Materials extracted from atherosclerotic arteries can disclose data about the molecular pathology of cardiovascular disease, but obtaining such samples is complex and requires invasive surgery. To overcome this barrier, this study investigated whether angioplasty balloons inflated during standard percutaneous coronary interventions retain proteins from treated (dilated) atherosclerotic lesions and whether proteomic analysis of this material could provide data on lesion protein profiles and distinguish between patients with stable and unstable coronary artery disease.
Patients with ST-segment-elevation myocardial infarction and stable angina pectoris were subjected to routine percutaneous coronary interventions. All angioplasty balloons inflated in a coronary artery were collected. Proteins retained on the balloons were extracted and analyzed using shotgun proteomic analysis.
Proteomics identified and quantified 1365 unique proteins captured on percutaneous coronary intervention balloons. Control balloonoronary artery lesions adhere to angioplasty balloons and constitute a source of material for proteomic analysis. This approach can identify proteins and processes occurring in unstable coronary atherosclerotic lesions and suggest novel therapeutic approaches.The aim of this study was to assess the agreement between anti-Toxoplasma gondii IgG antibody detection in serum and filter paper (FP) blood spots using the indirect immunofluorescence antibody assay (IFA) and to evaluate the potential impact of the packed cell volume (PCV) on antibody detection in FPs. A pair of a serum and an FP sample was collected from 96 sows at various farms in Greece, with previously identified high seropositivity and/or risk factors associated with high seropositivity against T. gondii. The PCV value was determined using the microhematocrit method. IFA was used for the detection of antibodies against T. gondii. T. gondii-specific antibodies were detected in 45.8% serum samples and 41.6% FP samples showing almost perfect agreement. Detection in FP samples presented high sensitivity (87.1-92.8%) and excellent specificity (100%) when compared with detection in serum, regardless of the PCV values. The findings of this study support the reliability of FPs for the evaluation of the serological status of swine against T. gondii. FPs could be a good alternative sample type compared with serum for large-scale epidemiological studies.Background Irisin has been recently reported to provide beneficial effects in obesity and diabetes. Elevation of high-sensitivity C-reactive protein (hs-CRP) reflects the inflammatory state that has been shown to play a key role in obesity and its comorbidities. Objective Our study aimed to compare the changes of circulating irisin levels in an obese diabetic population who underwent laparoscopic Roux-en-Y gastric bypass (LRYGB) versus a control-matched population who underwent usual medical care plus diabetes support education (DSE) program. In addition, we aimed to explore the association between circulating irisin and hs-CRP levels after the interventions. Methods In a prospective controlled trial, we studied 58 obese individuals with type 2 diabetes before and 12 months after intervention. Twenty-nine subjects underwent LRYGB and 29 subjects received DSE. Results At 12-month follow-up, compared with the DSE group, patients who underwent LRYGB lost more weight (LRYGB; -33.4 ± 11.2, and DSE; 0.2 ± 4.9 kg; P less then .001), fat mass (P less then .001), and fat-free mass (P less then .05). Circulating irisin (P less then .05) and hs-CRP level (P less then .05) were also significantly lower. Within the LRYGB group, the reduction of irisin level was positively associated with the changes of hs-CRP levels (r = 0.39, P less then .05). Conclusions To the best of our knowledge, this is the first study showing that LRYGB significantly reduces circulating irisin levels compared with usual medical care and DSE, in an obese diabetic population. After LRYGB, the irisin reduction significantly correlates with the reduction of hs-CRP. The elevation of circulating irisin levels suggests irisin resistance in the obese state and its decrease after LRYGB might reflect the resolution of irisin resistance. Future investigations are needed to confirm and explore the mechanisms of irisin resistance in obesity, its resolution after LRYGB, and the pathophysiological significance.Shigella flexneri, a common Gram-negative foodborne pathogen, is widely distributed in fresh-cut fruits and vegetables, unpasteurized milk, and food processing environments. The aims of this study were to evaluate the antibacterial effects of 405-nm light-emitting diode (LED) treatment on S. flexneri and to investigate the possible mechanism. The results showed that LED irradiation (360 min) reduced the number of S. flexneri in phosphate-buffered saline by 3.29 log colony-forming unit (CFU)/mL (initial bacterial count 6.81 log CFU/mL). The cells in reconstituted infant formula, cells on fresh-cut carrot slices, and biofilm-associated cells on stainless steel surfaces were reduced by 1.83 log CFU/mL, 7.00 log CFU/cm2, and 4.35 log CFU/cm2 following LED treatment for 360, 120, and 120 min, respectively. LED treatment damaged both DNA and cell wall of S. flexneri and changed cell morphology and cell membrane permeability. In addition, LED treatment decreased total cell protein concentration of S. flexneri. These results indicated that 405-nm LED treatment effectively controlled S. flexneri contamination of foods and food contact surfaces and that the bacterial inactivation may be the result of damage to multiple cellular components. These findings highlight the potential of LED technology in controlling S. flexneri during food processing, storage, and preparation.Knowledge and perceptions of the population on services in case of need for long-term care A standardized telephone survey Abstract. Aim The study investigates the care of people in need for care, the level of knowledge about and stress factors in the assumption of care from the point of view of informal caregivers and non-caregivers. Methods A cross-sectional study was conducted using standardized Computer Assisted Telephone Interviews (CATI) with persons aged 18 and older in Saxony. Results Care provided by an outpatient care service or by family caregivers was rated significantly better by 259 informal caregivers compared with 342 non-caregivers among 601 participants (N = 601). Conclusions In individual areas of the queried counseling services and relief options, the response behavior of family caregivers differed significantly from the other participants. It can be assumed that family caregivers have a slight information advantage of family caregivers in the use of support services.
Strokes in the working-age population represent a relevant share of ischemic strokes and re-employment is a major factor for well-being in these patients. Income differences by sex have been suspected a barrier for women in returning to paid work following ischemic stroke. We aim to identify predictors of (not) returning to paid work in patients with large vessel occlusion treated with mechanical thrombectomy (MT) to identify potential areas of targeted vocational rehabilitation.
From 6635 patients enrolled in the German Stroke Registry Endovascular Treatment between 2015 and 2019, data of 606 patients of the working population who survived large vessel occlusion at least 90 days past MT were compared based on employment status at day 90 follow-up. Univariate analysis, multiple logistic regression and analyses of area under the curve were performed to identify predictors of re-employment.
We report 35.6% of patients being re-employed 3 months following MT (median age 54.0 years; 36.1% of men, 34.5% of waracteristic curve 0.887 versus 0.835,
<0.001).
There is more to re-employment after MT than functional outcome alone. In particular, attention should be paid to possible systemic barriers deterring women from resuming paid work.

The optimal endovascular strategy for reperfusing distal medium-vessel occlusions (DMVO) remains unknown. This study evaluates angiographic and clinical outcomes of thrombectomy strategies in DMVO stroke of the posterior circulation.
TOPMOST (Treatment for Primary Medium Vessel Occlusion Stroke) is an international, retrospective, multicenter, observational registry of patients treated for DMVO between January 2014 and June 2020. This study characterized CBD pharmacokinetics in healthy adults using a population pharmacokinetic approach, informing drug development of oral-based dose forms of CBD. Materials and Methods CBD concentration-time data were obtained from a phase I, randomized, open-label, four-way crossover study (n=12) and modeled using Phoenix NLME. Monte Carlo simulations were conducted to estimate CBD exposure with chronic dosing as intended for clinical use (50 mg b.i.d.). Results A three-compartment pharmacokinetic model with a chain of absorption transit compartments and first-order elimination most adequately described CBD pharmacokinetics. Substantial variability in population pharmacokinetic parameters was identified (up to 60%CV), which could not be accounted for by any covariates.
