Cardiovascular disease is a leading cause of death worldwide, and thus there remains great interest in regenerative approaches to treat heart failure. In the past 20 years, the field of heart regeneration has entered a renaissance period with remarkable progress in the understanding of endogenous heart regeneration, stem cell differentiation for exogenous cell therapy, and cell-delivery methods. In this review, we highlight how this new understanding can lead to viable strategies for human therapy. For the near term, drugs, electrical and mechanical devices, and heart transplantation will remain mainstays of cardiac therapies, but eventually regenerative therapies based on fundamental regenerative biology may offer more permanent solutions for patients with heart failure.The endometrium is a dynamic, multicellular tissue that is constantly remodeled in response to regulating hormones. In a recent issue of Nature Genetics, Garcia-Alonso et al. delineate the unique genetic signatures of the endometrial cells. Their findings validate a three-dimensional epithelial organoid system for modeling endometrial glands ex utero.How the physical nature of the cytoplasm shapes cellular processes has been increasingly studied in recent years. Work in this issue demonstrates that cytoplasmic viscosity affects the dynamic instability of microtubules. Surprisingly, both polymerization and depolymerization slow with increasing viscosity. This raises interesting questions for future work.In this issue of Developmental Cell, Hernández-Coronado et al. present genetic and pharmacological evidence that reveals the central role of plant glutamate receptor-like proteins (GLRs) in the tradeoff between wounding-triggered regeneration and defense, offering new strategies to improve plant regeneration.The capacity for long-distance axon regeneration and functional recovery after spinal cord injury in the adult has long been thought to be a unique feature of certain non-mammalian vertebrates. However, in this issue of Developmental Cell, Nogueira-Rodrigues et al. report an astonishingly high regenerative ability in the spiny mouse.The presence of intratumoral tertiary lymphoid structures (TLS) is associated with positive clinical outcomes and responses to immunotherapy in cancer. Here, we used spatial transcriptomics to examine the nature of B cell responses within TLS in renal cell carcinoma (RCC). B cells were enriched in TLS, and therein, we could identify all B cell maturation stages toward plasma cell (PC) formation. B cell repertoire analysis revealed clonal diversification, selection, expansion in TLS, and the presence of fully mature clonotypes at distance. In TLS+ tumors, IgG- and IgA-producing PCs disseminated into the tumor beds along fibroblastic tracks. TLS+ tumors exhibited high frequencies of IgG-producing PCs and IgG-stained and apoptotic malignant cells, suggestive of anti-tumor effector activity. Therapeutic responses and progression-free survival correlated with IgG-stained tumor cells in RCC patients treated with immune checkpoint inhibitors. Thus, intratumoral TLS sustains B cell maturation and antibody production that is associated with response to immunotherapy, potentially via direct anti-tumor effects.Individuals are faster at detecting threatening stimuli than neutral stimuli. While generally considered a rapid bottom-up response, this threat superiority effect is also modulated by top-down mechanisms known to rely on the dorsolateral prefrontal cortex (DLPFC). What remains unclear is whether the response is modulated only at later stages of processing, or whether rapid attention to threat itself is controlled in a top-down manner. To test this, we used repetitive transcranial magnetic stimulation (rTMS) to inhibit activity in the DLPFC, and measured EEG to index the immediate neural response to threat. Twenty participants attended two sessions where they performed a visual search task with threatening or neutral targets. Prior to this, they received 15 min of 1 Hz inhibitory or sham rTMS targeting the right DLPFC. Since autism and epilepsy are common neurodevelopmental disorders with high coincidence, we tested whether early-life chronic subconvulsive activity compared to convulsive activity alters handling, spatial memory, lateralization, coping strategy and the seizure threshold in a sex-dependent manner. A hypothesis is that convulsive seizures may alter sex differences induced by subconvulsive (SC) activity. Serial subconvulsive doses of kainic acid (KA) were administered postnatally (0.25-1 mg/kg) for 15 days to induce the chronic subconvulsive phenotype (SC group). Age-matched controls and a subset of SC pups were exposed to a convulsive dose of KA (KA and SC + KA groups; 7.5 mg/kg) or flurothyl vapors. In our open handling test, controls and the ASD groups escaped to a similar degree whereas after convulsive seizures, the pups exhibited freezing beseizures than control females which disappeared after the sub-convulsive pre-treatment. Data suggest that behavioral manifestations are more readily detectable between males and females when low levels of hyperexcitation are present chronically in early postnatal development but diminished after tonic-clonic convulsions persist. Therefore, therapeutic interventions may benefit patients if initiated upon the initial onset of sex-related autistic pathologies, particularly in males, which may reduce subsequent vulnerability to seizures.

To examine the relationships among depression and anxiety symptomatology, stigma of mental illness, levels of social support, and select demographics among hospitalized women in the postpartum period.

Descriptive, cross-sectional, correlational.

A convenience sample of 105 English-speaking and Spanish-speaking women was recruited and enrolled from a 208-bed free-standing Southern California women's community hospital postpartum unit serving a diverse community.

Data were collected on potential covariates including participants' characteristics, depression and anxiety symptomatology, social support, and stigma.

Sixteen participants were classified as high risk for depression or anxiety based on the Edinburgh Postnatal Depression Scale and Generalized Anxiety Disorder-7. Anxiety was significantly associated with all stigma subscales; the strongest association was with Internal Stigma (r= .46, p< .001, moderate effect), followed by Disclosure Stigma (r= .36, p< .001, moderate effect) and External and anxiety and the relevance of stigma and social support in aggravating or mitigating symptomatology. There is a need for comprehensive standardized screening to ensure the identification of and referral to treatment for women at risk.Trichinellosis is an important meat-borne zoonotic parasitic disease caused by ingesting raw or semi-cooked meat of pigs and other animals infected with Trichinella sp. muscle larvae. Epidemiological data on human and animal Trichinella sp. infection in the People's Republic of China (PRC) during 2009-2020 were analyzed in this review. The results showed that the endemic foci of human trichinellosis are principally localized in southwestern areas, and eight outbreaks covering 479 cases and 2 deaths were reported. Pork is still the primary source of trichinellosis outbreaks. Seven out of 8 outbreaks (87.50%) were caused by ingesting raw or semi-cooked pork. The seroprevalence of swine anti-Trichinella IgG ranged from 0 to 42.11% in 11 provinces/autonomous regions (P/As), and swine Trichinella infection was detected in six P/A slaughterhouses. The Trichinella-infected pigs came from small backyard farms and outdoor free-ranging pigs in western and southwestern PRC. To prevent trichinellosis, the traditional pig-rearing mode should be improved, more industrialized pig farms should be developed, all pigs should be raised in piggeries under controlled management conditions, and mandatory inspection of Trichinella sp. in slaughtered pigs should be implemented in rural areas of western and southwestern PRC. A One Health approach with participation from governments, public health officials, and medical and veterinary practitioners is vital for controlling zoonotic foodborne trichinellosis.Two major environmental challenges of our time are responding to climate change and reversing biodiversity decline. Interventions that simultaneously tackle both challenges are highly desirable. To date, most studies aiming to find synergistic interventions for these two challenges have focused on protecting or restoring vegetation and soils but overlooked how conservation or restoration of large wild animals might influence the climate mitigation and adaptation potential of ecosystems. However, interactions between large animal conservation and climate change goals may not always be positive. Here, we review wildlife conservation and climate change mitigation in terrestrial and marine ecosystems. We elucidate general principles about the biome types where, and mechanisms by which, positive synergies and negative trade-offs between wildlife conservation and climate change mitigation are likely. We find that large animals have the greatest potential to facilitate climate change mitigation at a global scale via three mechanisms changes in fire regime, especially in previously low-flammability biomes with a new or intensifying fire regime, such as mesic grasslands or warm temperate woodlands; changes in terrestrial albedo, particularly where there is potential to shift from closed canopy to open canopy systems at higher latitudes; and increases in vegetation and soil carbon stocks, especially through a shift towards below-ground carbon pools in temperate, tropical and sub-tropical grassland ecosystems. Large animals also contribute to ecosystem adaptation to climate change by promoting complexity of trophic webs, increasing habitat heterogeneity, enhancing plant dispersal, increasing resistance to abrupt ecosystem change and through microclimate modification.Wolbachia is a genus of globally distributed endosymbiotic bacteria found in thousands of species of arthropods and nematodes.
