Horseradish root is used as a condiment due to its extremely pungent flavour, deriving from the high content of glucosinolates and their breakdown products such as isothiocyanates and other sulfur compounds. Horseradish also has a long history in ethnomedicine. In this study the anti-inflammatory potential of three accessions of Armoracia rusticana on lipopolysaccharide from E. coli treated J774A.1 murine macrophages was evaluated. Our results demonstrate that Armoracia rusticana reduced nitric oxide, tumor necrosis factor-α and interleukin-6 release and nitric oxide synthase and cyclooxygenase-2 expression in macrophages, acting on nuclear transcription factor NF-κB p65 activation. Moreover Armoracia rusticana reduced reactive oxygen species release and increased heme-oxygenase-1 expression, thus contributing to the cytoprotective cellular effect during inflammation.Alterations in platelet function are a common finding in surgical procedures involving cardiopulmonary bypass and hypothermia. Although the combined impact of hypothermia and artificial circulation on platelets has been studied before, the ultimate strategy to safely minimize the risk for bleeding and thrombosis is yet unknown. The aim of this study was to evaluate the use of a mock circulation loop to study the impact of hypothermia for platelet-related hemostatic changes. Venous blood was collected from healthy adult humans (n = 3). Closed mock circulation loops were assembled, each consisting of a centrifugal pump, an oxygenator with integrated heat exchanger, and a hardshell venous reservoir. The experiment started with the mock circulation temperature set at 37°C (T0 [0 h]). Cooling was then initiated at T1 (+2 h), where temperature was adjusted from 37°C to 32°C. Hypothermia was maintained from T2 (+4 h) to T3 (+28 h). From that point in time, rewarming from 32°C to 37°C was initiated with similar speedwarming.

None.The successful synthesis and solid state NMR characterization of silica-based organic-inorganic hybrid materials is presented. For this, collagen-like peptides are immobilized on carboxylate functionalized mesoporous silica (COOH/SiOx) materials. A pre-activation of the silica material with TSTU (O-(N-Succinimidyl)-N,N,N',N'-tetramethyluronium tetrafluoroborate) is performed to enable a covalent binding of the peptides to the linker. The success of the covalent immobilization is indicated by the decrease of the (13)C CP-MAS NMR signal of the TSTU moiety. A qualitative distinction between covalently bound and adsorbed peptide is feasible by (15)N CP-MAS Dynamic Nuclear Polarization (DNP). The low-field shift of the (15)N signal of the peptide's N-terminus clearly identifies it as the binding site. The DNP enhancement allows the probing of natural abundance (15)N nuclei, rendering expensive labeling of peptides unnecessary.Under Magic-Angle Spinning (MAS), a long radio-frequency (rf) pulse applied on resonance achieves the selective excitation of the center-band of a wide NMR spectrum. We show herein that these rf pulses can be applied on the indirect channel of Hetero-nuclear Multiple-Quantum Correlation (HMQC) sequences, which facilitate the indirect detection via spin-1/2 isotopes of nuclei exhibiting wide spectra. Numerical simulations show that this indirect excitation method is applicable to spin-1/2 nuclei experiencing a large chemical shift anisotropy, as well as to spin-1 isotopes subject to a large quadrupole interaction, such as (14)N. The performances of the long pulses are analyzed by the numerical simulations of scalar-mediated HMQC (J-HMQC) experiments indirectly detecting spin-1/2 or spin-1 nuclei, as well as by dipolar-mediated HMQC (D-HMQC) experiments achieving indirect detection of (14)N nuclei via (1)H in crystalline γ-glycine and N-acetyl-valine samples at a MAS frequency of 60kHz. We show on these solids that for the acquisition of D-HMQC spectra between (1)H and (14)N nuclei, the efficiency of selective moderate excitation with long-pulses at the (14)N Larmor frequency, ν0((14)N), is comparable to those with strong excitation pulses at ν0((14)N) or 2ν0((14)N) frequencies, given the rf field delivered by common solid-state NMR probes. Furthermore, the D-HMQC experiments also demonstrate that the use of long pulses does not produce significant spectral distortions along the (14)N dimension. In summary, the use of center-band selective weak pulses is advantageous for HMQC experiments achieving the indirect detection of wide spectra since it (i) requires a moderate rf field, (ii) can be easily optimized, (iii) displays a high robustness to CSAs, offsets, rf-field inhomogeneities, and fluctuations in MAS frequency, and (iv) is little dependent on the quadrupolar coupling constant.Molecular crystals demonstrate drastically different behavior in solid and liquid state, mainly due to their difference in structural frameworks. Therefore, designing of unique structured molecular compound which can work at both these interfaces has been a challenge. Here, we present remarkable 'molecular' property by non-porous molecular solid crystal, dinuclear copper complex (C6H5CH(X)NH2)2CuCl2, to reversibly 'adsorb' HCl gas at solid-gas interface as well as 'accommodate' azide anion at solid-liquid interface with crystal to crystal transformation. The latter process is driven by molecular recognition, self-assembly, and anchimeric assistance. The observed transformations are feasible due to breathing of inner and outer coordination sphere around metal center resulting in change in metal polyhedra for 'accommodating' guest molecule. These transformations cause changes in optical, magnetic, and/or ferroelectric property offering diversity in 'sensing' application. With the proposed underlying principles in these exceptional reversible and cyclic transformations, we prepared a series of compounds, can facilitate designing of novel multifunctional molecular materials.

Vena cava compression is a relatively rare initial manifestation of underlying malignancy. The superior vena cava syndrome, which is characterized by facial plethora, jugular venous distension, and arm swelling, is a well-known entity associated with bronchogenic carcinoma. Less common is the compression of the inferior vena cava. To the best of my knowledge, this is the first reported case of newly diagnosed gastric adenocarcinoma presenting initially as the inferior vena cava syndrome. Heart rate-corrected QT interval (QTc) is an indicator of risk, however it is imperfect, and subtle abnormalities of repolarization have been linked with arrhythmogenesis.

Identification of electrocardiographic predictors of torsadogenic risk through the application of a novel T wave analysis tool.

Among all patients admitted to Mayo Clinic for initiation of dofetilide or sotalol, we identified 13 cases who developed drug-induced TdP and 26 age and sex matched controls that did not develop TdP. The immediate pre-TdP ECG of those with TdP was compared to the last ECG performed prior to hospital discharge in controls using a novel T wave program that quantified subtle changes in T wave morphology.

The QTc and 12T wave parameters successfully distinguished TdP cases from controls. The top performing parameters were the QTc in lead V3 (mean case vs control 480 vs 420msec, p < 0.001, r = 0.72) and T wave right slope in lead I (mean case vs control -840.29 vs -1668.71mV/s, p = 0.002, r = 0.45). The addition of T wave right slope to QTc improved prediction accuracy from 79 to 88%.

Our data demonstrate that, in addition to QTc, the T wave right slope is correlated strongly with TdP risk. This suggests that a computer-based repolarization measurement tool that integrates additional data beyond the QTc may identify patients with the greatest torsadogenic potential.
