A total of 68 patients with active SLE treated in our hospital between March 2015 and January 2018 were enrolled into the active SLE group, and they were divided into three groups according to the mild, moderate, and heavy active periods, and also divided into two groups according to the positive and negative anti dsDNA. A total of 60 healthy individuals in the same period were included in the control group (con group). ARA014418 The levels of serum IL-6, IL-10, and TNF-α in all participants were detected via an enzyme-linked immunosorbent assay (ELISA), and the correlation of these values with SLE activity was analyzed. The independent prognostic factors were analyzed through multivariate logistic regression. It was found that the levels of serum IL-6, IL-10, and TNF-α in the SLE groups were all higher than those in the control group; the levels of the inflammatory markers in the severe active SLE group were higher than those in the mild and moderate active SLE groups, and the levels in the moderate active SLE group were higher than those in the mild active SLE group. Additionally, the anti dsDNA positive group showed much higher levels of these than the anti dsDNA negative group. Pearson correlation analysis revealed a positive correlation between anti dsDNA antibody and IL-6, IL-10, and TNF-α levels. The multivariate logistic regression results, the mean course of disease and IL-10 were independent prognostic factors of SLE. The abnormal secretion of peripheral blood cytokines in SLE patients can affect the prognosis of the disease. Monitoring serum cytokines is helpful to understand the activity and prognosis of patients with lupus and guide clinical treatment.

This study aimed to explore the effect of COX-2 selective inhibitor (celecoxib) on adenomyosis and its mechanism.

By establishing a mouse model of adenomyosis and using celecoxib to treat adenomyosis, newly born female mice were randomly divided into a control group, adenomyosis model group, and celecoxib group. Hematoxylin-eosin (H&E) staining was used to observe the depth of endometrial infiltration of mouse adenomyosis. RT-PCR (reverse transcription PCR) and western blot were used to detect the expression of Cyclooxygenase-2 (COX-2), Vascular growth factor (VEGF), Nerve growth factor (NGF), and Corticotropin-releasing hormone (CRH) mRNA and protein in mice before and after celecoxib treatment.

After treatment with celecoxib, the depth of endometrial infiltration of mouse adenomyosis was reduced. COX-2 and VEGF decreased significantly after celecoxib inhibited expression of COX-2 (P<0.001), but there was no significant difference in the expression of NGF or CRH (P>0.05).

This study indicated that COX-2 may be an important factor related to the pathogenesis of adenomyosis, and it may become an important molecular target for the treatment of adenomyosis.

This study aimed at exploring the diagnostic values and pathological conditions of serum HGF and CA199 in endometriosis (EMT).

From May 2017 to January 2019, 86 patients with EMT, who were admitted to our hospital, were grouped as Group B, whereas 78 healthy women undergoing physical examinations during the same period were grouped as Group A. Serum HGF and CA199 levels in both groups were detected to analyze their diagnostic values for EMT and their relationships with clinical data.

Compared with those in Group A, both serum HGF and CA199 expression elevated significantly in Group B (P<0.05). Their expression was correlated with the age, course of disease, number of nodules and infiltration depth of the patients (P<0.05). Besides, their expression was remarkably higher in patients with stages III ± IV than that in patients with stages I ± II (P<0.05). According to the Kaplan-Meier curves for tumor-free survival, the high expression of HGF and CA199 was related to the poor prognosis of the patients (P<0.05). According to the COX regression analysis for risk factors, the number of nodules, infiltration depth, American Fertility Society (AFS) staging, HGF and CA199 were the independent risk factors of EMT recurrence (P<0.05).

HGF and CA199 increase in patients with EMT, and can be used as the biomarkers for diagnosing the disease and predicting its prognosis.

This study aims to determine the role of SIX1 in endometrial cancer (EC). Methods In this study, we analyzed the SIX1 expression profile and the correlation with the corresponding clinical characteristics of EC samples from the Cancer Genome Atlas (TCGA), Gene Expression Omnibus (GEO) and Clinical Proteomic Tumor Analysis Consortium (CPTAC) databases. Wilcoxon signed-rank test was applied to analyze the difference between tumor group and control group. The potential biological processes or signaling pathways related to SIX1 activity in EC was also assessed. Results The results showed that SIX1 was overexpressed in EC tissues compared to normal tissues (P=2.029e-15, P=6.25e-6). The SIX1 level was correlated with tumor grade (P=2.91e-4), peritoneal cytology (P=0.005), and the subsequent tumor surgery (P=1.169e-4). SIX1 expression was negatively associated with overall survival rate (P=4.241e-4, P=0.000241) and served as an independent factor that affected EC overall survival rate (P=0.005063), similar to other factors such as age, Figo stage, and tumor (T) stage. SIX1 participates in cancer pathogenesis through gene regulation that involves PI3K/AKT/MTOR signaling, mitotic spindle, G2M checkpoint, E2F targets, NOTCH signaling, glycolysis, cholesterol homeostasis, DNA repair and early estrogen response. Conclusions Our data demonstrate that SIX1 is overexpressed in EC and associated with adverse clinicopathological outcomes, which can function as an independent factor for EC prognosis.

To analyze the efficacy of sequential treatment with high-flow nasal cannula (HFNC) in chronic obstructive pulmonary disease (COPD) concomitant with respiratory failure.

A total of 100 COPD patients concomitant with respiratory failure requiring invasive mechanical ventilation from June 2019 to May 2020 in our hospital were enrolled and then divided into two groups according to the random number table, with 50 in each group. Pulmonary infection control window (PIC) was used as a switching point for sequential ventilation. The control group (CNG) received non-invasive positive pressure ventilation (NIPPV), while the study group (SG) underwent HFNC. The efficacy, complications and 48 h reintubation rate of the two groups were statistically analyzed. The respiratory parameters, diaphragmatic parameters, diaphragmatic excursion during quiet breathing (DEq), diaphragmatic rapid shallow breathing index (D-RSBI), COPD score (CAT), 6-min walk test (6 MWT) score (Borg), General Comfort Questionnaire (GCQ), sputum espiratory function and diaphragmatic function, reduce dyspnea and fatigue, reduce sputum viscosity, regulate serum factors, and make patients enjoy higher comfort.

To explore the effectiveness of a targeted nursing model for patients undergoing thyroid surgery, and to analyse the influence of intervention on the negative emotions of patients.

Eighty patients who received thyroid surgery in our hospital were enrolled and divided into a study group (n=40, given targeted nursing) and a control group (n=40, given routine surgical nursing) according to the difference of intervention measures. The postoperative bed-leaving time, hospitalization time, medical expenses, drainage tube indwelling time and incidence of postoperative adverse reactions were compared between the two groups. The degree of pain, anxiety and depression was compared between the two groups at 1 d, 3 d, 5 d, and 7 d after surgery. The scales of voice handicap index (VHI-10) and standard swallowing assessment (SSA) were used to evaluate voice quality and swallowing function in the two groups.

The postoperative bed-leaving time, hospitalization time, medical expenses, drainage tube indwelling time and ealthy emotions and pain of patients, and help improve their voice quality and swallowing function, which also has a positive effect on reducing postoperative complications. Therefore, it is worthy of clinical popularization and application.

To investigate the effect of dose-painted intensity-modulated radiotherapy (DP-IMRT) combined with chemotherapy on stage IIIB cervical cancer.

A total of 107 stage IIIB cervical cancer patients were treated with DP-IMRT combined with chemotherapy. The planning target volume (PTV) was divided into regions with different prescribed absorbed doses (so-called PTV-subvolume [PTVsv]) PTVsv

(the part of the PTV that overlaps with the organ at risk (OAR)) received 39.6-45 Gy, 1.8 Gy/fraction (fx); and PTVsv

(the part of the PTV that does not overlap with the OAR) received 44-50 Gy, 2.0 Gy/fx. The lymph nodes were simultaneously boosted; lymph nodes with a short axis dimension <1 cm received 50-55 Gy, 2.0-2.4 Gy/fx, while nodes with a short axis dimension >1 cm received 55-66 Gy, 2.2-2.6 Gy/fx. External radiotherapy was followed by intracavitary brachytherapy. Patients were followed up regularly to collect the survival information.

Five years after therapy, the overall survival rate and progression-free survival rate were 61.0% and 55.0%, respectively. The cumulative rates for total grade 3 or higher chronic gastrointestinal or genitourinary toxicity were 4.67% and 1.9% respectively.

Without compromising the primary PTV, DP-IMRT achieved good outcomes for stage IIIB cervical cancer patients with a favorable gastrointestinal toxicity profile.
