ignores the distortion of age and cohort patterns of adult health and mortality and the increased load of chronic illness and disability that ensues. In this paper, we use a microsimulation model combined with empirical estimates of incidence and prevalence of obesity, type 2 diabetes, and associated disability in low- and middle-income countries to assess the magnitude of delayed effects on adult healthy life expectancy and on compression (or expansion) of morbidity at older ages. The main goal is to determine if, in what ways, and to what extent delayed effects due to early conditions can influence cohorts' chronic illness and disability profiles.
