Metabolism serves mammalian feeding and active behavior, and is controlled by circadian clock. The molecular mechanism by which clock factors regulate metabolic homeostasis under oxidative stress is unclear. Here, we have characterized that the daily oxygen consumption rhythm was deregulated in Per1 deficient mice. Per1 deficiency impaired daily mitochondrial dynamics and deregulated cellular GPx-related ROS fluctuations in the peripheral organs. We identified that PER1 enhanced GPx activity through PER1/GPX1 interaction in cytoplasm, consequently improving the oxidative phosphorylation efficiency of mitochondria. Per1 expression was specifically elevated in the fasting peripheral organs for protecting mitochondrial from oxidation stress. These observations reveal that Per1-driven mitochondrial dynamics is a critical effector mechanism for the regulation of mitochondrial function in response to oxidation stress.Ferroptosis is a newly defined form of cell death induced by iron-dependent accumulation of lethal lipid peroxidation. Ferroptosis represent a therapeutic strategy to suppress therapy-resistant cancer cells with more property of epithelial-mesenchymal transition (EMT). However, epigenetic reprogramming of EMT has been rarely studied in the context of ferroptosis susceptibility. Therefore, we examined the therapeutic potentiality of EMT epigenetic reprogramming in promoting ferroptosis in head and neck cancer (HNC) cells. The effects of ferroptosis inducers and EMT inhibition or induction were tested in HNC cell lines and mouse tumor xenograft models. These effects were analyzed concerning cell viability and death, lipid reactive oxygen species and iron production, labile iron pool, glutathione contents, NAD/NADH levels, and mRNA/protein expression. Cell density and the expression levels of E-cadherin, vimentin, and ZEB1 were associated with the different susceptibility to ferroptosis inducers. CDH1 silencing or ZEB1 overexpression increased the susceptibility to ferroptosis, whereas CDH overexpression or ZEB1 silencing decreased the susceptibility, in vitro and in vivo. Histone deacetylase SIRT1 gene silencing or pharmacological inhibition by EX-527 suppressed EMT and consequently decreased ferroptosis, whereas SIRT inducers, resveratrol and SRT1720, increased ferroptosis. MiR-200 family inhibitors induced EMT and increased ferroptosis susceptibility. In HNC cells with low expression of E-cadherin, the treatment of 5-azacitidine diminished the hypermethylation of CDH1, resulting in increased E-cadherin expression and decreased ferroptosis susceptibility. Our data suggest that epigenetic reprogramming of EMT contributes to promoting ferroptosis in HNC cells.Glaucoma is a leading cause of blindness worldwide in individuals 60 years of age and older. Despite its high prevalence, the factors contributing to glaucoma progression are currently not well characterized. Glia-driven neuroinflammation and mitochondrial dysfunction play critical roles in glaucomatous neurodegeneration. Here, we demonstrated that elevated intraocular pressure (IOP) significantly decreased apolipoprotein A-I binding protein (AIBP; gene name Apoa1bp) in retinal ganglion cells (RGCs), but resulted in upregulation of TLR4 and IL-1β expression in Müller glia endfeet. Apoa1bp-/- mice had impaired visual function and Müller glia characterized by upregulated TLR4 activity, impaired mitochondrial network and function, increased oxidative stress and induced inflammatory responses. We also found that AIBP deficiency compromised mitochondrial network and function in RGCs and exacerbated RGC vulnerability to elevated IOP. Administration of recombinant AIBP prevented RGC death and inhibited inflammatory responses and cytokine production in Müller glia in vivo. These findings indicate that AIBP protects RGCs against glia-driven neuroinflammation and mitochondrial dysfunction in glaucomatous neurodegeneration and suggest that recombinant AIBP may be a potential therapeutic agent for glaucoma.The study of visual illusions has proven to be a very useful approach in vision science. In this work we start by showing that, while convolutional neural networks (CNNs) trained for low-level visual tasks in natural images may be deceived by brightness and color illusions, some network illusions can be inconsistent with the perception of humans. On one hand, the proposed linear eigenanalysis explains the overall similarities in simple CNNs trained for tasks like denoising or deblurring, the linear version of the network has center-surround receptive fields, and global transfer functions are very similar to the human achromatic and chromatic contrast sensitivity functions in human-like opponent color spaces. These similarities are consistent with the long-standing hypothesis that considers low-level visual illusions as a by-product of the optimization to natural environments. Specifically, here human-like features emerge from error minimization. On the other hand, the observed differences must be due to the behavior of the human visual system not explained by the linear approximation. However, our study also shows that more 'flexible' network architectures, with more layers and a higher degree of nonlinearity, may actually have a worse capability of reproducing visual illusions. This implies, in line with other works in the vision science literature, a word of caution on using CNNs to study human vision on top of the intrinsic limitations of the L + NL formulation of artificial networks to model vision, the nonlinear behavior of flexible architectures may easily be markedly different from that of the visual system.A rise in the shipping of heavier hydrocarbon products increases the potential for an oil to sink after a spill. Further, sunken oil is difficult to locate and recover, and appropriate response technologies depend on the sinking mechanism. In this review, principal sinking mechanisms for oil are described and appropriate response technologies are suggested. Then, models appropriate for tracking sunken oil are compared. Oil may sink as burn residue, microscopic oil-particle aggregates (OPAs) or macroscopic oil-sediment mixtures (OSMs), marine oil snow during a MOSSFA event, or due to its high density. The most common mechanism is by sediment entrainment, and in such scenarios manual recovery has been reported as a successful response option. Among oil tracking models, trajectory models and Bayesian oil search models are compared for sunken oil capabilities. Many oil spill models require hydrodynamic inputs, whereas Bayesian models infer parameters based on available field concentration and bathymetric data.The characterization of petroleum or its products spilled in the environment in relation to its source rocks is an important tool to assist in the resolution of issues of environmental impact and legal responsibility since it clarifies the possible region or the producing country of that type of petroleum. link2 This article analyzes the application of analytical techniques for petroleum geochemistry to identify the type and origin of oily material collected from beaches in Northeast Brazil in 2019. Samples of the oily material collected on the beaches in the states of Bahia and Sergipe were analyzed and for comparison purposes, samples of crude oils produced in Brazil, Middle East, Nigeria and Venezuela were also analyzed. The analytical results showed that the oily material that reached the beaches has geochemical characteristics compatible with those of the Venezuelan oil, indicating it was severely weathered or a product made with heavy oil produced.In the South Pacific (SP) pearl farming atolls, wind is the main driver of lagoon water circulation, affecting dispersal and survival of pearl oyster larvae. To characterize typical wind conditions in the SP, wind regime classifications are performed from regional climate simulations using the WRF model, for present-day and for the end of the 21st century under RCP8.5 scenario conditions. At the daily time-scale, 4 regimes are identified a trade-wind, a north-easterly, and two easterly regimes. Their characteristics are driven by large-scale circulation and climate modes of variability. In future projection, all regimes are characterized by a ~15% wind speed increase, while directions and occurrence frequencies undergo marginal changes. At the monthly time-scale that corresponds to pearl oyster pelagic larval duration, nine wind regimes are determined including three regimes with wind reversals. These regimes can be used to model typical lagoon conditions during larval dispersal.We evaluated the incidence of microplastics in the digestive tract contents of four fish species Engraulis encrasicolus, Sardina pilchardus, Callionymus lyra and Mullus surmuletus. Alkaline digestion (10% KOH) was used to degrade the organic matter. The percentage of fish with microplastics was 78% (88% fibres, 12% fragments). The main types of polymers identified by Raman spectroscopy were polyethylene and polypropylene. The diet of the four species was also studied and two feeding types were determined plankton-feeders and benthic-feeders. The effect of a set of biological variables (Length, Fullness index, Fulton's condition factor and Feeding type) on the number and size of microplastics ingested was studied using Generalised Additive Models (GAMs). A significant increase in the number of microplastics with increasing length was observed. No significant effect of trophic variables (fullness index and feeding type) on the number and size of microplastics was found.This review summarizes the current literature on the epidemiology of orthostatic hypotension (OH) in the elderly and in patients with autonomic impairment also known as neurogenic OH (nOH); these two conditions have distinct pathophysiologies and affect different patient populations. The prevalence of OH in the elderly varies depending on the study population. link3 In community dwellers, OH prevalence is estimated at 16%, whereas in institutionalized patients, it may be as high as 60%. The prevalence of OH increases exponentially with age, particularly in those 75 years and older. Multiple epidemiological studies have identified OH as a risk factor for all-cause mortality and cardiovascular disease including heart failure and stroke. Real-world data from administrative databases found polypharmacy, multiple co-morbid conditions, and high health-care utilization as common characteristics in OH patients. A comprehensive evaluation of medications associated with OH is discussed with particular emphasis on the use of anti-hypertensive therapy from two large clinical trials on high-intensive versus standard blood pressure management. Finally, we also review the epidemiology of nOH based on the underlying neurodegenerative disorder (either Parkinson's disease or multiple system atrophy), and the presence of co-morbid conditions such as hypertension and cognitive impairment.The study aimed at understanding the biochemical and molecular level modifications in Desmodesmus sp. under lipid inducing stress conditions. The low-temperature (5 °C) incubation and nitrogen starvation reduced the PS II electron transport in microalga with a maximum reduction of 50-57% in ET0/ABS values. The PS II electron transport recovered in UV treated cultures after an initial reduction of 87-93% in ET0/ABS values. A 2.7-4.4 fold increase in ROS and MDA levels was observed under low-temperature incubation, and nitrogen starvation. The UV treatment caused 1.3-2.4 fold higher ROS and MDA levels than control. The low-temperature incubated, nitrogen starved, and UV treated cultures showed 2.4-4 fold higher acc D gene expression. A higher rbc L gene expression was observed under low-temperature stress. The study showed modifications in PS II electron transport, oxidative status, and expression of acc D and rbc L genes under stress conditions.
