luation and treatment of patients, and are worthy of widespread clinical application. To provides a reference basis for the apoptosis of breast cancer (BC) cells and the carcinogenesis of BC, the effects of 7, 12-dimethylbenz (a) anthracene (DMBA) on apoptosis regulators FasL and B-cell lymphoma-2 (Bcl-2) were investigated. In this study, 62 female C57BL/6 mice aged from 4 to 6 weeks were randomly divided into control group (CG) and test group (TG), with 31 mice in each group. The TG was given DMBA solution by gavage at a dose of 50 mg/kg, and the CG was given normal saline of equal volume. On the second day after the experiment, all the mice were killed by cervical dislocation. The morphology of the mammary gland was observed by hematoxylin-eosin (HE) staining, and the differences of FasL and Bcl-2 protein expression (PE) were detected by immunohistochemistry. The mRNA expression levels of FasL and Bcl-2 were detected by quantitative real-time PCR (qPCR). Breast cell apoptosis status of mice in the two groups was detected by the terminal deoxyribonucleotidyl transferase (TdT)-mediated biotin-in the CG was (2.93±0.28) %, and there was an extremely obvious difference (P less then 0.01). It indicated that DMBA inhibited the apoptosis of BC cells by regulating the up-regulation of FasL and Bcl-2 expression.Early preeclampsia is a pregnancy-specific clinical disease, its pathogenesis is not clear, and clinical treatment is limited. Therefore, it is particularly important to study the pathogenesis of early preeclampsia, so as to provide evidence for the diagnosis and effective treatment of early preeclampsia. This research was carried out to investigate the expression levels and clinical significance of mir-145-5p and stagflation specific gene 6 (Gas6) in serum of women with early preeclampsia. For this purpose, 142 patients with preeclampsia were divided into the early-onset group (n=78) and the late-onset group (n=64) according to the onset time. Meanwhile, 70 normal pregnant women were selected as the control group. The levels of serum Gas6, mir-145-5p and inflammatory factors were detected. Logistic regression analysis showed that the expression levels of serum gas-6 and serum mir-145-5p were valuable in the diagnosis of early preeclampsia. ROC curve was established to evaluate the diagnostic efficacy of serum Gas6 level in early-onset preeclampsia. Its sensitivity and specificity were 91.23% and 64.2%, respectively, and the cutting value was 255.71pg/mL. The serum Gas6 level was negatively correlated with systolic blood pressure (r=-0.349, P less then 0.05) and positively correlated with platelet count (r=0.391, P less then 0.05). Compared with the control group, serum levels of mir-145-5p and TNF-a in women with early-onset preeclampsia were negatively correlated (r=-0.460~0.622, P less then 0.05). Conclusion the serum level of Gas6 was reduced in patients with early-onset preeclampsia, and TNF-a inhibited the invasion of SVneo cells by upregulating mir-145-5p.In this study, rainbow trout were fed for a total of 90 days with different feed mixes containing 0.00%, 1.00%, 1.50% and 2.00% garlic (Allium sativum). The effect of garlic on the growth performance, body composition and fatty acid profile of rainbow trout was researched. The starting weight of fish in the experimental group was approximately 64.12±0.29 gr, which increased to 234.12±3.54gr, 246.31±4.41gr, 258.74±4.42gr, and 268.79±4.79gr, while the difference in growth between the groups was observed to be statistically significant (p less then 0.05). At the end of the feeding period, the specific growth rate (SGR), the economic conversion ratio (ECR), the feed conversion ratio (FCR), the economic profit index (EPI) and the protein efficiency ratio (PER) were checked. In this study, the nutritional composition and fatty acid profiles of fish after 90 days of feeding were examined. As a result of this study, the supplementation of garlic to the diet given to rainbow trout caused an increase in their protein ratio and a decrease in their moisture content and fat ratio. Additionally, it led to a decrease in fatty acid and monounsaturated fatty acid levels and an increase in polyunsaturated fatty acids. The DNA methyltransferase (DNMT) family constitutes a conserved set of DNA-modifying enzymes which have essential functions in the modulation of epigenetics. The fundamental role of epigenetic changes in carcinogenesis and metastasis is increasingly being appreciated. DNMTs (DNMT1, DNMT3A and DNMT3B) have been shown to drive metastasis. Epigenetic machinery is installed at the target sites for the regulation of a wide variety of genes. Moreover, microRNAs, long non-coding RNAs and circular RNAs also shape the epigenetic landscape during metastasis. In this review, we have provided a snapshot of the quintessential role of DNMTs in metastasis. SP-2577 We also summarize how lncRNAs and circRNAs play roles in the epigenetic regulation of a myriad of genes.LncRNA prostate cancer-associated transcript 1 (PCAT1) is a well-known oncogene, but the mechanisms of exosomes PCAT1 in colorectal cancer (CRC) remain largely unknown. Thus, the mechanisms of exosomes lncRNA PCAT1 were investigated. The expressions of exosomes lncRNA PCAT1 in tissues from stage 0-I and stage II-III CRC patients, and intestinal epithelial cell line FHC and two CRC cell lines, HT29 and HCT8 were measured by real-time quantitative PCR. The effects of lncRNA PCAT1 on adhesion and invasion of two CRC cell lines were investigated by cell-matrix adhesion and transwell assays. In addition, the target of PCAT1 (ZNF217) was validated using an RNA immune precipitation assay. Finally, the protein levels of MTA2, MTA3, SNAI1, and E-cadherin in normal participants, stage 0-I and stage II-III CRC patients, as well as two cell lines with stable ZNF217 knockdown were investigated by western blotting. The plasma exosomal lncRNA PCAT1 was found to be significantly increased in the CRC tissues and cell lines. In addition, lncRNA PCAT1 knockdown significantly inhibited the adhesion and invasion of HT29 and HCT8 cells. RIP assay results showed lncRNA PCAT1 could target ZNF217, and downregulation of lncRNA PCAT1 could decrease the protein expressions of ZNF217 in two CRC cells lines. Moreover, ZNF217 knockdown significantly decreased MTA2, MTA3, and SNAI1 expressions, but increased E-cadherin expressions in both CRC cells lines. Exosomal lncRNA PCAT1 can promote the adhesion and invasion of CRC cells, and PCAT1 overexpression may lead to ZNF217 upregulation that regulates EMT-related MTA2/MTA3/Snai1/E-cadherin signaling. It has been recognized that Citrus reticulata and Pinellia ternata have a good therapeutic effect on NSCLC. However, the potential mechanism of C. reticulata and P. ternata in the treatment of NSCLC based on network pharmacology analysis is not clear. The "Drug-Component-Target-Disease" network was constructed by Cytoscape, and the protein interaction (PPI) network was constructed by STRING. Our study indicated that 18 active ingredients of C. reticulata and P. Ternata were screened from the TCMSP database, and 56 target genes of C. reticulata and P. Ternata for the treatment of NSCLC were identified, and we constructed the "Drug-Component-Target-Disease" network. In this study, we screened 56 PPI core genes to establish a PPI network. We concluded that the network pharmacology mechanism of the effect of C. reticulata and P. Ternata on NSCLC may be closely related to the protein expressed by TP53, ESR1, FOS, NCOA3 and MAPK8, and these may play the therapeutic roles by regulating the IL-17 signaling pathway, antigen processing and presentation, microRNAs in cancer and endocrine resistance.Ischemia-reperfusion injury (IRI) is associated with ischemic heart disease (IHD) which leads to patients a poor progression. According to Pubmed Datasets, we analyzed different gene and mRNA expressions in IHD patients with IRI. The relevant mRNA expression detected in H9C2 cells undergo hypoxia and reoxygenation, we selected and structured miR-525-5p gene mutation H9C2 cells, the results performed miR-525-5p mutated restored H9C2 metabolism of mitochondria which detected by relevant genes and proteins. At the same time, miR-525-5p silence resisted hypoxia and reoxygenation induced H9C2 cells apoptosis. All the results indicated miR-525-5p maybe protect H9C2 cells without hypoxia and reoxygenation induced injury through regulating the mitochondria metabolism.Balloon atrial septostomy (BAS) remains an essential palliative interventional procedure in neonates with transposition of great arteries (TGA) and restrictive inter-circulatory mixing (low saturation) that does not respond to prostaglandin E1 therapy for maintaining ductal patency. The short- and intermediate-term outcomes of BAS were evaluated in patients with TGA, and the association of C677T polymorphism in the MTHFR gene was considered on these patients at Solemani Center for Heart Diseases, Dr. Jamal Ahmad Rasheed Pediatric Teaching Hospital, and Shar Hospital in Sulaymaniyah city. For this purpose, a retrospective study was used to evaluate short- and intermediate-term outcomes of BAS in 65 consecutive neonates who have been diagnosed with TGA and restrictive inter-circulatory mixing using echocardiography in three leading hospitals in Sulaymaniyah city, during the study period from June 2011 until January 2020. Also, for genetic polymorphism assessments, in addition to 65 patients, 65 healthy children also showed a significant difference between the two groups in genotypes and alleles (P less then 0.05). This result confirms the relationship between C667T polymorphism of the MTHFR gene and transposition of great arteries disease. BAS is an effective palliative procedure in neonates with dextro-transposition of the great arteries (d-TGA) and can be safely performed at the bedside using echocardiography. It has an excellent short-term outcome and has a relatively good intermediate-term effect, but this depends on the later performed correction operation.Excessive use of refined flour, solid fats, and sugar in preparing baked products are considered to be unhealthy and is intricately linked with the development of lifestyle diseases. Replacing refined flour with whole wheat flour and solid fats with cold-pressed oil serves as an alternate option. The study was aimed at evaluating the physicochemical properties, nutrient composition, sensory attributes, and shelf life of cupcakes enriched using pomegranate seed oil (PSO). Vanilla and chocolate cupcake variants were prepared using 25 and 50% of PSO. A sensory panel consisting of 30 semi-trained participants was selected for evaluating the formulated products using a five-point hedonic scale. Nutrient content was estimated using standard techniques. The stability of the formulated product was determined by evaluating the physicochemical traits and microbial growth on the 0th, 4th, and 7th day. Mean scores of the sensorial analysis showed that the incorporation of PSO in cupcakes was highly accepted by the panel members. Chocolate cupcake containing 50% of PSO was found to be the most preferred product (3.53±0.94), followed by vanilla cupcake containing 25% of PSO (3.4±0.62). The moisture, protein, and fat content of chocolate cupcakes containing 25% of PSO were high. Cupcakes prepared with PSO can be stored for four days at room temperature. GC-MS analysis showed the presence of punicic acid, oleic acid, tocopherols, campesterol, sitosterols, stigmasterol, and α-tocopheryl acetate as pre-dominant fatty acid in unheated and heated PSO. In conclusion, cupcakes prepared using PSO showed acceptable physicochemical qualities and sensory properties which indicated its successful consumption by people affected with metabolic disorders.
