The goal of this study was to describe patterns of selective serotonin reuptake inhibitor (SSRI) use during pregnancy in a US cohort (2005-2014) of > 1 million commercially insured women using administrative claims. We used international classification of disease (ICD-9) diagnosis and procedure and current procedural terminology codes in the OptumLabs® Data Warehouse to identify deliveries (including losses) among US women aged 15-45 (n = 1,061,023). SSRI dispensings that overlapped with the timing of pregnancy were identified using national drug codes in linked pharmacy claims. Demographic characteristics were imputed based on residential location, census data, and consumer information. We investigated patterns by trimester, agent, and demographic subgroups. A total of 46,087 of women (4.34%) were dispensed SSRIs during the estimated pregnancy period. Sertraline was the most common overall and had the highest initial use after trimester 1, including women who switched from another SSRI, although dispensing for > 1 SSRI during pregnancy was uncommon. Use of vilazodone was rare and had the highest discontinuation after trimester 1, followed by paroxetine. SSRI use was more common among women who were older, White, college-educated, higher income (≥ $100,000), or resided in the Midwest. Paroxetine and dispensings for > 1 SSRI were more common in lower education subgroups. White women had the highest proportion of use in all trimesters of pregnancy, whereas Hispanic women had the lowest. Among commercially insured US women, SSRI use during pregnancy differed by agent and demographics. More research is needed to understand whether these differences are due to symptom reporting, cultural beliefs, and/or physician preferences.The CIAO3 protein operates at a crossroad of the cytosolic iron-sulfur protein assembly (CIA) machinery. Although the functional role of CIAO3 has been recently characterized, a description of its interaction network is still not complete. Literature data suggested that CIAO3 interacts individually with CIA2A and CIAO1 protein, with the latter two interacting each other. However, no experimental data are available yet showing the formation of a possible ternary complex composed by CIAO3, CIAO1, and CIA2A. This work shows, for the first time, via size exclusion chromatography coupled with multiangle light scattering, UV-vis absorption and electron paramagnetic resonance (EPR) spectroscopies, the formation of a stable, [4Fe-4S]-bound, complex, composed by CIAO3 and the hetero-CIA2A-CIAO1 complex. Moreover, site-directed mutagenesis data suggested a structural role for the C-terminal [4Fe-4S] cluster of the CIAO3 protein. These findings can provide solid bases for further investigation of the molecular mechanisms involving these CIA machinery proteins.INTRODUCTION The design of training programs for football players is not straightforward due to intra- and inter-individual variability that leads to different physiological responses under similar training loads. OBJECTIVE To study the association between the external load, defined by variables obtained using electronic performance tracking systems (EPTS), and the urinary metabolome as a surrogate of the metabolic adaptation to training. METHODS Urine metabolic and EPTS data from 80 professional football players collected in an observational longitudinal study were analyzed by ultra-performance liquid chromatography coupled to electrospray ionization quadrupole time-of-flight mass spectrometry and assessed by partial least squares (PLS) regression. RESULTS PLS models identified steroid hormone metabolites, hypoxanthine metabolites, acetylated amino acids, intermediates in phenylalanine metabolism, tyrosine, tryptophan metabolites, and riboflavin among the most relevant variables associated with external load. Metabolic network analysis identified enriched pathways including steroid hormone biosynthesis and metabolism of tyrosine and tryptophan. The ratio of players showing a deviation from the PLS model of adaptation to exercise was higher among those who suffered a muscular lesion compared to those who did not. CONCLUSIONS There was a significant association between the external load and the urinary metabolic profile, with alteration of biochemical pathways associated with long-term adaptation to training. Future studies should focus on the validation of these findings and the development of metabolic models to identify professional football players at risk of developing muscular injuries.While the contribution of industrial parks to the socio-economic aspect has been widely acknowledged over the past two decades in Vietnam, the problem of environmental pollution due to the wastes generated from industrial parks, especially wastewater, has still been an emerging issue, which places a great pressure on the Government. The wastewater generation from industrial parks was ordinarily required to report in all environmental impact assessment (EIA) reports as a base for the construction of on-site wastewater treatment plants within the industrial park. In Vietnam, this data was, however, often higher than the actual generated number due to inaccurate technical guidelines on predicting wastewater generation and many different methods to be applied. This study aimed to evaluate different approaches used to calculate the effluent in the industrial parks. The results showed that all of the four approaches (i.e., previous operation-based, water-based, wastewater-based, or experience-based methods) rendered significant gaps between predicted and actual values. None can be acceptable for use at present. A revision of technical guideline should be conducted to provide more detailed instruction for the better prediction. This shall minimize the investment capital and increase the efficiency of industrial parks in pollution control.Early detection and treatment of cardiovascular disease by identifying markers is important to improve the health of our citizens and simultaneously reduce the cost of health care. Therefore, there is an urgent need for early detection and treatment of asymptomatic cardiovascular disease. Herein, we have developed highly sensitive vertical flow immunokit (VFIK) for the detection of C-reactive protein (CRP), comprising an antibody/citrate conjugated gold nanoparticle in a fixed orientation surface. Our CRP test kit has been optimized with various parameters such as buffer composition, the quantity of captured antibody and sensitivity of the assay. Prominently, our developed CRP test kit is highly sensitive and rapid point of care test (POCT) which is capable to detect CRP with minimal volume (50-60 μl) and time (1-2 min) with diminutive detection up to 10 ng/ml. The reliability of our kit was evaluated and validated with spiked and clinical whole blood samples. The results demonstrated that our developed vertical flow-based point-of-care Immunokit may serve fruitful and semi-quantitative routine diagnostic for the early detection of cardiac disease.Individuals of the annual halophyte Atriplex centralasiatica produce three kinds of diaspores that differ in dispersal, dormancy/germination response and type of seed bank formed, which likely is a bet-hedging strategy in the rainfall-unpredictable environment on the semi-arid, saline Ordos Plateau in Inner Mongolia, China. Seasonal fluctuations in environmental conditions provide germination cues for the establishment of seedlings at the right time and place to ensure plant survival and population regeneration. Diaspore heteromorphism is a phenomenon in which diaspores with stark qualitative differences in morphology and ecology are produced by the same maternal plant. Germination responses and dispersal times of the annual halophyte Atriplex centralasiatica were examined to determine the role of diaspore heteromorphism in its adaptation to salt desert conditions. A. centralasiatica is a tumbleweed that produces three types of diaspores that differ in morphology and ecophysiology. The relative potential dispersal ability and intensity of dormancy of the three diaspore types was type A (fan-shaped diaspores with yellow fruits) 90.0% remained in the soil seedbank 2 years after dispersal, respectively. The dormancy, dispersal and salt tolerance of type B diaspores were intermediate between those of A and C. Type A exhibited low dispersal-nondormancy, type B exhibited intermediate dispersal-intermediate dormancy and type C exhibited high dispersal ability-high dormancy. In the unpredictable salt desert habitat, the functional differences in germination and dispersal of the three diaspores act as a bet-hedging mechanism and ensure population establishment in different years by spreading germination over time and space.Primary sclerosing cholangitis (PSC) is a progressive liver disease, histologically characterized by inflammation and fibrosis of the bile ducts, and clinically leading to multi-focal biliary strictures and with time cirrhosis and liver failure. Patients bear a significant risk of cholangiocarcinoma and colorectal cancer, and frequently have concomitant inflammatory bowel disease and autoimmune disease manifestations. To date, no medical therapy has proven significant impact on clinical outcomes and most patients ultimately need liver transplantation. Several treatment strategies have failed in the past and whilst prescription of ursodeoxycholic acid (UDCA) prevails, controversy regarding benefits remains. Lack of statistical power, slow and variable disease progression, lack of surrogate biomarkers for disease severity and other challenges in trial design serve as critical obstacles in the development of effective therapy. Despite its relatively characteristic histology, it may sometimes mimic other lesions. Here, we report two female non-smoker patients diagnosed with low-grade mucoepidermoid carcinoma with oncocytic epithelium and prominent lymphoid (Warthin-like) stroma and with molecularly confirmed MAML2 rearrangement. In addition, we screened a consecutive series of 114 Warthin tumour cases by means of MAML2 break apart fluorescence in situ hybridization to assess its value in differential diagnosis. MAML2 rearrangement was detected in both mucoepidermoid carcinoma cases, while all Warthin tumours were negative. Taking into account the literature data, Warthin-like mucoepidermoid carcinomas are more frequently observed in women, while a slight male predominance and smoking history are typical for Warthin tumour. In addition, the patients with Warthin-like mucoepidermoid carcinoma were significantly younger than those with Warthin tumour. To conclude, Warthin-like mucoepidermoid carcinoma may usually be suspected based on histology, while the diagnosis can be confirmed by means of molecular assays such as FISH.
