Nanomechanical sensors and their arrays have been attracting significant attention for detecting, discriminating and identifying target analytes. The sensing responses can be partially explained by the physical properties of the receptor layers coated on the sensing elements. Analytical solutions of nanomechanical sensing are available for a simple cantilever model including the physical parameters of both a cantilever and a receptor layer. These analytical solutions generally rely on the simple structures, such that the sensing element and the receptor layer are fully attached at their boundary. However, an actual interface in a real system is not always fully attached because of inhomogeneous coatings with low affinity to the sensor surface or partial detachments caused by the exposure to some analytes, especially with high concentration. Here, we study the effects of such macroscopic interfacial structures, including partial attachments/detachments, for static nanomechanical sensing, focusing on a Membrane-type Surface stress Sensor (MSS), through finite element analysis (FEA). We simulate various macroscopic interfacial structures by changing the sizes, numbers and positions of the attachments as well as the elastic properties of receptor layers (e.g., Young's modulus and Poisson's ratio) and evaluate the effects on the sensitivity. It is found that specific interfacial structures lead to efficient sensing responses, providing a guideline for designing the coating films as well as optimizing the interfacial structures for higher sensitivity including surface modification of the substrate.Mutations of the TP53 gene occur in a subset of patients with acute myeloid leukemia (AML) and confer an exceedingly adverse prognosis. However, whether different types of TP53 mutations exert a uniformly poor outcome has not been investigated yet. Proteasome assay Here, we addressed this issue by analyzing data of 1537 patients intensively treated within protocols of the German-Austrian AML study group. We classified TP53 mutations depending on their impact on protein structure and according to the evolutionary action (EAp53) score and the relative fitness score (RFS). In 98/1537 (6.4%) patients, 108 TP53 mutations were detected. While the discrimination depending on the protein structure and the EAp53 score did not show a survival difference, patients with low-risk and high-risk AML-specific RFS showed a different overall survival (OS; median, 12.9 versus 5.5 months, p = 0.017) and event-free survival (EFS; median, 7.3 versus 5.2 months, p = 0.054). In multivariable analyses adjusting for age, gender, white blood cell count, cytogenetic risk, type of AML, and TP53 variant allele frequency, these differences were statistically significant for both OS (HR, 2.14; 95% CI, 1.15-4.0; p = 0.017) and EFS (HR, 1.97; 95% CI, 1.06-3.69; p = 0.033). We conclude that the AML-specific RFS is of prognostic value in patients with TP53-mutated AML and a useful tool for therapeutic decision-making.Thioredoxin fold proteins (TFPs) form a family of diverse proteins involved in thiol/disulfide exchange in cells from all domains of life. Leptospirillum spp. are bioleaching bacteria naturally exposed to extreme conditions like acidic pH and high concentrations of metals that can contribute to the generation of reactive oxygen species (ROS) and consequently the induction of thiol oxidative damage. Bioinformatic studies have predicted 13 genes that encode for TFP proteins in Leptospirillum spp. We analyzed the participation of individual tfp genes from Leptospirillum sp. CF-1 in the response to oxidative conditions. Genomic context analysis predicted the involvement of these genes in the general thiol-reducing system, cofactor biosynthesis, carbon fixation, cytochrome c biogenesis, signal transduction, and pilus and fimbria assembly. All tfp genes identified were transcriptionally active, although they responded differentially to ferric sulfate and diamide stress. Some of these genes confer oxidative protection to a thioredoxin-deficient Escherichia coli strain by restoring the wild-type phenotype under oxidative stress conditions. These findings contribute to our understanding of the diversity and complexity of thiol/disulfide systems, and of adaptations that emerge in acidophilic microorganisms that allow them to thrive in highly oxidative environments. These findings also give new insights into the physiology of these microorganisms during industrial bioleaching operations.Phalaenopsis amabilis, one of the most important plants in the international flower market due to its graceful shape and colorful flowers, is an orchid that undergoes vernalization and requires low-temperature treatment for flowering. There have been few reports on the proteomics of the development of flower buds. In this study, isobaric tags for relative and absolute quantification (iTRAQ) were used to identify 5064 differentially expressed proteins in P. amabilis under low-temperature treatment; of these, 42 were associated with early floral induction, and 18 were verified by mass spectrometry multi-reaction monitoring (MRM). The data are available via ProteomeXchange under identifier PXD013908. Among the proteins associated with the vernalization pathway, PEQU_11434 (glycine-rich RNA-binding protein GRP1A-like) and PEQU_19304 (FT, VRN3 homolog) were verified by MRM, and some other important proteins related to vernalization and photoperiod pathway that were detected by iTRAQ but not successfully verified by MRM, such as PEQU_11045 (UDP-N-acetylglucosamine diphosphorylase), phytochromes A (PEQU_13449, PEQU_35378), B (PEQU_09249), and C (PEQU_41401). Our data revealed a regulation network of the early development of flower buds in P. amabilis under low temperature induction.The aim of the study is to develop a compact, robust and maintenance free gas concentration and humidity monitoring system for industrial use in the field of inert process gases. Our multiparameter gas-monitoring system prototype allows the simultaneous measurement of the fluid physical properties (density, viscosity) and water vapor content (at ppm level) under varying process conditions. This approach is enabled by the combination of functionalized and non-functionalized resonating microcantilevers in a single sensing platform. Density and viscosity measuring performance is evaluated over a wide range of gases, temperatures and pressures with non-functionalized microcantilevers. For the humidity measurement, microporous Y-type zeolite and mesoporous silica MCM48 are evaluated as sensing materials. An easily scalable functionalization method to high-throughput production is herein adopted. Experimental results with functionalized microcantilevers exposed to water vapor (at ppm level) indicate that frequency changes cannot be attributed to a mass effect alone, but also stiffness effects dependent on adsorption of water and working temperature must be considered.
